Distinct emphysema subtypes defined by quantitative CT analysis are associated with specific pulmonary matrix metalloproteinases.
BACKGROUND: Emphysema is characterised by distinct pathological sub-types, but little is known about the divergent underlying aetiology. Matrix-metalloproteinases (MMPs) are proteolytic enzymes that can degrade the extracellular matrix and have been identified as potentially important in the development of emphysema. However, the relationship between MMPs and emphysema sub-type is unknown. We investigated the role of MMPs and their inhibitors in the development of emphysema sub-types by quantifying levels and determining relationships with these sub-types in mild-moderate COPD patients and ex/current smokers with preserved lung function. METHODS: Twenty-four mild-moderate COPD and 8 ex/current smokers with preserved lung function underwent high resolution CT and distinct emphysema sub-types were quantified using novel local histogram-based assessment of lung density. We analysed levels of MMPs and tissue inhibitors of MMPs (TIMPs) in bronchoalveolar lavage (BAL) and assessed their relationship with these emphysema sub-types. RESULTS: The most prevalent emphysema subtypes in COPD subjects were mild and moderate centrilobular (CLE) emphysema, while only small amounts of severe centrilobular emphysema, paraseptal emphysema (PSE) and panlobular emphysema (PLE) were present. MMP-3, and -10 associated with all emphysema sub-types other than mild CLE, while MMP-7 and -8 had associations with moderate and severe CLE and PSE. MMP-9 also had associations with moderate CLE and paraseptal emphysema. Mild CLE occurred in substantial quantities irrespective of whether airflow obstruction was present and did not show any associations with MMPs. CONCLUSION: Multiple MMPs are directly associated with emphysema sub-types identified by CT imaging, apart from mild CLE. This suggests that MMPs play a significant role in the tissue destruction seen in the more severe sub-types of emphysema, whereas early emphysematous change may be driven by a different mechanism. Quasi-Elastic Scattering, Random Fields and phonon-coupling effects in PbMg1/3Nb2/3O3
The low-energy part of the vibration spectrum in PbMgNbO
(PMN) relaxor ferroelectric has been studied by neutron scattering above and
below the Burns temperature, T. The transverse acoustic and the lowest
transverse optic phonons are strongly coupled and we have obtained a model for
this coupling. We observe that the lowest optic branch is always underdamped. A
resolution-limited central peak and quasi-elastic scattering appear in the
vicinity of the Burns temperature. It is shown that it is unlikely that the
quasi-elastic scattering originates from the combined effects of coupling
between TA and TO phonons with an increase of the damping of the TO phonon
below T. The quasi-elastic scattering has a peak as a function of
temperature close to the peak in the dielectric constant while the intensity of
the central peak scattering increases strongly below this temperature. These
results are discussed in terms of a random field model for relaxors
Marked increase in community-associated methicillin-resistant Staphylococcus aureus infections, Western Australia, 2004ā2018
This study presents enhanced surveillance data from 2004 ā 2018 for all community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) specimens collected in Western Australia (WA), and describes the changing epidemiology over this period. A total of 57,557 cases were reviewed. Annual incidence rates increased from 86.2 cases per 100,000 population to 245.6 per 100,000 population (IRR = 2.9, CI95 2.7 ā 3.0). The proportion of isolates carrying Panton-Valentine leukocidin (PVL)-associated genes increased from 3.4% to 59.8% (Ļ2 test for trend 7021.9, p\u3c0.001). The emergence of PVL-positive, āQueensland CA-MRSAā (ST93- IV) and āWA 121ā (ST5-IV) accounted for the majority of increases in CA-MRSA across the study period. It is unclear why some clones are more prolific in certain regions. In WA, CA-MRSA rates increase as indices of temperature and humidity increase after controlling for socioeconomic disadvantage. We suggest climatic conditions may contribute to transmission, along with other socio-behavioural factors. A better understanding of the ability for certain clones to form ecological niches and cause outbreaks is required

Microbiological influences on fracture surfaces of intact mudstone and the implications for geological disposal of radioactive waste
The significance of the potential impacts of microbial activity on the transport properties of host rocks for geological repositories is an area of active research. Most recent work has focused on granitic environments. This paper describes pilot studies investigating changes in transport properties that are produced by microbial activity in sedimentary rock environments in northern Japan. For the first time, these short experiments (39 days maximum) have shown that the denitrifying bacteria, Pseudomonas denitrificans, can survive and thrive when injected into flow-through column experiments containing fractured diatomaceous mudstone and synthetic groundwater under pressurized conditions. Although there were few significant changes in the fluid chemistry, changes in the permeability of the biotic column, which can be explained by the observed biofilm formation, were quantitatively monitored. These same methodologies could also be adapted to obtain information from cores originating from a variety of geological environments including oil reservoirs, aquifers and toxic waste disposal sites to provide an understanding of the impact of microbial activity on the transport of a range of solutes, such as groundwater contaminants and gases (e.g. injected carbon dioxide)
