    Transcription of Nonrepeated DNA in Neonatal and Fetal Mice

    The transcription of nonrepeated DNA sequences was measured by hybridization of RNA from neonatal and fetal mice to mouse DNA using three different techniques. The measurements indicate that a large part (about 70%) of the rapidly-labeled fetal RNA is transcribed from nonrepeated DNA sequences. It appears that more than 12% of the single-copy DNA sequences are represented in the RNA of newborn mice

    Reversal of hemochromatosis by apotransferrin in non-transfused and transfused Hbbth3/+ (heterozygous b1/b2 globin gene deletion) mice

    Intermediate beta-thalassemia has a broad spectrum of sequelae and affected subjects may require occasional blood transfusions over their lifetime to correct anemia. Iron overload in intermediate beta-thalassemia results from a paradoxical intestinal absorption, iron release from macrophages and hepatocytes, and sporadic transfusions. Pathological iron accumulation in parenchyma is caused by chronic exposure to non-transferrin bound iron in plasma. The iron scavenger and transport protein transferrin is a potential treatment being studied for correction of anemia. However, transferrin may also function to prevent or reduce iron loading of tissues when exposure to non-transferrin bound iron increases. Here we evaluate the effects of apotransferrin administration on tissue iron loading and early tissue pathology in non-transfused and transfused Hbb(th3/+) mice. Mice with the Hbb(th3/+) phenotype have mild to moderate anemia and consistent tissue iron accumulation in the spleen, liver, kidneys and myocardium. Chronic apotransferrin administration resulted in normalization of the anemia. Furthermore, it normalized tissue iron content in the liver, kidney and heart and attenuated early tissue changes in non-transfused Hbb(th3/+) mice. Apotransferrin treatment was also found to attenuate transfusion-mediated increases in plasma non-transferrin bound iron and associated excess tissue iron loading. These therapeutic effects were associated with normalization of transferrin saturation and suppressed plasma non-transferrin bound iron. Apotransferrin treatment modulated a fundamental iron regulatory pathway, as evidenced by decreased erythroid Fam132b gene (erythroferrone) expression, increased liver hepcidin gene expression and plasma hepcidin-25 levels and consequently reduced intestinal ferroportin-1 in apotransferrin-treated thalassemic mice

    HST/STIS low dispersion spectroscopy of three Compact Steep Spectrum sources Evidence for jet-cloud interaction

    We present Hubble Space Telescope Imaging Spectrograph long-slit spectroscopy of the emission line nebulae in the compact steep spectrum radio sources 3C 67, 3C 277.1, and 3C 303.1. We derive BPT (Baldwin- Philips-Terlevich; Baldwin et al. 1981) diagnostic emission line ratios for the nebulae which are consistent with a mix of shock excitation and photoionization in the extended gas. In addition, line ratios indicative of lower ionization gas are found to be associated with higher gas velocities. The results are consistent with a picture in which these galaxy scale radio sources interact with dense clouds in the interstellar medium of the host galaxies, shocking the clouds thereby ionizing and accelerating them.Comment: Accepted for publication in A&A. 6 pages, 8 figure

    Corn nitrogen rate recommendation tools’ performance across eight US midwest corn belt states

    Determining which corn (Zea mays L.) N fertilizer rate recommendation tools best predict crop N need would be valuable for maximizing profits and minimizing environmental consequences. Simultaneous comparisons of multiple tools across various environmental conditions have been limited. The objectives of this research were to evaluate the performance of publicly‐available N fertilizer recommendation tools across diverse soil and weather conditions for: (i) prescribing N rates for planting and split‐fertilizer applications, and (ii) economic and environmental effects. Corn N‐response trials using standardized methods were conducted at 49 sites, spanning eight US Midwest states and three growing seasons. Nitrogen applications included eight rates in 45 kg N ha−1 increments all at‐planting and matching rates with 45 kg N ha−1 at‐planting plus at the V9 development stage. Tool performances were compared to the economically optimal N rate (EONR). Over this large geographic region, only 10 of 31 recommendation tools (mainly soil nitrate tests) produced N rate recommendations that weakly correlated to EONR (P ≤ .10; r2 ≤ .20). With other metrics of performance, the Maximum Return to N (MRTN) soil nitrate tests, and canopy reflectance sensing came close to matching EONR. Economically, all tools but the Maize‐N crop growth model had similar returns compared to EONR. Environmentally, yield goal based tools resulted in the highest environmental costs. Results show that no tool was universally reliable over this study\u27s diverse growing environments, suggesting that additional tool development is needed to better represent N inputs and crop utilization at a larger regional level

