    An Investigation of the Large-scale Variability of the Apparently Single Wolf-Rayet Star WR 1

    In recent years, much studies have focused on determining the origin of the large-scale line-profile and/or photometric patterns of variability displayed by some apparently single Wolf-Rayet stars, with the existence of an unseen (collapsed?) companion or of spatially extended wind structures as potential candidates. We present observations of WR 1 which highlight the unusual character of the variations in this object. Our narrowband photometric observations reveal a gradual increase of the stellar continuum flux amounting to Delta v = 0.09 mag followed by a decline on about the same timescale (3-4 days). Only marginal evidence for variability is found during the 11 following nights. Strong, daily line-profile variations are also observed but they cannot be easily linked to the photometric variations. Similarly to the continuum flux variations, coherent time-dependent changes are observed in 1996 in the centroid, equivalent width, and skewness of He II 4686. Despite the generally coherent nature of the variations, we do not find evidence in our data for the periods claimed in previous studies. While the issue of a cyclical pattern of variability in WR 1 is still controversial, it is clear that this object might constitute in the future a cornerstone for our understanding of the mechanisms leading to the formation of largely anisotropic outflows in Wolf-Rayet stars.Comment: 11 pages, 9 figures, accepted for publication in Astronomy & Astrophysic

    A Case of Amblyomma testudinarium Tick Bite in a Korean Woman

    A case of tick bite was found in the inguinal region of a 74-year-old Korean woman. She was attacked by the tick while working in her vegetable garden in the vicinity of mountain located in Suncheon City, the southern coastal area of the Korean Peninsula. On admission she complained of mild discomfort and itching around the bite area. The causative tick was 23 mm long and had slender pedipalps. The scutum was quite ornate and had eyes at the edge. The genital aperture was located anterior to the level of the coxa II. The spiracular plate was comma-shaped and the anus was surrounded posteriorly by the anal groove. The coxa I had subequal 2 spurs; the external one slightly larger. The spur of coxa IV was slightly longer than those of coxae II and III. The tarsus IV had 2 distinct subapical ventral spurs. It was identified as the fully engorged adult female of Amblyomma testudinarium. This is the first human case of Amblyomma bite in Korea

    New insights into the nature of the SMC WR/LBV binary HD 5980

    We present the results of optical wavelength observations of the unusual SMC eclipsing binary system HD 5980 obtained in 1999 and 2004--2005. Radial velocity curves for the erupting LBV/WR object (star A) and its close WR-like companion (star B) are obtained by deblending the variable emission-line profiles of N IV and N V lines under the simplistic assumption that these lines originate primarily in the winds of star A and star B. The derived masses M_A=58--79 Mo and M_B=51--67 Mo, are more consistent with the stars' location near the top of the HRD than previous estimates. The presence of a wind-wind interaction region is inferred from the orbital phase-dependent behavior of He I P Cygni absorption components. The emission-line intensities continued with the declining trend previously seen in UV spectra. The behavior of the photospheric absorption lines is consistent with the results of Schweickhardt (2002) who concludes that the third object in the combined spectrum, star C, is also a binary system with P(starC)~96.5 days, e=0.83.     Additive Protection by Antioxidant and Apoptosis-Inhibiting Effects on Mosquito Cells with Dengue 2 Virus Infection

    Cytopathic effects (CPEs) in mosquito cells are generally trivial compared to those that occur in mammalian cells, which usually end up undergoing apoptosis during dengue virus (DENV) infection. However, oxidative stress was detected in both types of infected cells. Despite this, the survival of mosquito cells benefits from the upregulation of genes related to antioxidant defense, such as glutathione S transferase (GST). A second defense system, i.e., consisting of antiapoptotic effects, was also shown to play a role in protecting mosquito cells against DENV infection. This system is regulated by an inhibitor of apoptosis (IAP) that is an upstream regulator of caspases-9 and -3. DENV-infected C6/36 cells with double knockdown of GST and the IAP showed a synergistic effect on activation of these two caspases, causing a higher rate of apoptosis (>20%) than those with knockdown of each single gene (∼10%). It seems that the IAP acts as a second line of defense with an additional effect on the survival of mosquito cells with DENV infection. Compared to mammalian cells, residual hydrogen peroxide in DENV-infected C6/36 cells may signal for upregulation of the IAP. This novel finding sheds light on virus/cell interactions and their coevolution that may elucidate how mosquitoes can be a vector of DENV and probably most other arboviruses in nature

    The Zinc Transporter SLC39A14/ZIP14 Controls G-Protein Coupled Receptor-Mediated Signaling Required for Systemic Growth

    Aberrant zinc (Zn) homeostasis is associated with abnormal control of mammalian growth, although the molecular mechanisms of Zn's roles in regulating systemic growth remain to be clarified. Here we report that the cell membrane-localized Zn transporter SLC39A14 controls G-protein coupled receptor (GPCR)-mediated signaling. Mice lacking Slc39a14 (Slc39a14-KO mice) exhibit growth retardation and impaired gluconeogenesis, which are attributable to disrupted GPCR signaling in the growth plate, pituitary gland, and liver. The decreased signaling is a consequence of the reduced basal level of cyclic adenosine monophosphate (cAMP) caused by increased phosphodiesterase (PDE) activity in Slc39a14-KO cells. We conclude that SLC39A14 facilitates GPCR-mediated cAMP-CREB signaling by suppressing the basal PDE activity, and that this is one mechanism for Zn's involvement in systemic growth processes. Our data highlight SLC39A14 as an important novel player in GPCR-mediated signaling. In addition, the Slc39a14-KO mice may be useful for studying the GPCR-associated regulation of mammalian systemic growth

    Surface-Associated Plasminogen Binding of Cryptococcus neoformans Promotes Extracellular Matrix Invasion

    BACKGROUND:The fungal pathogen Cryptococcus neoformans is a leading cause of illness and death in persons with predisposing factors, including: malignancies, solid organ transplants, and corticosteroid use. C. neoformans is ubiquitous in the environment and enters into the lungs via inhalation, where it can disseminate through the bloodstream and penetrate the central nervous system (CNS), resulting in a difficult to treat and often-fatal infection of the brain, called meningoencephalitis. Plasminogen is a highly abundant protein found in the plasma component of blood and is necessary for the degradation of fibrin, collagen, and other structural components of tissues. This fibrinolytic system is utilized by cancer cells during metastasis and several pathogenic species of bacteria have been found to manipulate the host plasminogen system to facilitate invasion of tissues during infection by modifying the activation of this process through the binding of plasminogen at their surface. METHODOLOGY:The invasion of the brain and the central nervous system by penetration of the protective blood-brain barrier is a prerequisite to the establishment of meningoencephalitis by the opportunistic fungal pathogen C. neoformans. In this study, we examined the ability of C. neoformans to subvert the host plasminogen system to facilitate tissue barrier invasion. Through a combination of biochemical, cell biology, and proteomic approaches, we have shown that C. neoformans utilizes the host plasminogen system to cross tissue barriers, providing support for the hypothesis that plasminogen-binding may contribute to the invasion of the blood-brain barrier by penetration of the brain endothelial cells and underlying matrix. In addition, we have identified the cell wall-associated proteins that serve as plasminogen receptors and characterized both the plasminogen-binding and plasmin-activation potential for this significant human pathogen. CONCLUSIONS:The results of this study provide evidence for the cooperative role of multiple virulence determinants in C. neoformans pathogenesis and suggest new avenues for the development of anti-infective agents in the prevention of fungal tissue invasion
