    Origins and evolution of stomatal development

    The fossil record suggests stomata-like pores were present on the surfaces of land plants over 400 million years ago. Whether stomata arose once or whether they arose independently across newly evolving land plant lineages has long been a matter of debate. In Arabidopsis, a genetic toolbox has been identified that tightly controls stomatal development and patterning. This includes the basic helix-loop-helix (bHLH) transcription factors SPEECHLESS (SPCH), MUTE, FAMA, and ICE/SCREAMs (SCRMs), which promote stomatal formation. These factors are regulated via a signaling cascade, which includes mobile EPIDERMAL PATTERNING FACTOR (EPF) peptides to enforce stomatal spacing. Mosses and hornworts, the most ancient extant lineages to possess stomata, possess orthologs of these Arabidopsis (Arabidopsis thaliana) stomatal toolbox genes, and manipulation in the model bryophyte Physcomitrella patens has shown that the bHLH and EPF components are also required for moss stomatal development and patterning. This supports an ancient and tightly conserved genetic origin of stomata. Here, we review recent discoveries and, by interrogating newly available plant genomes, we advance the story of stomatal development and patterning across land plant evolution. Furthermore, we identify potential orthologs of the key toolbox genes in a hornwort, further supporting a single ancient genetic origin of stomata in the ancestor to all stomatous land plants

    Stomata and sporophytes of the model moss physcomitrium patens

    Mosses are an ancient land plant lineage and are therefore important in studying the evolution of plant developmental processes. Here, we describe stomatal development in the model moss species Physcomitrium patens (previously known as Physcomitrella patens) over the duration of sporophyte development. We dissect the molecular mechanisms guiding cell division and fate and highlight how stomatal function might vary under different environmental conditions. In contrast to the asymmetric entry divisions described in Arabidopsis thaliana, moss protodermal cells can enter the stomatal lineage directly by expanding into an oval shaped guard mother cell (GMC). We observed that when two early stage P. patens GMCs form adjacently, a spacing division can occur, leading to separation of the GMCs by an intervening epidermal spacer cell. We investigated whether orthologs of Arabidopsis stomatal development regulators are required for this spacing division. Our results indicated that bHLH transcription factors PpSMF1 and PpSCRM1 are required for GMC formation. Moreover, the ligand and receptor components PpEPF1 and PpTMM are also required for orientating cell divisions and preventing single or clustered early GMCs from developing adjacent to one another. The identification of GMC spacing divisions in P. patens raises the possibility that the ability to space stomatal lineage cells could have evolved before mosses diverged from the ancestral lineage. This would have enabled plants to integrate stomatal development with sporophyte growth and could underpin the adoption of multiple bHLH transcription factors and EPF ligands to more precisely control stomatal patterning in later diverging plant lineages. We also observed that when P. patens sporophyte capsules mature in wet conditions, stomata are typically plugged whereas under drier conditions this is not the case; instead, mucilage drying leads to hollow sub-stomatal cavities. This appears to aid capsule drying and provides further evidence for early land plant stomata contributing to capsule rupture and spore release

    Australian livestock export industry workers’ attitudes toward animal welfare

    Understanding live export industry workers’ attitudes and beliefs toward animal welfare can provide insight into their decision-making processes and likely behavior. Industry workers (n = 265) with various roles within the supply chain were surveyed from different global regions. Participants were divided into ten categories according to their industry roles and compared using ordinal regression. Respondents were highly likely to have a positive attitude toward animal welfare; the majority of workers enjoyed working with livestock (95.8%) and agreed that livestock should be treated with respect (97.7%). Workers demonstrated a strong understanding of animal welfare concepts, 168 respondents (63.4%) provided examples of ways they had improved animal welfare in their workplace, and 164 workers (61.9%) suggested ways that animal welfare could be improved further. Most workers (95.8%) agreed that animal welfare was satisfactory in their workplace. Five out of the 24 multiple-choice responses differed significantly by the participant’s industry role, but no particular group displayed consistently divergent beliefs or attitudes. Given the community concern regarding animals in the livestock export supply chain, it is imperative to understand the attitudes of industry personnel who are responsible for the daily management of the animals. This knowledge assists in the development of animal welfare policy and can inform strategies to manage public perception

    The molecular mechanisms regulating the biogenesis of the first desmosomes to form during mouse embryogenesis have been studied. A sensitive modification of a reverse transcriptase-cDNA amplification procedure has been used to detect transcripts of the desmosomal adhesive cadherin, desmocollin. Sequencing of cDNA amplification products confirmed that two splice variants, a and b, of the DSC2 gene are transcribed coordinately. Transcripts were identified in unfertilized eggs and cumulus cells and in cleavage stages up to the early 8-cell stage, were never detected in compact 8-cell embryos, but were evident again either from the 16-cell morula or very early blastocyst (approx 32-cells) stages onwards. These two phases of transcript detection indicate DSC2 is encoded by maternal and embryonic genomes. Previously, we have shown that desmocollin protein synthesis is undetectable in eggs and cleavage stages but initiates at the early blastocyst stage when desmocollin localises at, and appears to regulate assembly of, nascent desmosomes that form in the trophectoderm but not in the inner cell mass (Fleming, T. P., Garrod, D. R. and Elsmore, A. J. (1991), Development 112, 527–539). Maternal DSC2 mRNA is therefore not translated and presumably is inherited by blastomeres before complete degradation. Our results suggest, however, that initiation of embryonic DSC2 transcription regulates desmocollin protein expression and thereby desmosome formation. Moreover, data from blastocyst single cell analyses suggest that embryonic DSC2 transcription is specific to the trophectoderm lineage. Inhibition of E-cadherin-mediated cell-cell adhesion did not influence the timing of DSC2 embryonic transcription and protein expression. However, isolation and culture of inner cell masses induced an increase in the amount of DSC2 mRNA and protein detected. Taken together, these results suggest that the presence of a contact-free cell surface activates DSC2 transcription in the mouse early embryo. <br/

