Antigen-driven clonal proliferation of B cells within the target tissue of an autoimmune disease: the salivary glands of patients with Sjögren's syndrome
Structures resembling germinal centers are seen in the salivary glands of patients with Sjögren's syndrome, but it is not known whether the microenvironment of these cell clusters is sufficient for the induction of a germinal center response. Therefore, we cloned and sequenced rearranged Ig V genes expressed by B cells isolated from sections of labial salivary gland biopsies from two Sjögren's syndrome patients. Rearranged V genes from B cells within one cell cluster were polyclonal and most had few somatic mutations. Two adjacent clusters from another patient each contained one dominant B cell clone expressing hypermutated V genes. None of the rearranged V genes was found in both clusters, suggesting that cells are unable to migrate out into the surrounding tissue and seed new clusters. The ratios of replacement to silent mutations in the framework and complementarity determining regions suggest antigen selection of high-affinity mutants. These results show that an antigen-driven, germinal center-type B cell response is taking place within the salivary glands of Sjögren's syndrome patients. In view of the recent demonstration of a germinal center response within the rheumatoid synovial membrane and the existence of similar structures in the target tissues of other autoimmune. diseases, we propose that germinal center- type responses can be induced in the nonlymphoid target tissues of a variety of autoimmune diseases
Massive Quark Production in Electron Positron Annihilation to Order
Recent analytical and numerical results for the three-loop polarization
function allow to present a phenomenological analysis of the cross section for
massive quark production in electron positron annihilation to order
. Numerical predictions based on fixed order perturbation theory
are presented for charm and bottom production above 5 and 11.5 GeV,
respectively. The contribution from these energy regions to ,
the running QED coupling constant at scale M_Z, are given. The dominant terms
close to threshold, i.e. in an expansion for small quark velocity , are

Matching the heavy-quark fields in QCD and HQET at four loops
The QCD/HQET matching coefficient for the heavy-quark field is calculated up
to four loops. It must be finite; this requirement produces analytical results
for some terms in the four-loop on-shell heavy-quark field renormalization
constant which were previously only known numerically. The effect of a non-zero
lighter-flavor mass is calculated up to three loops. A class of on-shell
integrals with two masses is analyzed in detail. By specifying our result to
QED, we obtain the relation between the electron field and the Bloch--Nordsieck

Heavy-light current correlators at order in QCD and HQET
The non-diagonal correlators of vector and scalar currents are considered at
three-loop order in QCD. The full mass dependence is computed in the case where
one of the quarks is massless and the other one carries mass . We exploit
the decoupling relations between the full theory and the heavy quark effective
theory (HQET) in order to obtain the logarithmic parts of the leading threshold
terms. With the help of conformal mapping and Pad\'e approximation numerical
estimates for the non-logarithmic terms are extracted which in turn lead to a
prediction of the correlator in HQET at order . As applications of
the vector and scalar correlator we consider the single-top-quark production
via the process and the decay rate of a charged Higgs
boson into hadrons, respectively. In both cases the computed NLO corrections
are shown to be numerically much less important than the leading ones. On the
contrary, the NLO order QCD corrections to the HQET sum rule for the leptonic
decay rate of a heavy-light meson proves to be comparable to the leading one.Comment: 31 page
