    Detecting changes of spatial cone-excitation ratios in dichoptic viewing

    AbstractSpatial ratios of cone excitations produced by light reflected by different surfaces in a scene may provide the cue for discriminating changes in illuminant from changes in surface reflectances. To test whether these ratios can be computed across the two eyes, observers were presented with simulations on a computer-controlled monitor of pairs of juxtaposed or separated Munsell surfaces undergoing an illuminant change with a small change in cone-excitation ratios or a change with constant cone-excitation ratios. Surfaces were viewed either binocularly or dichoptically. Observers reliably discriminated the two changes in both viewing conditions, although less well dichoptically. Cone-excitation ratios, which may in principle be computed retinally, may also be computed cortically

    Human streptococcus agalactiae strains in aquatic mammals and fish

    <p>Background: In humans, Streptococcus agalactiae or group B streptococcus (GBS) is a frequent coloniser of the rectovaginal tract, a major cause of neonatal infectious disease and an emerging cause of disease in non-pregnant adults. In addition, Streptococcus agalactiae causes invasive disease in fish, compromising food security and posing a zoonotic hazard. We studied the molecular epidemiology of S. agalactiae in fish and other aquatic species to assess potential for pathogen transmission between aquatic species and humans.</p> <p>Methods: Isolates from fish (n = 26), seals (n = 6), a dolphin and a frog were characterized by pulsed-field gel electrophoresis, multilocus sequence typing and standardized 3-set genotyping, i.e. molecular serotyping and profiling of surface protein genes and mobile genetic elements.</p> <p>Results: Four subpopulations of S. agalactiae were identified among aquatic isolates. Sequence type (ST) 283 serotype III-4 and its novel single locus variant ST491 were detected in fish from Southeast Asia and shared a 3-set genotype identical to that of an emerging ST283 clone associated with invasive disease of adult humans in Asia. The human pathogenic strain ST7 serotype Ia was also detected in fish from Asia. ST23 serotype Ia, a subpopulation that is normally associated with human carriage, was found in all grey seals, suggesting that human effluent may contribute to microbial pollution of surface water and exposure of sea mammals to human pathogens. The final subpopulation consisted of non-haemolytic ST260 and ST261 serotype Ib isolates, which belong to a fish-associated clonal complex that has never been reported from humans.</p> <p>Conclusions: The apparent association of the four subpopulations of S. agalactiae with specific groups of host species suggests that some strains of aquatic S. agalactiae may present a zoonotic or anthroponotic hazard. Furthermore, it provides a rational framework for exploration of pathogenesis and host-associated genome content of S. agalactiae strains.</p&gt

    Development of a Biocompatible Layer-by-Layer Film System Using Aptamer Technology for Smart Material Applications

    Aptamers are short, single-stranded nucleic acids that fold into well-defined three dimensional (3D) structures that allow for binding to a target molecule with affinities and specificities that can rival or in some cases exceed those of antibodies. The compatibility of aptamers with nanostructures such as thin films, in combination with their affinity, selectivity, and conformational changes upon target interaction, could set the foundation for the development of novel smart materials. In this study, the development of a biocompatible aptamer-polyelectrolyte film system was investigated using a layer-by-layer approach. Using fluorescence microscopy, we demonstrated the ability of the sulforhodamine B aptamer to bind its cognate target while sequestered in a chitosan-hyaluronan film matrix. Studies using Ultraviolet-visible (UV-Vis) spectrophotometry also suggest that deposition conditions such as rinsing time and volume play a strong role in the internal film interactions and growth mechanisms of chitosan-hyaluronan films. The continued study and development of aptamer-functionalized thin films provides endless new opportunities for novel smart materials and has the potential to revolutionize the field of controlled release

