    Population pharmacokinetics-based recommendations for a single delayed or missed dose of nusinersen

    Nusinersen is an antisense oligonucleotide approved for the treatment of spinal muscular atrophy. The drug is given intrathecally at 12 mg, beginning with 3 loading doses at 2-week intervals, a fourth loading dose 30 days thereafter, and maintenance doses at 4-month intervals. This population pharmacokinetic model was developed to clarify how to maintain targeted nusinersen exposure after an unforeseen one-time delay or missed dose. Simulations demonstrated that the impact of a one-time delay in dosing or a missed dose on median cerebrospinal fluid exposures depended on duration of interruption and the regimen phase in which it occurred. Delays in loading doses delayed reaching the peak trough concentration by approximately the duration of the interruption. Resumption of the regimen as soon as possible resulted in achieving steady state trough concentration upon completion of the loading phase. A short delay (30-90 days) during the maintenance phase led to prolonged lower median cerebrospinal fluid concentration if all subsequent doses were shifted by the same 4-month interval. However, administration of the delayed dose, followed by the subsequent dose as originally scheduled, rapidly restored trough concentration. If a dose must be delayed, patients should return to the original dosing schedule as soon as possible

    Diving And Amphibious Behaviour In Free-Living Crocodylus porosus

    Twelve estuarine crocodiles (6-51 kg) were fitted with recording back packs in order to study the longevity, frequency and daily/tidal rhythm of natural dives by crocodiles free-living in their familiar habitat. Despite disinterest shown by captive crocodiles in removing their jackets in tank trials, all but one of the seven crocodiles recaptured 5-16 days after release had shed their jackets and recorders. Results from the 9.75 kg animal showed that it had a prolonged emergence during each daylight low tide (basking?), that it dived mainly in daylight hours (feeding?), predominantly in the upper half of the tide and that most dives were of very short duration (1-5 minutes). Even the longest dive, 30 minutes, was well within the aerobic capabilities of a crocodile this size. Although these results are from a single animal, and may turn out to be quite atypical, if attachment problems are solved, the method has clear potential for revealing much about the daily activity patterns of free-ranging crocodiles and other animals

    Clinical Meaningfulness of the Changes in Muscle Performance and Physical Function Associated With Testosterone Administration in Older Men With Mobility Limitation

    Context. Testosterone in Older Men with Mobility Limitations Trial determined the effects of testosterone on muscle performance and physical function in older men with mobility limitation. Trial's Data and Safety Monitoring Board recommended enrollment cessation due to increased frequency of adverse events in testosterone arm. The changes in muscle performance and physical function were evaluated in relation to participant's perception of change. Methods. Men aged 65 years and older, with mobility limitation, total testosterone 100-350 ng/dL, or free testosterone less than 50 pg/mL, were randomized to placebo or 10 g testosterone gel daily for 6 months. Primary outcome was leg-press strength. Secondary outcomes included chest-press strength, stair-climb, 40-m walk, muscle mass, physical activity, self-reported function, and fatigue. Proportions of participants exceeding minimally important difference in study arms were compared. Results. Of 209 randomized participants, 165 had follow-up efficacy measures. Mean (SD) age was 74 (5.4) years and short physical performance battery score 7.7 (1.4). Testosterone arm exhibited greater improvements in leg-press strength, chest-press strength and power, and loaded stair-climb than placebo. Compared with placebo, significantly greater proportion of men receiving testosterone improved their leg-press and chest-press strengths (43% vs 18%, p = .01) and stair-climbing power (28% vs 10%, p = .03) more than minimally important difference. Increases in leg-press strength and stair-climbing power were associated with changes in testosterone levels and muscle mass. Physical activity, walking speed, self-reported function, and fatigue did not change. Conclusions. Testosterone administration in older men with mobility limitation was associated with patient-important improvements in muscle strength and stair-climbing power.     Viral trans-factor independent replication of human papillomavirus genomes

