Biossorção de íons metálicos presentes nas águas de efluentes de indústrias químicas.
As atividades industriais têm introduzido metais nas águas numa quantidade
muito maior do que aquela que seria natural, causando grandes poluições. Os
tratamentos convencionais (redução química, troca iônica, ultrafiltração e
osmose inversa), normalmente usados para a remoção de metais dos efluentes
líquidos, apresentam algumas desvantagens, pois além de serem, na maioria,
processos caros, não conseguem remover totalmente os íons metálicos. A
necessidade de tratamentos eficientes e econômicos para remoção de íons
metálicos de efluentes tem resultado no desenvolvimento de novas tecnologias.
Muitas destas descrevem técnicas envolvendo o uso de bactérias, fungos,
microalgas, e macroalgas. A biossorção é o processo no qual sólidos de origem
biológica ou seus derivados são usados na retenção de íons metálicos de um
ambiente aquoso. Comparadas às metodologias convencionais para a remoção
de íons metálicos de efluentes industriais, o processo de biossorção tem como
principais vantagens o baixo custo operacional e alta seletividade. Dentre os
microrganismos mais utilizados destacam-se os fungos, que são utilizados em
uma variedade de processos industriais servindo como fonte constante e
econômica de suprimento de biomassa para remoção de íons metálicos. Sendo
o Brasil, o maior produtor mundial de álcool etílico via processo fermentativo
utilizando-se da Saccharomyces cerevisiae (levedura) como o microrganismo
agente da fermentação, é prática comum nas indústrias de produção de álcool
etílico, no Brasil, a sangria do creme de levedura, que consiste em retirar parte
do creme do processo de centrifugação. Desta forma, a Saccharomyces
cerevisiae é uma fonte excedente do processo de fermentação que pode ser
utilizada, por exemplo, como bioadsorvente de íons metálicos em processos de
descontaminação ambiental. Experimentos de biossorção de íons metálicos
utilizando a Saccharomyces cerevisiae para remoção do Cd2+ e Pb2+ foram
realizados para investigar os fatores que influenciam e otimizam o processo de
biossorção. Através do estudo da cinética estático verificou-se que o tempo 48h
já é suficiente para o processo alcançar o equilíbrio. Para a cinética dinâmica a
partir de 90 min não ocorreu mais alteração no valor da concentração final dos
dois íons metálicos, portanto, sendo este tempo suficiente para o sistema
alcançar o equilíbrio num sistema operando em condições dinâmicas. A técnica
de planejamento experimental foi utilizada para avaliar os efeitos das variáveis
quantidade de biomassa, concentração dos íons metálicos, temperatura, pH e
estado da biomassa (viva ou morta), que influenciam no processo Os efeitos da
quantidade de biomassa e concentração inicial dos íons metálicos estudados
foram as variáveis que apresentaram mais efeitos significativos quando
comparado com o efeito do pH, estado da biomassa (viva ou morta) e
temperatura nas condições avaliadas. O modelo que melhor ajustou o processo
foi o de Langmuir com valores de qmax de 210,5 mg.g-1 para o cádmio e 1486,88
mg.g-1 para o chumbo. A levedura imobilizada apresentou uma certa queda na
sua eficiência de remoção observando-se redução média na eficiência de q de
78,5% para o Cd2+ e de 73,92% para o Pb2+.The industrial activities have introduced metals in waters in quantities which are
greater than those found naturally, thereby causing heavy pollutions. The
conventional treatments (chemical reduction, ionic exchange, ultrafiltration and
inverse osmosis), normally used for the metal removal of the liquid effluents,
present some disadvantages, because majority of them apart from being
expensive processes are not capable to remove metal ions compleatly. The
necessity of efficient and economic treatments for removal of metallic ions from
effluents has resulted in the development of new technologies. Many of these
techniques are based on the use of bacteria, fungi, microseaweed, and
macroseaweed. The biosorption is a process in which solids of natural origin or
its derivatives are used in the retention of metallic ions from an aqueous
medium. The biosorption process has as main advantages the low operational
cost and high selectivity when compared to the conventional methods for the
removal of metallic ions from industrial effluents. Amongst the used
microorganisms fungi are more distinguished, as these are used in a variety of
industrial processes and serves as constant and economic source of
supplement of biomass for removal of metallic ions. Brazil being a world-wide
big producer of ethyl alcohol by a fermentative process that uses the
Saccharomyces cerevisiae (ferment) as the microorganism, it is a common
practice in the industries of alcohol to remove the excess of the ferment cream
by centrifuging. This way, the excessive Saccharomyces cerevisiae of the
fermentation process is a source that can be used, for example, as biosorbent
of metallic íons for the decontamination of environment. Experiments of
biosorption of metallic íons using the Saccharomyces cerevisiae for removal of
the Cd2+ and Pb2+ had been carried out to investigate the factors that
influence and optimize the biosorption process. Through the study of the static
kinetics it was verified that a period of 48 hours is enough for the process to
reach the equilibrium. For the dynamic kinetics after 90 min no more change in
the final concentration of the two metallic ions occurred. Therefore, this time is
enough for the system to reach the equilibrium when the process is operating in
dynamic conditions. The design of experiment technique was used to evaluate
the effect of the variables: biomass amount, metallic ions concentrations,
temperature, pH and state of the biomass (alive or dead). The effects of the
amount of biomass amount and initial metallic ion concentration were the
variables that presented more significant effects when compared with the effect
of pH and state of the biomass (alive or dead) under the evaluated conditions.
