    Argonaute 2 drives miR-145-5p-dependent gene expression program in breast cancer cells

    To perform their regulatory functions, microRNAs (miRNAs) must assemble with any of the four mammalian Argonaute (Ago) family of proteins, Ago1–4, into an effector complex known as the RNA-induced silencing complex (RISC). While the mature miRNA guides the RISC complex to its target mRNA, the Ago protein represses mRNA translation. The specific roles of the various Ago members in mediating miRNAs activity, however, haven’t been clearly established. In this study, we investigated the contribution of Ago2, the only human Ago protein endowed with nuclease activity, to the function of tumor-suppressor miR-145-5p in breast cancer (BC). We show that miR-145-5p and Ago2 protein are concomitantly downregulated in BC tissues and that restoration of miR-145-5p expression in BC cells leads to Ago2 protein induction through the loosening of Ago2 mRNA translational repression. Functionally, miR-145-5p exerts its inhibitory activity on cell migration only in presence of Ago2, while, upon Ago2 depletion, we observed increased miR-145/Ago1 complex and enhanced cell motility. Profiling by microarray of miR-145-5p target mRNAs, in BC cells depleted or not of Ago2, revealed that miR-145-5p drives Ago2-dependent and -independent activities. Our results highlight that the Ago2 protein in cancer cells strictly dictates miR-145-5p tumor suppressor activity

    The effects of Glycine max L. (SOY) vs. Pisum sativum L. (PEA) in the diet were investigated on in vivo performance of two medium-growing broiler genotypes and on environmental impact. Sixty Kabir Red Plus and sixty New Red chickens were randomly allocated in 20 pens (n = 6 birds per pen). Each pen, i.e., experimental unit, received 1.18 kg dry matter (DM) including soybean (3.39%) in SOY, or pea bean (6.78%) in PEA groups. DM intake, DM refusals and bodyweight (BW) were recorded on pen basis, and average daily gain (ADG) and feed conversion rate (FCR) were calculated. Data on in vivo performance were processed by ANOVA General Linear Model followed by Student–Newman–Keuls post hoc test. Greenhouse gas (GHG) emissions were evaluated on a year basis by GLEAM-i software. The diet did not affect (p > 0.05) DM intake and DM refusals, BW, ADG and FCR. Diet x genotypes affected (p < 0.05) DM intake and DM refusals. PEA diet, cheaper than SOY diet, allowed the reduction in total GHG and CO2, and of meat emission intensity. However, an increase in total CH4 and N2O emissions was observed. The replacement of soybean with pea bean can represent a possible management strategy to reach trade-off between good farming practices and environmental protection on small-scale poultry farms of inner Mediterranean areas

    Macro Minerals and Trace Elements in Milk of Dairy Buffaloes and Cows Reared in Mediterranean Areas

    Aim of this study was to evaluate the differences in Ca, P, K, Na, Mg, Zn, Fe, Cu, Mn, Se, Mo, Co, Li, B, Ti, Rb, and Sr concentrations in milk from buffaloes and cows reared in the same farm in Mediterranean areas and fed diets including the same ingredients. Individual milk samples were obtained from 32 Mediterranean buffaloes and 29 Italian Friesian cows and samples of milk, dietary ingredients and drinking water were analyzed for the investigated chemical elements by inductively coupled plasma-mass spectrometry. Data about milk element concentrations were processed by one-way analysis of variance. Buffalo milk contains higher concentrations of Ca, P, Mg, Zn, Fe, Cu, B, Ti, and Sr, and lower concentrations of K, Na, Mo, Li, and Rb compared to cow milk, whereas milk from both species contains similar concentrations of Mn, Se, and Co. The concentrations of the investigated elements in the diet were similar for both species and the differences observed between buffalo and cow milk were not dependent on environmental factors

    A study was carried out to evaluate the effect of the total replacement of flaked soybean (Glycine max L., SOY) with raw pea (Pisum sativum L., PEA) on the carcass and meat quality traits of two medium-growing broiler strains (Kabir Rosso Plus, KB; New Red, NR). Birds were housed in 20 pens (five replications/groups, six birds each). At 83 days of age, 40 birds (2/replication) were slaughtered and the pectoral muscle (PM) was removed for analyses. Diet did not affect slaughter weight, carcass traits and meat quality. A pea diet determined a significant increase of MUFA and a decrease of PUFA, n-3 and n-6 PUFA; hence, the pea-fed group had a lower PUFA/SFA and a higher n-6/n-3 ratios compared to the soy-fed. NR chickens were heavier, with higher carcass and cut weights (p p p < 0.05) pH. Fatty acids were slightly affected by genotype.