    <p>Abstract</p> <p>Background</p> <p>Papillomaviruses (PVs) establish a persistent infection in the proliferating basal cells of the epithelium. The viral genome is replicated and maintained as a low-copy nuclear plasmid in basal keratinocytes. Bovine and human papillomaviruses (BPV and HPV) are known to utilize two viral proteins; E1, a DNA helicase, and E2, a transcription factor, which have been considered essential for viral DNA replication. However, growing evidence suggests that E1 and E2 are not entirely essential for stable replication of HPV.</p> <p>Results</p> <p>Here we report that multiple HPV16 mutants, lacking either or both E1 and E2 open reading frame (ORFs) and the long control region (LCR), still support extrachromosomal replication. Our data clearly indicate that HPV16 has a mode of replication, independent of viral trans-factors, E1 and E2, which is achieved by origin activity located outside of the LCR.</p

    Human Computer Interaction Meets Psychophysiology: A Critical Perspective

    Human computer interaction (HCI) groups are more and more often exploring the utility of new, lower cost electroencephalography (EEG) interfaces for assessing user engagement and experience as well as for directly controlling computers. While the potential benefits of using EEG are considerable, we argue that research is easily driven by what we term naïve neurorealism. That is, data obtained with psychophysiological devices have poor reliability and uncertain validity, making inferences on mental states difficult. This means that unless sufficient care is taken to address the inherent shortcomings, the contributions of psychophysiological human computer interaction are limited to their novelty value rather than bringing scientific advance. Here, we outline the nature and severity of the reliability and validity problems and give practical suggestions for HCI researchers and reviewers on the way forward, and which obstacles to avoid. We hope that this critical perspective helps to promote good practice in the emerging field of psychophysiology in HCI

    MSMEG_2731, an Uncharacterized Nucleic Acid Binding Protein from Mycobacterium smegmatis, Physically Interacts with RPS1

    While the M. smegmatis genome has been sequenced, only a small portion of the genes have been characterized experimentally. Here, we purify and characterize MSMEG_2731, a conserved hypothetical alanine and arginine rich M. smegmatis protein. Using ultracentrifugation, we show that MSMEG_2731 is a monomer in vitro. MSMEG_2731 exists at a steady level throughout the M. smegmatis life-cycle. Combining results from pull-down techniques and LS-MS/MS, we show that MSMEG_2731 interacts with ribosomal protein S1. The existence of this interaction was confirmed by co-immunoprecipitation. We also show that MSMEG_2731 can bind ssDNA, dsDNA and RNA in vitro. Based on the interactions of MSMEG_2731 with RPS1 and RNA, we propose that MSMEG_2731 is involved in the transcription-translation process in vivo

    Preparative isolation, fractionation and chemical characterization of dissolved organics from natural and industrially derived bitumen-influenced groundwaters from the Athabasca River watershed

    Recent analytical advances have provided evidence that groundwater affected by oil sands process-affected water (OSPW) is reaching the Athabasca River at one location. To understand and discriminate the toxicological risks posed by OSPW-influenced groundwater relative to groundwaters associated with natural oil sands deposits, these highly complex mixtures of soluble organics were subjected to toxicological characterization through effects directed analysis. A recently-developed preparative fractionation methodology was applied to bitumen-influenced groundwaters and successfully isolated dissolved organics from both industrial and natural sources into three chemically distinct fractions (F1, F2, F3), enabling multiple toxicological assessments. Analytical techniques included electrospray ionization high resolution mass spectrometry (ESI-HRMS), liquid chromatography quadrupole time-of-flight mass spectrometry (LC-QToF/MS), gas chromatography mass spectrometry (GC–MS), and synchronous fluorescence spectroscopy (SFS) methods, which did not reveal obvious differences between sources. Comparisons between fractions within each source consistently demonstrated that F3 contained compounds with greater polarity than F2, which in turn was more polar than F1. The abundance of O2 species was confined to F1, including naphthenic acids often cited for being the primary toxicants within bitumen-influenced waters. This result is consistent with earlier work on aged OSPW, as well as with other extraction methods, suggesting that additional factors other than molecular weight and the presence of acid functionalities play a prominent role in defining compound polarities and toxicities within complex bitumen-derived organic mixtures. The similarities in organic abundances, chemical speciation, aromaticity, and double bond equivalents, concomitant with inorganic mixture similarities, demonstrate the resemblances of bitumen-influenced groundwaters regardless of the source, and reinforce the need for more advanced targeted analyses for source differentiation.This work was funded under the Oil Sands Monitoring Program, and is a contribution to the Program, but does not necessarily reflect the position of the Program.
