Health related quality of life in patients with multiple myeloma undergoing a double transplantation
Applicability and reproducibility of acute myeloid leukaemia stem cell assessment in a multi-centre setting
Leukaemic stem cells (LSC) have been experimentally defined as the leukaemia-propagating population and are thought to be the cellular reservoir of relapse in acute myeloid leukaemia (AML). Therefore, LSC measurements are warranted to facilitate accurate risk stratification. Previously, we published the composition of a one-tube flow cytometric assay, characterised by the presence of 13 important membrane markers for LSC detection. Here we present the validation experiments of the assay in several large AML research centres, both in Europe and the United States. Variability within instruments and sample processing showed high correlations between different instruments (Rpearson > 0Β·91, P < 0Β·001). Multi-centre testing introduced variation in reported LSC percentages but was found to be below the clinical relevant threshold. Clear gating protocols resulted in all laboratories being able to perform LSC assessment of the validation set. Participating centres were nearly unanimously able to distinguish LSChigh (>0Β·03% LSC) from LSClow (<0Β·03% LSC) despite inter-laboratory variation in reported LSC percentages. This study proves that the LSC assay is highly reproducible. These results together with the high prognostic impact of LSC load at diagnosis in AML patients render the one-tube LSC assessment a good marker for future risk classification
Assessment of the appropriate use criteria for amyloid PET in an unselected memory clinic cohort: The ABIDE project
Introduction
The objective of this study was to assess the usefulness of the appropriate use criteria (AUC) for amyloid imaging in an unselected cohort.
Methods
We calculated sensitivity and specificity of appropriate use (increased confidence and management change), as defined by Amyloid Imaging Taskforce in the AUC, and other clinical utility outcomes. Furthermore, we compared differences in postβpositron emission tomography diagnosis and management change between βAUC-consistentβ and βAUC-inconsistentβ patients.
Results
Almost half (250/507) of patients were AUC-consistent. In both AUC-consistent and AUC-inconsistent patients, postβpositron emission tomography diagnosis (28%β21%) and management (32%β17%) change was substantial. The Amyloid Imaging Taskforce's definition of appropriate use occurred in 55/507 (13%) patients, detected by the AUC with a sensitivity of 93%, and a specificity of 56%. Diagnostic changes occurred independently of AUC status (sensitivity: 57%, specificity: 53%).
Discussion
The current AUC are not sufficiently able to discriminate between patients who will benefit from amyloid positron emission tomography and those who will not

Here, we show that high-efficiency CD40-targeted adenoviral gene transfer of GM-CSF to CML-derived DC induces long-lived maturation in the absence of exogenous cytokines and may thus ensure protracted stimulation of CML-specific T cells upon vaccination