The Langmuir and Freundlich models were used to adjust to the experimental
data of the adsorption isotherms of metal ions studied. The model that adjusted
better the adsorption isotherms was of Langmuir having qmax values as 210.5
mg.g-1 for cadmium and 1486.88 mg.g-1 (Langmuir not linearized) for the lead.
The immobilized ferment presented a decrease in its efficiency of metal ion
removal. In the study for the immobilized ferment an average efficiency of the
adsorbed amount per unit of mass of 78.5% for the Cd2+ and 73.92% for the
Pb2+ was observed.CNP
Como fazer a diferença? Relato de uma intervenção em psicologia organizacional realizada por uma empresa júnior em um hospital filantrópico
This article presents the report of an intervention in Organizational and Labor Psychology in a philanthropic hospital, performed by Perfil, the junior company of the Psychology program of the Federal University of São João del-Rei (UFSJ). For this, an organizational diagnosis was performed by means of focus groups with employees, and interviews were conducted with the managers of the institution, so as to identify intervention needs and perspectives which did not appear in the initial demand. The intervention actions were based on the motivational theory of Abraham Maslow, as well as on concepts of leadership, internal communication, and feedback. This work highlights the relevance of providing greater visibility to the activities developed by junior companies to non-academic audiences, praising the knowledge produced within the academy in exchange with the labor market, and attempting to show how it is possible for an organizational and labor psychology initiative to make a difference in a health care institution.Este artigo traz o relato de uma intervenção em Psicologia Organizacional e do Trabalho, realizada pela Perfil, empresa júnior do curso de Psicologia da Universidade Federal de São João del-Rei (UFSJ), num hospital filantrópico. Para isso, foi realizado um diagnóstico organizacional, por meio de grupos focais, com os funcionários e entrevistas com os gestores da instituição, de modo a se identificar necessidades de intervenção e perspectivas que não apareceram na demanda inicial. As ações interventivas tiveram como base a teoria motivacional de Abraham Maslow, além de conceitos sobre liderança, comunicação interna e feedback. Este trabalho ressalta a pertinência de se dar maior visibilidade às atividades desenvolvidas pelas empresas juniores para o público não-acadêmico, valorizando o conhecimento produzido dentro da academia em intercâmbio com o mercado de trabalho, e procurando demonstrar como é possível uma iniciativa de Psicologia Organizacional e do Trabalho fazer a diferença numa instituição de saúde
Parenteral adjuvant effects of an enterotoxigenic Escherichia coli natural heat-labile toxin variant
Native type I heat-labile toxins (LTs) produced by enterotoxigenic Escherichia coli (ETEC) strains exert strong adjuvant effects on both antibody and T cell responses to soluble and particulate antigens following co-administration via mucosal routes. However, inherent enterotoxicity and neurotoxicity (following intra-nasal delivery) had reduced the interest in the use of these toxins as mucosal adjuvants. LTs can also behave as powerful and safe adjuvants following delivery via parenteral routes, particularly for activation of cytotoxic lymphocytes. In the present study, we evaluated the adjuvant effects of a new natural LT polymorphic form (LT2), after delivery via intradermal (i.d.) and subcutaneous (s.c.) routes, with regard to both antibody and T cell responses. A recombinant HIV-1 p24 protein was employed as a model antigen for determination of antigen-specific immune responses while the reference LT (LT1), produced by the ETEC H10407 strain, and a non-toxigenic LT form (LTK63) were employed as previously characterized LT types. LT-treated mice submitted to a four dose-base immunization regimen elicited similar p24-specific serum IgG responses and CD4(+) T cell activation. Nonetheless, mice immunized with LT1 or LT2 induced higher numbers of antigen-specific CD8(+) T cells and in vivo cytotoxic responses compared to mice immunized with the non-toxic LT derivative. These effects were correlated with stronger activation of local dendritic cell populations. In addition, mice immunized with LT1 and LT2, but not with LTK63, via s.c. or i.d. routes developed local inflammatory reactions. Study of optimal conditions by the response surface analysis method for acetic fermentation efficiency.
A fermentação acética e um processo de oxidação realizada por um conjunto de
bactérias do gênero Acetobacter. O acido acético produzido por bactérias desse
gênero e o composto principal do vinagre. A produção de um bom vinagre depende de
uma serie de fatores como concentração de álcool, temperatura, oxigenação, pH e
nutrientes, Este trabalho teve por objetivo estudar em escala de bancada as melhores
condições da fermentação acética em batelada, processo muito utilizado nas
industrias de pequeno porte do Estado da Paraíba na produção do vinagre.
