    Use of a dual reporter plasmid to demonstrate bactofection with an attenuated aroa- derivative of Pasteurella multocida b:2

    A reporter plasmid pSRG has been developed which expresses red fluorescent protein (RFP) from a constitutive prokaryotic promoter within Pasteurella multocida B:2 and green fluorescent protein (GFP) from a constitutive eukaryotic promoter within mammalian cells. This construct has been used to determine the location and viability of the bacteria when moving from the extracellular environment into the intracellular compartment of mammalian cells. Invasion assays with embryonic bovine lung (EBL) cells and an attenuated AroA- derivative of Pasteurella multocida B:2 (strain JRMT12), harbouring the plasmid pSRG, showed that RFP-expressing bacteria could be detected intracellularly at 3 h post-invasion. At this stage, some EBL cells harbouring RFP-expressing bacteria were observed to express GFP simultaneously, indicating release of the plasmid into the intracellular environment. At 5 h post-invasion, more EBL cells were expressing GFP, while still harbouring RFP-expressing bacteria. Concurrently, some EBL cells were shown to express only GFP, indicating loss of viable bacteria within these cells. These experiments proved the functionality of the pSRG dual reporter system and the potential of P. multocida B:2 JRMT12 for bactofection and delivery of a DNA vaccine

    Influenza viruses in healthy wild birds in Hong Kong

    Poster Presentations: Animal Influenza EcologyWild waterfowl are considered the natural reservoir of influenza A viruses. The recent outbreak of highly pathogenic avian influenza (HPAI) H5N1 in Asia which has now spread as far as Africa highlights the importance of defining the influenza virus gene pool in these birds and understanding the potential role played by migratory waterfowl in such HPAI outbreaks. Seventy-three influenza viruses were isolated from 16,724 samples collected from feral waterfowls or their fecal droppings during 2003-7 at Mai Po Marshes and Lok Ma Chau in Hong Kong. A diversity of influenza viruses representing hemagglutinin subtypes of H1, H2, H4, H5, H6, H7, H8, H9, H10 and H11; neuramidinase subtypes of N1, N2, N3, N4, N6, N7, N8 and N9, were isolated. Seventy-two of these 73 positive isolates were collected during the winter period coinciding with the southern migration of waterfowl along the East Asian flyway. No HPAI H5N1 viruses were isolated from healthy birds sampled in this study, though H5N1 viruses have been isolated from dead wild birds found in Hong Kong. Phylogenetic analyses of the HA gene of the H5 viruses isolated in the study showed that they clustered with other LP H5 viruses isolated from Hokkaido, Mongolia and Siberia but they seemed not to be very closely related to the HP H5N1.     Detection of iron-oxide magnetic nanoparticles using magnetic tunnel junction sensors with conetic alloy

    This journal issue contain selected papers of the 2011 IEEE International Magnetics (INTERMAG) Conference, Taipei, Taiwan, 25-29 April 2011.We demonstrated the detection of 20-nm iron-oxide magnetic nanoparticles (MNPs) using Al2 O3 magnetic tunnel junction sensors (MTJs) with Conetic alloy. Conetic alloy Ni77Fe14Cu 5Mo4 was deposited as the MTJ free layer and pinned layer due to its magnetically soft properties. The magnetoresistance (MR) curves of MTJs with Conetic alloy showed tunneling magnetoresistance of 8.0% with small hysteresis and high linearity in the sensing region, after applying an external magnetic field of 14 Oe along the hard axis. The sensitivity of the MTJ sensors with Conetic alloy was determined to be 0.3%/Oe within a linear region at room temperature. The MNPs of three different concentrations were successfully detected by the shifts of the MR loops of the MTJs, and it was observed that the resistance deviations of the MTJ sensors increased with the logarithm of MNP concentrations. The maximum resistance deviation was 0.16 Ω for an MNP concentration of 20.0 mg/mL. MTJ sensors, together with MNPs, are a promising platform for future biosensor applications, and this paper shows that Conetic alloy is feasible for improving the performance of this platform. © 2011 IEEE.published_or_final_versio

    Sinter formation during directed energy deposition of titanium alloy powders

    During directed energy deposition (DED) additive manufacturing, powder agglomeration and sintering can occur outside of the melt pool when using titanium alloy powders. Using in situ synchrotron radiography we investigate the mechanisms by which sintering of Ti6242 powder occurs around the pool, performing a parametric study to determine the influence of laser power and stage traverse speed on sinter build-up. The results reveal that detrimental sinter can be reduced using a high laser power or increased stage traverse speed, although the latter also reduces deposition layer thickness. The mechanism of sinter formation during DED was determined to be in-flight heating of the powder particles in the laser beam. Calculations of particle heating under the processing conditions explored in this study confirm that powder particles can reasonably exceed 700 °C, the threshold for Ti surface oxide dissolution, and thus the powder is prone to sintering if not incorporated into the melt pool. The build-up of sinter powder layer on deposit surfaces led to lack of fusion pores. To mitigate sinter formation and its detrimental effects on DED component quality, it is essential that the powder delivery spot area is smaller than the melt pool, ensuring most powder lands in the melt pool

    Macular Telangiectasia Type 2: A Classification System Using MultiModal Imaging MacTel Project Report Number 10