    Spatial distributions of local illumination color in natural scenes

    In natural complex environments, the elevation of the sun and the presence of occluding objects and mutual reflections cause variations in the spectral composition of the local illumination across time and location. Unlike the changes in time and their consequences for color appearance and constancy, the spatial variations of local illumination color in natural scenes have received relatively little attention. The aim of the present work was to characterize these spatial variations by spectral imaging. Hyperspectral radiance images were obtained from 30 rural and urban scenes in which neutral probe spheres were embedded. The spectra of the local illumination at 17 sample points on each sphere in each scene were extracted and a total of 1904 chromaticity coordinates and correlated color temperatures (CCTs) derived. Maximum differences in chromaticities over spheres and over scenes were similar. When data were pooled over scenes, CCTs ranged from 3000 K to 20,000 K, a variation of the same order of magnitude as that occurring over the day.     The Swinburne Intermediate Latitude Pulsar Survey

    We have conducted a survey of intermediate Galactic latitudes using the 13-beam 21-cm multibeam receiver of the Parkes 64-m radio telescope. The survey covered the region enclosed by 5 deg < |b| < 15 deg and -100 deg < l < 50 deg with 4,702 processed pointings of 265 s each, for a total of 14.5 days of integration time. Thirteen 2x96-channel filterbanks provided 288 MHz of bandwidth at a centre frequency of 1374 MHz, one-bit sampled every 125 microsec and incurring ~DM/13.4 cm^-3 pc samples of dispersion smearing. The system was sensitive to slow and most millisecond pulsars in the region with flux densities greater than approximately 0.3--1.1 mJy. Offline analysis on the 64-node Swinburne workstation cluster resulted in the detection of 170 pulsars of which 69 were new discoveries. Eight of the new pulsars, by virtue of their small spin periods and period derivatives, may be recycled and have been reported elsewhere. The slow pulsars discovered are typical of those already known in the volume searched, being of intermediate to old age. Several pulsars experience pulse nulling and two display very regular drifting sub-pulses. We discuss the new discoveries and provide timing parameters for the 48 slow pulsars for which we have a phase-connnected solution.Comment: 19 pages, 11 figures, accepted to MNRA

    Smart materials based on DNA aptamers: Taking aptasensing to the next level

    "Smart" materials are an emerging category of multifunctional materials with physical or chemical properties that can be controllably altered in response to an external stimulus. By combining the standard properties of the advanced material with the unique ability to recognize and adapt in response to a change in their environment, these materials are finding applications in areas such as sensing and drug delivery. While the majority of these materials are responsive to physical or chemical changes, a particularly exciting area of research seeks to develop smart materials that are sensitive to specific molecular or biomolecular stimuli. These systems require the integration of a molecular recognition probe specific to the target molecule of interest. The ease of synthesis and labeling, low cost, and stability of DNA aptamers make them uniquely suited to effectively serve as molecular recognition probes in novel smart material systems. This review will highlight current work in the area of aptamer-based smart materials and prospects for their future applications

    Increasing rates of fluoroquinolone resistance in escherichia coli isolated from the blood and urine of patients with hematologic malignancies and stem cell transplant recipients

    Fluoroquinolone (FQ) antibiotics have been shown to reduce mortality and the number of febrile episodes when used as prophylaxis during neutropenia. Prior studies suggest that prophylaxis may result in increasing rates of FQ resistance. Fluoroquinolone non-susceptibility trends in Escherichia coli isolated from blood and urine cultures were evaluated over a 16-year period during which prophylaxis was initiated in patients with hematologic malignancies and stem cell transplants. Non-susceptibility rates increased after the introduction of prophylaxis, with yearly non-susceptibility rates rising from 30%–33% to 40%–88% in blood isolates. The high rates of non-susceptibility now observed raise concerns about the continued efficacy of FQ prophylaxis. This concern exists particularly in those patients undergoing stem cell transplants where the total FQ non-susceptibility rates over the study period were 82.3%. Further evaluation of the effect of FQ prophylaxis on antibiotic resistance and its efficacy in the setting of increased rates of resistance is warranted
