    Investigation of Tribological Performances for Porous Structure of Diatom Frustule with FSI Method

    Tribological performances of the diatom frustule are investigated with the liquid-solid interaction (FSI) method. Take, for example, the representative Coscinodiscus sp. shell; the diatom frustule with the porous structure is achieved by the scanning electron microscope (SEM). Based on the frustule, the representative diatom frustule is modeled. Further, tribological performances of the diatom at its different geometry sizes and velocities are solved with FSI method and compared with corresponding values for the nonporous structure. The numerical result shows that the existence of the porous structure of the diatom helps to reduce friction between it and ambient water and to increase its load-carrying capacity

    Micro-Inertial-Aided High-Precision Positioning Method for Small-Diameter PIG Navigation

    Pipeline leakage or explosion has caused huge economic losses, polluted the environments and threatened the safety of civilian’s lives and assets, which even caused negative influences to the society greatly. Fortunately, pipeline inspection gauge (PIG) could accomplish the pipeline defect (corrosions, cracks, grooves, etc.) inspection effectively and meanwhile to localize these defects precisely by navigation sensors. The results are utilized for pipeline integrity management (PIM) and pipeline geographic information system construction. Generally, the urban underground pipeline presents with small-diameter and complicated-distribution properties, which are of great challenges for the pipeline defects positioning by PIG. This chapter focuses on in-depth research of the high-precision positioning method for small-diameter PIG navigation. In the beginning, the problems and system errors statement of MEMS SINS-based PIG are analyzed step by step. Then, the pipeline junction (PJ) identification method based on fast orthogonal search (FOS) is studied. After that, a PIG positioning system that comprises of micro-inertial/AGM/odometer/PJ is proposed, and also the application mechanism of extended Kalman filter and its smoothing technology on PIG navigation system is researched to improve the overall positioning precision for the small-diameter PIG. Finally, the proposed methods and research conclusions are verified by the indoor wheel robot simulation platform

    Identification of forensically significant beetles (Coleoptera: Staphylinoidae) based on COI gene in China