    Animal welfare indicators for sheep during sea transport: The effect of voyage day and time of day

    Ensuring the well-being of animals during transport is becoming an increasingly important societal concern. The Australian livestock export industry recognises the need for comprehensive monitoring and reporting on animal welfare during sea transport. It is predicted that pen-side assessments of sheep can be used to monitor environmental conditions, resource access, and animal health and behavioural outcomes throughout a sea voyage. Pen-side assessments by observation are non-invasive and practical to apply in an industry setting. This study monitored sheep using a pilot list of welfare indicators during two sea voyages from Australia to the Middle East, in contrasting seasons. Sheep behaviour, environment and resources were recorded three times daily via pen-side observations of six pens of Merino wethers (castrated males), repeated over three decks for each voyage. Behavioural outcomes were examined for the effect of sampling frequency on group assessments. The number of behavioural measures were reduced via Principal Component (PC) analysis. The primary three PC factors were tested against the time of sampling and pen location after accounting for the effect of environmental- and resource-based predictor variables. PC 1 (24.0 % of the total variance) described activity levels, with sheep on Voyage B being more active in the morning and resting or recumbent in the middle of the afternoon and evening. PC 2 (14.7 %) reflected heat responses with the majority of the variation in these data accounted for by changes in Wet Bulb Globe Temperature (WBGT) and manure pad moisture. The heat responses described by PC 2 also varied by voyage day (p < 0.001) and time point (p < 0.001). PC 3 scores (9.5 %) reflected flight distances and feeding behaviour and strongly correlated to WBGT and pellet consumption per head per day. Feeding behaviour generally became more competitive, and flight distances reduced as both voyages progressed. Results indicate that a comprehensive welfare monitoring protocol requires repeated daily sampling throughout a voyage. The findings of this study are pertinent for developing a sampling strategy to assess sheep welfare during sea transport

    Review of livestock welfare indicators relevant for the Australian live export industry

    Animal welfare is an important issue for the live export industry (LEI), in terms of economic returns, community attitudes and international socio-political relations. Mortality has traditionally been the main welfare measure recorded within the LEI; however, high mortality incidents are usually acted upon after adverse events occur, reducing the scope for proactive welfare enhancement. We reviewed 71 potential animal welfare measures, identifying those measures that would be appropriate for use throughout the LEI for feeder and slaughter livestock species, and categorised these as animal-, environment- and resource-based. We divided the live export supply chain into three sectors: (1) Australian facilities, (2) vessel and (3) destination country facilities. After reviewing the relevant regulations for each sector of the industry, we identified 38 (sector 1), 35 (sector 2) and 26 (sector 3) measures already being collected under current practice. These could be used to form a ‘welfare information dashboard’: a LEI-specific online interface for collecting data that could contribute towards standardised industry reporting. We identified another 20, 25 and 28 measures that are relevant to each LEI sector (sectors 1, 2, 3, respectively), and that could be developed and integrated into a benchmarking system in the future

    Mode-hop-free tuning over 135 GHz of external cavity diode lasers without anti-reflection coating

    We report an external cavity diode laser (ECDL), using a diode whose front facet is not antireflection (AR) coated, that has a mode-hop-free (MHF) tuning range greater than 135 GHz. We achieved this using a short external cavity and by simultaneously tuning the internal and external modes of the laser. We find that the precise location of the pivot point of the grating in our laser is less critical than commonly believed. The general applicability of the method, combined with the compact portable mechanical and electronic design, makes it well suited for both research and industrial applications.Comment: 5 pages, 5 figure

    Notes on a scandal: the official enquiry into deviance and corruption in New Zealand police

    Since 2004, the New Zealand Police Service has been engulfed by a series of scandals relating to allegations that officers have committed rape and sexual assault and conducted inappropriate sexual relations with vulnerable people. Moreover, it has been claimed that other officers engaged in corrupt practices to thwart the investigation and prosecution of criminal behaviour of police officers. In 2007, a Commission of Inquiry report established a program of reform intended to shape the future direction of the police service. This article provides an overview of these scandals, the context in which they have emerged, and the political and policing response to them. The analysis contained in the Commission report is compared with that offered by comparable investigations of police deviance and corruption in other countries. The methodological and conceptual limitations of the Commission are outlined and the prospects of the recommendations are considered
