    Synergy between chemotherapeutic agents and CTLA-4 blockade in preclinical tumor models

    Ipilimumab, a cytotoxic T-lymphocyte antigen-4 (CTLA-4) binding agent, has proven to be an effective monotherapy for metastatic melanoma and has shown antitumor activity in trials when administered with other therapeutic agents. We hypothesized that the combination of ipilimumab with chemotherapeutic agents, such as ixabepilone, paclitaxel, etoposide, and gemcitabine, may produce therapeutic synergy based on distinct but complementary mechanisms of action for each drug and unique cellular targets. This concept was investigated using a mouse homolog of ipilimumab in preclinical murine tumor models, including SA1N fibrosarcoma, EMT-6 mammary carcinoma, M109 lung carcinoma, and CT-26 colon carcinoma. Results of CTLA-4 blockade in combination with one of various chemotherapeutic agents demonstrate that synergy occurs in settings where either agent alone was not effective in inducing tumor regression. Furthermore, when combined with CTLA-4 blockade, ixabepilone, etoposide, and gemcitabine elicited prolonged antitumor effects in some murine models with induction of a memory immune response. Future investigations are warranted to determine which specific chemo-immunotherapy combinations, if any, will produce synergistic antitumor effects in the clinical setting

    The debate on transgenic, which is embebed in asymmetrical information, has as a focal point, the risks and uncertainties to them associated. Many are the concerns and questions about the risks for the human health and the environment. Considering the interests of the productive sector in the transgenic technology, the matter sets itself in terms of how fast it wants to be settled taking into consideration the incapability of establishing certainties about the damages or benefits within the process. With the recent liberation by the Brazilian government for its production the sector starts showing segmentation. Therefore, this issue reflects in the market itself. The question is: are the consumers willing to buy transgenic? As an attempt to answer this question, this paper used a Contingent Valuation procedure, with an exploratory character, estimating consumer willingness to pay (WTP) to avoid the consumption of transgenic.     A randomized, phase II, three-arm study of two schedules of ixabepilone or paclitaxel plus bevacizumab as first-line therapy for metastatic breast cancer

    The aim of this phase II trial was to estimate the objective response rate (ORR) of two different schedules of ixabepilone [weekly or every 3 weeks (Q3W)] combined with bevacizumab, relative to a reference arm of weekly paclitaxel and bevacizumab. Patients with human epidermal growth factor receptor 2-normal, chemotherapy-naïve metastatic breast cancer (MBC) were randomized 3:3:2 to ixabepilone 16 mg/m(2) weekly plus bevacizumab 10 mg/kg Q2W (Arm A: n = 46); ixabepilone 40 mg/m(2) Q3W (reduced to 32 mg/m(2) after four cycles of treatment) plus bevacizumab 15 mg/kg Q3W (Arm B: n = 45); or paclitaxel 90 mg/m(2) weekly plus bevacizumab 10 mg/kg intravenous infusion Q2W (Arm C: n = 32). Of 123 randomized patients, 122 were treated. All were followed for ≥19 months; 5 % of patients remained on study treatment at the time of this analysis. Grade 3 or 4 neutropenia was more common in Arm B (60 %) than Arms A (16 %) or C (22 %); other adverse events were similar. The investigator-assessed ORR was 48, 71, and 63 % for Arms A, B, and C, respectively. Median progression-free survival (randomized patients) was 9.6 months in Arm A, 11.9 months in Arm B, and 13.5 months in Arm C. In conclusion, ixabepilone Q3W plus bevacizumab has clinical activity as first-line therapy for MBC relative to paclitaxel plus bevacizumab, but with significantly greater risk of grade 3 or 4 neutropenia. In addition, these data suggest that weekly dosing of ixabepilone may be less active than Q3W dosing, but with less neutropenia

    For Generations to Come: Chinese American Philanthropy in the Bay Area

    In order to understand better philanthropy among Chinese Americans in the San Francisco Bay Area, Silicon Valley Community Foundation commissioned this report, sponsored by the Chinese American Community Foundation in which active Chinese American donors were asked about their giving practices and preferences. Interviews were conducted with 33 Chinese American individuals who encompassed a range of ages, origins, and counties of residence in the Bay Area.To address the questions motivating this report, participants were asked to describe the values and vision that guide their charitable giving. They spoke about their appreciation for the opportunities they had, and their desire to give back by making the world a better place. Linking the past to the future, they voiced their aspirations for the next generations, especially of Chinese Americans, that they may have those same opportunities and eventually contribute to the community through their leadership and philanthropy. Their efforts emphasize creating opportunities for all, but especially for children, youth, and young adults, by supporting schools (both locally and in China), creating university scholarships, and serving on boards of universities and leadership development organizations.     Initial Efforts to Coordinate Appreciative Inquiry: Facilitators’ Experiences and Perceptions

    Appreciative inquiry (AI) is an alternative approach to action research that moves participants beyond problem solving and builds on existing strengths as the participants co-construct a positive vision of the future and move toward that vision through collaborative inquiry. Ph.D. students enrolled in a doctoral seminar on AI (who also are co-authors of the article) facilitated three separate AI projects. This article describes the AI projects, carried out at the school, district, and university levels, and identifies doctoral students’ experiences and perceptions as first-time facilitators of AI. The authors also make recommendations to others considering or initiating AI in educational settings
