    An investigation into the pharmacological properties of propolis

    Propolis is a traditional value-added product from beekeeping, and has been widely used in medicine and the chemical industry because of its extensive biological activities. In this dissertation some pharmacological properties of propolis extracted by ethanol (EEP) and water (WSP) were studied by means of modem pharmacological methods. The results of the experiments show the following: 1. Both EEP and WSD led to decreased levels of fasting blood glucose (FBG), glycosylated haemoglobin (HBA1c), Fructosamine (FRU), blood urea nitrogen (BUN), uric acid (UA), malonaldehyde (MDA), nitric oxide (NO), nitric oxide synthetase (NOS), total cholesterol (TC), triglyceride (TG), low-density lipoprotein cholesterol (LDL-C) and very-low density lipoprotein cholesterol (VLDL-C) in the serum of fasting rats; and, to increased serum levels of high-density lipoprotein cholesterol (HDL-C) and superoxide dismutase (SOD) in rats with diabetes mellitus. This suggests that propolis can control blood glucose and modulate the metabolism of glucose, blood lipid and protein, leading to decreased outputs of lipid peroxidation and scavenging free radicals in rats with diabetes mellitus. 2. Both EEP and WSD showed inhibitory effects on swelling induced by Freund's complete adjuvant and decreased the degree of local inflammatory responses; significantly inhibited the increase of interleukin-6 (lL-6) in inflamed tissues, but had no significant effect on levels of interleukin-2 (IL-2) and interferon-y (IFN-y). The results are consistent with the interpretation that EEP and WSD may exert these effects by inhibiting the activation and differentiation of mononuclear macrophages. 3. Both EEP and WSD exhibited significant anti-inflammatory effects in animal models with respect to thoracic capillary vessel leakage in mice, Carrageenan-induced oedema, Carrageen-aninduced pleurisy and acute lung damage in rats. The mechanisms for the anti-inflammatory effects probably involve decreasing prostaglandin and NO levels. 4. Both EEP and WSD had an inhibitory effect on the level of TG, TC, LDL-C, glutamic-pyruvic transaminase (GPT) and glutamic-oxalacetic transaminase (GOT) in serum, and TC, TG and MDA in liver of hyperlipidemic SD rats; but were without effects on HDL-C, MDA, SOD and NO in serum. EEP also reduced body weight, liver weight and liver index, but WSD did not reduce those indexes. The results showed that the two extracts contribute to the improvement of lipid metabolism in hyperlipidemic rats and provide them with the required anti-oxidative activity. 5. The 80% ethanol extracts of propolis had the highest flavonoid content. The flavone content of water extracts of propolis were obtained with a natural surface-active agent, under ultrasonic perturbation at 80°C, for 12 h and was 6.7 times greater than that of propolis extracted with water at room temperature. Compared to EEP, WSD has the same or a greater anti-tumor or anti-inflammation effect at the same dosage, and shows considerable pharmacological potential especially because of its low side effects and ease of preparation

    Effects of Encapsulated Propolis on Blood Glycemic Control, Lipid Metabolism, and Insulin Resistance in Type 2 Diabetes Mellitus Rats

    The present study investigates the encapsulated propolis on blood glycemic control, lipid metabolism, and insulin resistance in type 2 diabetes mellitus (T2DM) rats. The animal characteristics and biological assays of body weight, fasting blood glucose (FBG), fasting serum insulin (FINS), insulin act index (IAI), triglycerides (TG), total cholesterol (TC), high-density lipoprotein cholesterol (HDL-C), and low-density lipoprotein cholesterol (LDL-C) were measured and euglycemic hyperinsulinemic glucose clamp technique were used to determine these effects. Our findings show that oral administration of encapsulated propolis can significantly inhibit the increasing of FBG and TG in T2DM rats and can improve IAI and M value in euglycemic hyperinsulinemic clamp experiment. There was no significant effects on body weight, TC, HDL-C, and LDL-C in T2DM rats treated with encapsulated propolis. In conclusion, the results indicate that encapsulated propolis can control blood glucose, modulate lipid metabolism, and improve the insulin sensitivity in T2DM rats

    Inhibitory Effect of Chinese Propolis on Phosphatidylcholine-Specific Phospholipase C Activity in Vascular Endothelial Cells

    To understand the mechanisms underlying the anti-inflammatory action of Chinese propolis, we investigated its effect on the activity of phosphatidylcholine-specific phospholipase C (PC-PLC) that plays critical roles in control of vascular endothelial cell (VEC) function and inflammatory responses. Furthermore, p53 and reactive oxygen species (ROS) levels and mitochondrial membrane potential (Δψm) were investigated. Our data indicated that treatment of Chinese propolis 6.25 and 12.5 μg/ml for 12 hours increased VEC viability obviously. Exposure to Chinese propolis 6.25, 12.5, and 25 μg/ml for 6 and 12 hours significantly decreased PC-PLC activity and p53 level, and ROS levels were depressed by Chinese propolis 12.5 μg/ml and 25 μg/ml dramatically. The Δψm of VECs was not affected by Chinese propolis at low concentration but disrupted by the propolis at 25 μg/ml significantly, which indicated that Chinese propolis depressed PC-PLC activity and the levels of p53 and ROS in VECs but disrupted Δψm at a high concentration