    Purpose: To develop a severity classification for macular telangiectasia type 2 (MacTel) disease using multimodal imaging. Design: An algorithm was used on data from a prospective natural history study of MacTel for classification development. Subjects: A total of 1733 participants enrolled in an international natural history study of MacTel. Methods: The Classification and Regression Trees (CART), a predictive nonparametric algorithm used in machine learning, analyzed the features of the multimodal imaging important for the development of a classification, including reading center gradings of the following digital images: stereoscopic color and red-free fundus photographs, fluorescein angiographic images, fundus autofluorescence images, and spectral-domain (SD)-OCT images. Regression models that used least square method created a decision tree using features of the ocular images into different categories of disease severity. Main Outcome Measures: The primary target of interest for the algorithm development by CART was the change in best-corrected visual acuity (BCVA) at baseline for the right and left eyes. These analyses using the algorithm were repeated for the BCVA obtained at the last study visit of the natural history study for the right and left eyes. Results: The CART analyses demonstrated 3 important features from the multimodal imaging for the classification: OCT hyper-reflectivity, pigment, and ellipsoid zone loss. By combining these 3 features (as absent, present, noncentral involvement, and central involvement of the macula), a 7-step scale was created, ranging from excellent to poor visual acuity. At grade 0, 3 features are not present. At the most severe grade, pigment and exudative neovascularization are present. To further validate the classification, using the Generalized Estimating Equation regression models, analyses for the annual relative risk of progression over a period of 5 years for vision loss and for progression along the scale were performed. Conclusions: This analysis using the data from current imaging modalities in participants followed in the MacTel natural history study informed a classification for MacTel disease severity featuring variables from SD-OCT. This classification is designed to provide better communications to other clinicians, researchers, and patients.     Perceptions and behaviors related to hand hygiene for the prevention of H1N1 influenza transmission among Korean university students during the peak pandemic period

    <p>Abstract</p> <p>Background</p> <p>This study was performed to better assess the perceptions, motivating factors, and behaviors associated with the use of hand washing to prevent H1N1 influenza transmission during the peak pandemic period in Korea.</p> <p>Methods</p> <p>A cross-sectional survey questionnaire was completed by 942 students at a university campus in Suwon, Korea, between December 1 and 8, 2009. The survey included questions regarding individual perceptions, motivating factors, and behaviors associated with hand washing for the prevention of H1N1 influenza transmission.</p> <p>Results</p> <p>Compared to one year prior, 30.3% of participants reported increasing their hand washing frequency. Female students were more likely to practice more frequent hand washing. Women also perceived the effectiveness of hand washing to be lower, and illness severity and personal susceptibility to H1N1 infection to be higher. Study participants who were female (OR: 1.79-3.90) who perceived of hand washing to be effective (OR: 1.34-12.15) and illness severity to be greater (OR: 1.00-3.12) washed their hands more frequently.</p> <p>Conclusions</p> <p>Korean students increased their frequency of hand hygiene practices during the pandemic, with significant gender differences existing in the attitudes and behaviors related to the use of hand hygiene as a means of disease prevention. Here, the factors that affected hand washing behavior were similar to those identified at the beginning of the H1N1 or SARS pandemics, suggesting that public education campaigns regarding hand hygiene are effective in altering individual hand hygiene habits during the peak periods of influenza transmission.</p

    Avoidance behaviors and negative psychological responses in the general population in the initial stage of the H1N1 pandemic in Hong Kong

    <p>Abstract</p> <p>Background</p> <p>During the SARS pandemic in Hong Kong, panic and worry were prevalent in the community and the general public avoided staying in public areas. Such avoidance behaviors could greatly impact daily routines of the community and the local economy. This study examined the prevalence of the avoidance behaviors (i.e. avoiding going out, visiting crowded places and visiting hospitals) and negative psychological responses of the general population in Hong Kong at the initial stage of the H1N1 epidemic.</p> <p>Methods</p> <p>A sample of 999 respondents was recruited in a population-based survey. Using random telephone numbers, respondents completed a structured questionnaire by telephone interviews at the 'pre-community spread phase' of the H1N1 epidemic in Hong Kong.</p> <p>Results</p> <p>This study found that 76.5% of the respondents currently avoided going out or visiting crowded places or hospitals, whilst 15% felt much worried about contracting H1N1 and 6% showed signs of emotional distress. Females, older respondents, those having unconfirmed beliefs about modes of transmissions, and those feeling worried and emotionally distressed due to H1N1 outbreak were more likely than others to adopt some avoidance behaviors. Those who perceived high severity and susceptibility of getting H1N1 and doubted the adequacy of governmental preparedness were more likely than others to feel emotionally distressed.</p> <p>Conclusions</p> <p>The prevalence of avoidance behaviors was very high. Cognitions, including unconfirmed beliefs about modes of transmission, perceived severity and susceptibility were associated with some of the avoidance behaviors and emotional distress variables. Public health education should therefore provide clear messages to rectify relevant perceptions.</p

    New Physics Signals in Longitudinal Gauge Boson Scattering at the LHC

    We introduce a novel technique designed to look for signatures of new physics in vector boson fusion processes at the TeV scale. This functions by measuring the polarization of the vector bosons to determine the relative longitudinal to transverse production. In studying this ratio we can directly probe the high energy E^2-growth of longitudinal vector boson scattering amplitudes characteristic of models with non-Standard Model (SM) interactions. We will focus on studying models parameterized by an effective Lagrangian that include a light Higgs with non-SM couplings arising from TeV scale new physics associated with the electroweak symmetry breaking, although our technique can be used in more general scenarios. We will show that this technique is stable against the large uncertainties that can result from variations in the factorization scale, improving upon previous studies that measure cross section alone
