TEAD1-dependent expression of the FoxO3a gene in mouse skeletal muscle
<p>Abstract</p> <p>Background</p> <p><it>TEAD1 </it>(TEA domain family member 1) is constitutively expressed in cardiac and skeletal muscles. It acts as a key molecule of muscle development, and trans-activates multiple target genes involved in cell proliferation and differentiation pathways. However, its target genes in skeletal muscles, regulatory mechanisms and networks are unknown.</p> <p>Results</p> <p>In this paper, we have identified 136 target genes regulated directly by TEAD1 in skeletal muscle using integrated analyses of ChIP-on-chip. Most of the targets take part in the cell process, physiology process, biological regulation metabolism and development process. The targets also play an important role in MAPK, mTOR, T cell receptor, JAK-STAT, calcineurin and insulin signaling pathways. TEAD1 regulates <it>foxo3a </it>transcription through binding to the M-CAT element in <it>foxo3a </it>promoter, demonstrated with independent ChIP-PCR, EMSA and luciferase reporter system assay. In addition, results of over-expression and inhibition experiments suggest that <it>foxo3a </it>is positively regulated by TEAD1.</p> <p>Conclusions</p> <p>Our present data suggests that TEAD1 plays an important role in the regulation of gene expression and different signaling pathways may co-operate with each other mediated by TEAD1. We have preliminarily concluded that TEAD1 may regulate <it>FoxO3a </it>expression through calcineurin/MEF2/NFAT and IGF-1/PI3K/AKT signaling pathways in skeletal muscles. These findings provide important clues for further analysis of the role of <it>FoxO3a </it>gene in the formation and transformation of skeletal muscle fiber types.</p
Distribution and Abundance of Archaea in South China Sea Sponge Holoxea sp. and the Presence of Ammonia-Oxidizing Archaea in Sponge Cells
Compared with bacterial symbionts, little is known about archaea in sponges especially about their spatial distribution and abundance. Understanding the distribution and abundance of ammonia-oxidizing archaea will help greatly in elucidating the potential function of symbionts in nitrogen cycling in sponges. In this study, gene libraries of 16S rRNA gene and ammonia monooxygenase subunit A (amoA) genes and quantitative real-time PCR were used to study the spatial distribution and abundance of archaea in the South China Sea sponge Holoxea sp. As a result, Holoxea sp. specific AOA, mainly group C1a (marine group I: Crenarchaeota) were identified. The presence of ammonia-oxidizing crenarchaea was observed for the first time within sponge cells. This study suggested a close relationship between sponge host and its archaeal symbionts as well as the archaeal potential contribution to sponge host in the ammonia-oxidizing process of nitrification
Measuring the regional availability of forest biomass for biofuels and the potential of GHG reduction
Forest biomass is an important resource for producing bioenergy and reducing greenhouse gas (GHG) emissions. The State of Michigan in the United States (U.S.) is one region recognized for its high potential of supplying forest biomass; however, the long-term availability of timber harvests and the associated harvest residues from this area has not been fully explored. In this study time trend analyses was employed for long term timber assessment and developed mathematical models for harvest residue estimation, as well as the implications of use for ethanol. The GHG savings potential of ethanol over gasoline was also modeled. The methods were applied in Michigan under scenarios of different harvest solutions, harvest types, transportation distances, conversion technologies, and higher heating values over a 50-year period. Our results indicate that the study region has the potential to supply 0.75–1.4 Megatonnes (Mt) dry timber annually and less than 0.05 Mt of dry residue produced from these harvests. This amount of forest biomass could generate 0.15–1.01 Mt of ethanol, which contains 0.68–17.32 GJ of energy. The substitution of ethanol for gasoline as transportation fuel has potential to reduce emissions by 0.043–1.09 Mt CO2eq annually. The developed method is generalizable in other similar regions of different countries for bioenergy related analyses
The synthesis and characterization of a xanthan gum-acrylamide-trimethylolpropane triglycidyl ether hydrogel
peer-reviewedTo improve the thermal stability and adsorption performance, xanthan gum was modified with acrylamide and trimethylolpropane triglycidyl ether (TTE). The modified xanthan gum (XGTTE) was characterized by Fourier transform infrared (FT-IR) spectroscopy, X-ray diffractogram (XRD), differential scanning calorimetry (DSC) and scanning electron microscopy (SEM). The characteristic peaks at 3449, 1655, 1611 and 1420 cm−1 in the FT-IR confirm the modification. The XGTTE crystal grew well upon addition of TTE. The XRD and DSC data revealed that the XGTTE enhanced its thermal stability. Analysis of SEM revealed that the grafting introduced major changes on the microstructure making it porous and resulting in the adsorption of crystal violet (CV) with flocculation. The CV adsorption capacity of the hydrogel with different dosages of TTE (XGTTE2, XGTTE3, XGTTE4, XGTTE5 and XGTTE6) were between 28.13 with 35.12 mg/g. Annexin A2 Coordinates STAT3 to Regulate the Invasion and Migration of Colorectal Cancer Cells In Vitro
The present study aimed to reveal the expression of STAT3 and Anxa 2 in CRC specimens and to investigate the effects of STAT3 and Anxa 2 signaling on the proliferation, invasion, and migration in CRC Caco-2 cells. Results demonstrated that both Anxa 2 and STAT3 were highly expressed in CRC specimens in both mRNA and protein levels, with or without phosphorylation (Tyrosine 23 in Anxa 2 and Tyrosine 705 in STAT3). And the upregulated Anxa 2 promoted the phosphorylation of STAT3 (Tyrosine 705) in CRC Caco-2 cells. The upregulated Anxa 2 promoted the proliferation, migration, and invasion of Caco-2 cells in vitro. Moreover, the STAT3 knockdown also repressed the proliferation, migration, and invasion of Caco-2 cells. In conclusion, the overexpressed Annexin A2 regulated the proliferation, invasion, and migration in CRC cells in an association with STAT3

Moreover, when the quantum state belongs to a set composed of two states, a
probabilistic teleportation scheme is presented using a non-maximally entangled