    Biological Activities of Chinese Propolis and Brazilian Propolis on Streptozotocin-Induced Type 1 Diabetes Mellitus in Rats

    Propolis is a bee-collected natural product and has been proven to have various bioactivities. This study tested the effects of Chinese propolis and Brazilian propolis on streptozotocin-induced type 1 diabetes mellitus in Sprague-Dawley rats. The results showed that Chinese propolis and Brazilian propolis significantly inhibited body weight loss and blood glucose increase in diabetic rats. In addition, Chinese propolis-treated rats showed an 8.4% reduction of glycated hemoglobin levels compared with untreated diabetic rats. Measurement of blood lipid metabolism showed dyslipidemia in diabetic rats and Chinese propolis helped to reduce total cholesterol level by 16.6%. Moreover, oxidative stress in blood, liver and kidney was improved to various degrees by both Chinese propolis and Brazilian propolis. An apparent reduction in levels of alanine transaminase, aspartate transaminase, blood urea nitrogen and urine microalbuminuria-excretion rate demonstrated the beneficial effects of propolis in hepatorenal function. All these results suggested that Chinese propolis and Brazilian propolis can alleviate symptoms of diabetes mellitus in rats and these effects may partially be due to their antioxidant ability

    Grouping, spectrum-effect relationship and antioxidant compounds of Chinese propolis from different regions using multivariate analyses and off-line anti-DPPH assay

    49 samples of propolis from different regions in China were collected and analyzed for their chemical compositions, contents of total flavonoids (TFC), total phenolic acid (TPC) and antioxidant activity. High-performance liquid chromatography (HPLC) analysis identified 15 common components, including key marker compounds pinocembrin, 3-O-acetylpinobanksin, galangin, chrysin, benzyl p-coumarate, pinobanksin and caffeic acid phenethyl ester (CAPE). Cluster analysis (CA) and correlation coefficients (CC) analysis showed that these propolis could be divided into three distinct groups. Principal component analysis (PCA) and multiple linear regression analysis (MLRA) revealed that the contents of isoferulic acid, caffeic acid, CAPE, 3,4-dimethoxycinnamic acid, chrysin and apigenin are closely related to the antioxidant properties of propolis. In addition, eight peak areas decreased after reacting with 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radicals, indicating that these compounds have antioxidant activity. The results indicate that the grouping and spectrum-effect relationship of Chinese propolis are related to their chemical compositions, and several compounds may serve as a better marker for the antioxidant activity of Chinese propolis than TFC and TPC. The findings may help to develop better methods to evaluate the quality of propolis from different geographic origins

    Ultrasound-assisted extraction of polyphenols from Chinese propolis

    IntroductionPropolis is a beneficial bioactive food with rich polyphenols content. Nowadays, an increasing interest is attracted to the extraction of polyphenols from raw propolis. This study utilized the novel ultrasound-assisted approach for polyphenol extraction from Chinese propolis, aiming to improve its extraction yield and reveal the relevant mechanisms via extraction kinetic study as well as the compositional and structural analysis.MethodsThe optimum ultrasound-assisted extraction conditions were optimized according to the total phenolic content and total flavonoids content. Compositional and structural analysis were conducted using high performance liquid chromatography-quadrupole-time of flight tandem mass spectrometry, high-performance liquid chromatography, Fourier-transform infrared spectroscopy (FT-IR) and scanning electron microscope (SEM).Results and discussionThe optimum ultrasound-assisted extraction conditions were as follows: ratio of liquid to solid, 60:1; ultrasound power, 135 W; ultrasound duration, 20 min. Under the optimum conditions, the antioxidant activities of the extract were increased by 95.55% and 64.46% by 2,2-diphenyl-1-picrylhydrazyl radical scavenging ability assay and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging ability assay, respectively, compared to those obtained by traditional maceration. The second-order kinetics model was employed to study the extraction process; it was found that ultrasound significantly accelerated the extraction of propolis and increased the maximum extraction volume of phenolic compounds. The qualitative and quantitative analysis of polyphenol compositions showed that ultrasound did not change the polyphenol types in the extract but it significantly improved the contents of various flavonoids and phenolic acids such as galangin, chrysin, pinocembrin, pinobanksin and isoferulic acid. Likewise, the FT-IR analysis indicated that the types of functional groups were similar in the two extracts. The SEM analysis revealed that the ultrasound-assisted extraction enhanced the contact areas between propolis and ethanol by breaking down the propolis particles and eroding the propolis surface