    _________________________________________________________________________________________ Abstract: Accurate and fast identification of insect species is an initial and crucial step of using entomological evidence in forensic cases. Beetles of Coleoptera usually take large parts of taxonomic composition of the carrion-arthropod community and have important meaning for the estimation of postmortem interval (PMI). Traditional morphological taxonomy of beetles poses a great challenge for forensic investigators without entomological background. A cytochrome oxidase subunits one (COI) sequence on mitochondrial DNA with 816-bp length was studied for molecular identification of forensic significant beetles. Six beetle species in superfamily Staphylinoidae collected from four locations in China were sampled. All species were successfully separated with high support value, which indicated this partial COI fragment was sufficient for distinguishing these beetles. Further step utilization of COI fragment in identification of forensically important beetles was also discussed.     <p><b>RNA extraction and preparation for RNA-Sequencing</b></p><p>Total RNA of each sample was isolated using TRIzol reagent (Invitrogen, Carlsbad, CA). The NanoPhotometer spectrophotometer (IMPLEN, Westlake Village, CA), the Qubit RNA Assay Kit in Qubit 2.0 Fluorometer (Life Technologies, Carlsbad, CA), and the RNA Nano 6000 Assay Kit of Bioanalyzer 2100 System (Agilent Technologies, Santa Clara, CA) ware utilized to check the purity, concentration, and integrity of RNA respectively.</p><p>In present study, eight complementary DNA (cDNA) libraries were constructed in total, i.e., four for rabbit VSMCs as control group and another four for ox-LDL-treated VSMCs. 3 µg RNA per sample was used as input material. Ribosomal RNA was removed by Epicentre Ribo-Zero rRNA Removal Kit (Epicentre, Madison, WI) firstly. Residue of rRNA-free was cleaned through ethanol precipitation. Second, sequencing libraries were generated using NEB-Next Ultra Directional RNA Library Prep Kit for Illumina (NEB, Ipswich, MA).</p><p>First-strand cDNA were synthesized through M-MuLV Reverse Transcriptase (RNaseH-) plus random hexamer primer. Then second-strand cDNA was synthesized subsequently using RNase H and DNA Polymerase I were utilized for synthesis of second-strand cDNA. The 3′ends of DNA fragments were adenylated, then NEBNext Adaptor with hair-pin loop structure were prepared for hybridization. AMPure XP System (Beckman Coulter, Beverly, MA) was used to purify the library fragments with preference of 150-200 bp in length. Finally, the library quality was assessed by Agilent Bioanalyzer 2100 System. </p><p>Clustering of samples was performed on cBot Cluster Generation System though TruSeq PE Cluster Kit v3-cBot-HS (Illumina). Then libraries were sequenced at the Novogene Bioinformatics Institute (Beijing, China) on Illumina HiSeq 4000 Platform, and paired-end reads with 150 bp were generated by Illumina HiSeq 2500 Platform.</p><p><b>Quality assessment, genome mapping, and transcriptome assembly </b></p><p>Clean reads were screen out from raw data by removing reads with adapter, poly-N or low quality. Then, calculation were performed to assess Q20, Q30, and GC contents of the clean reads. Clean reads of high-quality were used for subsequent analyses. </p><p>The rabbit (<i>Oryctolagus cuniculus</i>) genome (ftp://ftp.ensembl.org/pub/release-88/embl/oryctolagus_cuniculus/) was used as reference genome for reads mapping through TopHat v2.0.9. Cufflinks v2.1.1 was used for mapped reads assembly of each sample. </p><p><b>LncRNA identification </b></p><p>All successfully assembled reads were combined by Cuffcompare software. Then transcripts shorter than 200 bp in length or less than two exons was removed. Reads coverage was calculated by Cufflinks v2.1.1 and transcripts less than three reads coverage were abandoned. Comparison between transcripts and reference rabbit lncRNAs was performed by Cuffcompare software. RNA transcripts like tRNA, rRNA, snoRNA, snRNA, pre-miRNA, and pseudogenes were also detected and discarded. Then, transcripts of mRNA were gained from blasting with annotated rabbit mRNA. Software CNCI, CPC, PFAM, and phyloCSF were utilized to screen out lncRNA by assessing the coding potential of transcripts. The final results contained lncRNAs which classified into three subtypes as lincRNA, intronic lncRNA, and antisense lncRNA through class_code (http://coLetrapnelllabio/cufflinks/cuffcompare/index.Html#transfrag-classcodes).</p><p><b>Expression analysis</b></p><p> </p><p>The expression levels of lncRNAs and mRNAs in each sample were evaluated using Cuffdiff (v2.1.1). Fragments per kilo-base of exon per million fragments (FPKM) mapped of the transcripts was calculated using Cuffdiff (v2.1.1). Then results were used for evaluating the expression levels of lncRNAs and mRNAs in each sample. Expression of transcripts with <i>P</i> value <0.05 was defined as significant difference between control group and ox-LDL treated group.</p

    Influence of Annealing and UV Irradiation on Hydrophilicity of Ag- Nanostructured Thin Films

    Ag-TiO2 nanostructured thin films with silver content of 5 vol% have been deposited on silicon, glass, and quartz substrates by RF magnetron sputtering and annealed in ambient air at 900°C for 15, 30, 60, 90, and 120 min. Their crystal structure, surface morphology, and hydrophilicity have been characterized by X-ray diffractometer, atomic force microscope, and water contact angle apparatus, respectively. The influence of annealing time and UV irradiation time on hydrophilic property of Ag-TiO2 thin films have been studied in detail. It is shown that annealing time influences crystal structure of Ag-TiO2 thin films. The unannealed film is amorphous and shows poor hydrophilicity. With the increase of annealing time from 15 to 120 min, the grain-size slowly increases and tends to uniformity. A suitable annealing time can significantly enhance the hydrophilic behavior of Ag-TiO2 films. Water contact angle decreases with the increase of irradiation time.