Inicialmente, realizou-se um estudo cinético da fermentação acética com a intenção de
determinar a influencia das variáveis: concentração de etanol, concentração de fosforo
e concentração de nitrogênio sobre o processo em diferentes intervalos de tempo.
Para esse estudo, realizou-se ensaios com concentrações de etanol de 2, 4, 6 e 8%,
concentrações de fosforo de 0,05 e 0,2g.mr1, e concentrações de nitrogênio de 0,5 e
2,0 g.ml"1. Através da cinética não foi possível observar conclusivamente a influencia
das variáveis estudadas no processo. Foi utilizado a metodologia de planejamento
fatorial completo 23, onde as variáveis de entrada foram: concentração inicial de
etanol, concentração de fosforo e concentração de nitrogênio. Foram realizados
ensaios com concentrações de etanol de 3, 6 e 9%, em volume, concentrações de
fosforo de 0,01, 0,03 e 0,05 g.ml"1 e concentrações de nitrogênio de 0,1 , 0,3 e 0,5
g.ml"1. Apos as analises foi necessário elaborar um outro planejamento, ampliando as
faixas das variáveis, desta vez o planejamento foi do tipo 22 mais configuração estrela.
Através dos modelos matemáticos empíricos obtidos e analise de superfície de
resposta verificou-se que a variável controlada, dentre as estudadas, que apresentou
maior influencia sobre o rendimento e produtividade do processo, foi a concentração
inicial de etanol, que apresentou um valor ótimo em 4%. O nitrogênio apresentou uma
discreta influencia a uma concentração de 1,7 g.ml"1 enquanto que o fosforo não
exerceu nenhuma influencia perceptível. Foi possível também observar, através dos
dados da analise de variância, que o modelo empírico encontrado e estatisticamente
significativo.The acetic fermentation is an oxidation process accomplished by a group of
bacterias of the gender Acetobacter. The acetic acid produced by bacterias of this
gender is the main compound of the vinegar. The production of a good vinegar
depends on a series of factors as concentration of alcohol, temperature, oxygenation,
pH and nutritious. This work had objective to study on bench scale, the optimum
conditions of acetic acid fermentation, a process very much used in small industries of
the state of Paraiba in the production of the vinegar. Initially a kinetic study of the
acetic fermentation was performed with the intention of determining the influence of the
variables: ethanol concentration, phosphorous concentration and concentration of
nitrogen on the process in different intervals of time. For this a study with
concentrations of ethanol of 2%, 4%, 6% and 8%, of phosphorous 0,05g/ml and
0,2g/ml, and of nitrogen of 0,5g/ml and 2,0g/ml, was made. Through the kinetics, it was
not possible to observe conclusively the influence of the variables of the process
studied. The methodology of complete factorial 23 planning was used, where the
entrance variables were initial concentrations of ethanol, phosphorous and nitrogen.
Tests were, accomplished with concentrations of ethanol 3%, 6% and 9%, in volume,
concentrations of phosphorus 0,01 g/ml, 0,03g/ml and 0,05g/ml and concentrations of
nitrogen 0,1 g/ml,0,3g/ml and 0,5g/ml. After the analyses it was necessary to elaborate
another planning, enlarging the range of the variables. This time the planning was of
the type 22 plus star configuration. Through the empirical mathematical models
obtained and analysis of surface response it was verified that the controlled variable
that presented great influence on the yield and productivity of the process was initial
concentration of ethanol which has an optimum value of 4%. The nitrogen presented a
discreet influence at the concentration of 1,7g/ml while the phosphorus didn't show any
perceptible influence. It was also possible to observe, through the data of the variance
analysis, that the empirical model observed is statistically significant
Variables in the process of coagulation / flocculation / settling of leachate of municipal landfills
The careless disposal of waste may generate tremendous environmental and social impacts. For the treatment of landfill leachate, biological and physic-chemical treatments are routinely used. Chemical precipitation using calcium hydroxide has been used with great effectiveness in treating effluent with high concentrations of organic compounds and heavy metals. The leachate used in this study was collected from a flow box of the Muribeca landfill, located in the city of Jaboatão Guararapes, PE. Commercial calcium hydroxide was used as coagulant in aqueous solution. The experiment was implemented using a fractional factorial design, followed by a complete planning 24. It was observed that the lowest turbidity and color were obtained at the highest flocculation speeds. Color removal was favored by long flocculation time whereas this did not occur for turbidity removal. For the optimal point, a reduction of 52% of the color and a NTU turbidity within the standards established in Brazilian Laws (CONAMA 357/05 and CONAMA 430/11) were obtained

The biosorption, based on the use of biomass for removal of ions is distinguished as an innovative and promising technology when compared with the traditional methods. In this context, the aim of the present work is to use Saccharomyces cerevisiae as biosorbent for the retention of Pb2+ metal ions. Factorial design was used for evaluation of the process. The observed equilibrium data were well described by Langmuir and Freundlich adsorption isotherms. The maximum adsorption capacity was 1486.88 mg/g. The results indicated that Saccharomyces cerevisiae is suitable for biosorption of Pb2+ metal ions