    Implementation of a parentage control system in Portuguese beef-cattle with a panel of microsatellite markers

    A study was conducted to assess the feasibility of applying a panel of 10 microsatellite markers in parentage control of beef cattle in Portugal. In the first stage, DNA samples were collected from 475 randomly selected animals of the Charolais, Limousin and Preta breeds. Across breeds and genetic markers, means for average number of alleles, effective number of alleles, expected heterozygosity and polymorphic information content, were 8.20, 4.43, 0.733 and 0.70, respectively. Enlightenment from the various markers differed among breeds, but the set of 10 markers resulted in a combined probability above 0.9995 in the ability to exclude a random putative parent. The marker-set thus developed was later used for parentage control in a group of 140 calves from several breeds, where there was the suspicion of possible faulty parentage recording. Overall, 76.4% of the calves in this group were compatible with the recorded parents, with most incompatibilities due to misidentification of the dam. Efforts must be made to improve the quality of pedigree information, with particular emphasis on information recorded at the calf's birth

    Laparoscopic resection of a residual retroperitoneal tumor mass of nonseminomatous testicular germ cell tumors

    Resection of a residual retroperitoneal tumor mass (RRRTM) is standard procedure after combination chemotherapy for metastatic nonseminomatous testicular germ cell tumors (NSTGCT). At the University Medical Center Groningen, 79 consecutive patients with disseminated NSTGCT were treated with cisplatin combination chemotherapy between 2005 and 2007. Laparoscopic RRRTM was performed for patients with RRTM located less than 5 cm ventrally or laterally from the aorta or the vena cava. The 29 patients who fulfilled the criteria had a median age of 25 years (range, 16-59 years). The stages of disease before chemotherapy treatment according to the Royal Marsden classification were 2A (n = 6, 21%), 2B (n = 14, 48%), 2C (n = 3, 10%), and 4 with a lymph node status of N2 (n = 6, 21%). The median duration of laparoscopy was 198 min (range, 122-325 min). The median diameter of the RRTM was 21 mm (range, 11-47 mm). Laparoscopic resection was successful for 25 patients (86%). Conversion was necessary for three patients (10%): two due to bleeding and one because of obesity. One nonplanned hand-assisted procedure (3%) also had to be performed. Histologic examination of the specimens showed fibrosis or necrosis in 12 patients (41%), mature teratoma in 16 patients (55%), and viable tumor in 1 patient (3%). The median hospital stay was 1 day (range, 1-6 days). During a median follow-up period of 47 months (29-70 months), one patient experienced an early relapse (1 month after the end of treatment) (4%). For properly selected patients, laparoscopic resection of RRTM is an improvement in the combined treatment of disseminated NSTGCT and associated with a short hospital stay, minimal morbidity, rapid recovery, and a neat cosmetic result. Long-term data to prove oncologic efficacy are awaited

    NCF1 gene and pseudogene pattern: association with parasitic infection and autoimmunity

    <p>Abstract</p> <p>Background</p> <p>Neutrophil cytosolic factor 1, p47<sup>phox </sup>(NCF1) is a component of the leukocyte NADPH oxidase complex mediating formation of reactive oxygen intermediates (ROI) which play an important role in host defense and autoimmunity. An individual genomic pattern of <it>ncf1 </it>and its two types of pseudogenes (reflected by the ΔGT/GTGT ratio) may influence the individual capacity to produce ROI.</p> <p>Methods</p> <p>NCF1ΔGT/GTGT ratios were correlated with clinical parameters and ROI production during <it>Plasmodium falciparum </it>malaria and with susceptibility to the autoimmune disease multiple sclerosis (MS).</p> <p>Results</p> <p>Among Gabonese children with severe malaria, ROI production from peripheral blood tended to be higher in individuals with a ΔGT/GTGT ratio ≤ 1:1. ΔGT/GTGT ratios were not associated with susceptibility to MS, but to age-of-onset among MS patients.</p> <p>Conclusion</p> <p>The genomic pattern of <it>NCF1 </it>and its pseudogenes might influence ROI production but only marginally influence susceptibility to and outcome of malaria and MS.</p
