    Social media influence on viewer engagement

    Society is being reshaped through Social Media. The case of Social Media integration into the London 2012 Olympics mainstream media coverage is used to demonstrate this. Social Media has modified television viewing from a passive to an active activity. It has also allowed opinions to be shared more quickly and broadly. Analysis of practical examples through the theoretical lens of sociomateriality demonstrates that greater use of the components in the \u27Theoretical Framework of Member Needs\u27 reflect the presence of viewers who are more active. This paper discusses the inability to separate the use of technology from its social context, the power of Social Media and the impact of this on business use of Social Media. It also demonstrates a novel method for evaluating the benefits of emerging technologies in society

    Is \u27community\u27 important for Community Information Systems?

    Community information systems have the power to transform communities. However, without fully understanding the pre-requisite factors affecting community information system viability, and the complex relationships between these factors, communities struggle to manage such projects in a way that leads to viable systems that deliver real benefits. This paper develops and presents a Model of Community Information System Viability Pre-requisite Factors, based on both existing literature and the study of three community information system projects. This Model represents the generic factors that inform viability (i.e. leadership, active membership, funding, awareness, and system design and functionality), and also considers the impact of community context. This study argues that the viability of a Community Information System cannot be considered in isolation. All factors are directly impacted by the value of the Community Information System to the community. Management can also heavily impact on the success of a Community Information System

    Short-term genome stability of serial Clostridium difficile ribotype 027 isolates in an experimental gut model and recurrent human disease

Firstly, 62 isolates from 29 timepoints from three in vitro gut models, inoculated with a NAP1/027 strain. Secondly, 122 isolates from 44 patients (2–8 samples/patient) with mostly recurrent/on-going symptomatic NAP-1/027 C. difficile infection. Reference-based mapping was used to identify single nucleotide variants (SNVs).Across three gut model inductions, two with antibiotic treatment, total 137 days, only two new SNVs became established. Pre-existing minority SNVs became dominant in two models. Several SNVs were detected, only present in the minority of colonies at one/two timepoints. The median (inter-quartile range) [range] time between patients’ first and last samples was 60 (29.5–118.5) [0–561] days. Within-patient C. difficile evolution was 0.45 SNVs/called genome/year (95%CI 0.00–1.28) and within-host diversity was 0.28 SNVs/called genome (0.05–0.53). 26/28 gut model and patient SNVs were non-synonymous, affecting a range of gene targets.The consistency of whole genome sequencing data from gut model C. difficile isolates, and the high stability of genomic sequences in isolates from patients, supports the use of whole genome sequencing in detailed transmission investigations.Peer reviewe

    Epithelial-to-mesenchymal transition leads to disease-stage differences in circulating tumor cell detection and metastasis in pre-clinical models of prostate cancer

    Metastasis is the cause of most prostate cancer (PCa) deaths and has been associated with circulating tumor cells (CTCs). The presence of \u3e= 5 CTCs/7.5mL of blood is a poor prognosis indicator in metastatic PCa when assessed by the CellSearch (R) system, the gold standard clinical platform. However, similar to 35% of metastatic PCa patients assessed by CellSearch (R) have undetectable CTCs. We hypothesize that this is due to epithelial-to-mesenchymal transition (EMT) and subsequent loss of necessary CTC detection markers, with important implications for PCa metastasis. Two pre-clinical assays were developed to assess human CTCs in xenograft models; one comparable to CellSearch (R) (EpCAM-based) and one detecting CTCs semi-independent of EMT status via combined staining with EpCAM/HLA (human leukocyte antigen). In vivo differences in CTC generation, kinetics, metastasis and EMT status were determined using 4 PCa models with progressive epithelial (LNCaP, LNCaP-C42B) to mesenchymal (PC-3, PC-3M) phenotypes. Assay validation demonstrated that the CellSearch (R)-based assay failed to detect a significant number (similar to 40-50%) of mesenchymal CTCs. In vivo, PCa with an increasingly mesenchymal phenotype shed greater numbers of CTCs more quickly and with greater metastatic capacity than PCa with an epithelial phenotype. Notably, the CellSearch (R)-based assay captured the majority of CTCs shed during early-stage disease in vivo, and only after establishment of metastases were a significant number of undetectable CTCs present. This study provides important insight into the influence of EMT on CTC generation and subsequent metastasis, and highlights that novel technologies aimed at capturing mesenchymal CTCs may only be useful in the setting of advanced metastatic disease