    Population genetics and host specificity of Varroa destructor mites infesting eastern and western honeybees

    In a globalized world, parasites are often brought in contact with new potential hosts. When parasites successfully shift host, severe diseases can emerge at a large cost to society. However, the evolutionary processes leading to successful shifts are rarely understood, hindering risk assessment, prevention, or mitigation of their effects. Here, we screened populations of Varroa destructor, an ectoparasitic mite of the honeybee genus Apis, to investigate their genetic structure and reproductive potential on new and original hosts. From the patterns identified, we deduce the factors that influenced the macro- and microevolutionary processes that led to the structure observed. Among the mite variants identified, we found two genetically similar populations that differed in their reproductive abilities and thus in their host specificity. These lineages could interbreed, which represents a threat due to the possible increased virulence of the parasite on its original host. However, interbreeding was unidirectional from the host-shifted to the nonshifted native mites and could thus lead to speciation of the former. The results improve our understanding of the processes affecting the population structure and evolution of this economically important mite genus and suggest that introgression between shifted and nonshifted lineages may endanger the original host

    Protection of Bovine Mammary Epithelial Cells from Hydrogen Peroxide-Induced Oxidative Cell Damage by Resveratrol

    The mammary epithelial cells (MECs) of high-producing dairy cows are likely to be subject to oxidative stress (OS) due to the intensive cell metabolism. The objectives of this study were to investigate the cytoprotective effects of resveratrol against hydrogen peroxide- (H2O2-) induced OS in cultured bovine MECs (MAC-T). Pretreatment of MAC-T cells with resveratrol could rescue the decrease in cell viability and resulted in lower intracellular reactive oxygen species (ROS) accumulation after H2O2 exposure. Resveratrol helped MAC-T cells to prevent H2O2-induced endoplasmic reticulum stress and mitochondria-related cell apoptosis. Moreover, resveratrol induced mRNA expression of multiple antioxidant defense genes in MAC-T cells under normal/oxidative conditions. Nuclear factor erythroid 2-related factor 2 (Nrf2) was required for the cytoprotective effects on MAC-T cells by resveratrol, as knockdown of Nrf2 significantly abolished resveratrol-induced cytoprotective effects against OS. In addition, by using selective inhibitors, we further confirmed that the induction of Nrf2 by resveratrol was mediated through the prolonged activation of PI3K/Akt and ERK/MAPK pathways but negatively regulated by p38/MAPK pathway. Overall, resveratrol has beneficial effects on bovine MECs redox balance and may be potentially used as a therapeutic medicine against oxidative insult in lactating animals

    Derivation of Rabbit Embryonic Stem Cells from Vitrified–Thawed Embryos

    The rabbit is a useful animal model for regenerative medicine. We previously developed pluripotent rabbit embryonic stem cell (rbESC) lines using fresh embryos. We also successfully cryopreserved rabbit embryos by vitrification. In the present work, we combined these two technologies to derive rbESCs using vitrified?thawed (V/T) embryos. We demonstrate that V/T blastocysts (BLs) can be used to derive pluripotent rbESCs with efficiencies comparable to those using fresh BLs. These ESCs are undistinguishable from the ones derived from fresh embryos. We tested the developmental capacity of rbESCs derived from V/T embryos by BL injection experiments and produced chimeric kits. Our work adds cryopreservation to the toolbox of rabbit stem cell research and applications and will greatly expand the available research materials for regenerative medicine in a clinically relevant animal model.Peer Reviewedhttps://deepblue.lib.umich.edu/bitstream/2027.42/140328/1/cell.2015.0044.pd

    Propolis Reduces Phosphatidylcholine-Specific Phospholipase C Activity and Increases Annexin a7 Level in Oxidized-LDL-Stimulated Human Umbilical Vein Endothelial Cells

    To understand the mechanisms underlying the regulating dyslipidemia action of Chinese propolis and Brazilian green propolis, we investigated their effects on phosphatidylcholine-specific phospholipase C (PC-PLC) activity and annexin a7 (ANXA7) level which play crucial roles in the control of the progress of atherosclerosis. Furthermore, active oxygen species (ROS) levels, nuclear factor-KappaB p65 (NF-κB p65), and mitochondrial membrane potential (MMP) were also investigated in oxidized-LDL- (ox-LDL-) stimulated human umbilical vein endothelial cells (HUVECs). Our data indicated that the treatment of both types of propolis 12.5 μg/mL significantly increased cell viability and attenuated apoptosis rate, increased ANXA7 level, and decreased PC-PLC activity. Both types of propolis also inhibited ROS generation as well as the subsequent MMP collapse, and NF-κB p65 activation induced by ox-LDL in HUVECs. Our results also indicated that Chinese propolis and Brazilian green propolis had similar biological activities and prevented ox-LDL induced cellular dysfunction in HUVECs
