    Theory of ac electrokinetic behavior of spheroidal cell suspensions with an intrinsic dispersion

    The dielectric dispersion, dielectrophoretic (DEP) and electrorotational (ER) spectra of spheroidal biological cell suspensions with an intrinsic dispersion in the constituent dielectric constants are investigated. By means of the spectral representation method, we express analytically the characteristic frequencies and dispersion strengths both for the effective dielectric constant and the Clausius-Mossotti factor (CMF). We identify four and six characteristic frequencies for the effective dielectric spectra and CMF respectively, all of them being dependent on the depolarization factor (or the cell shape). The analytical results allow us to examine the effects of the cell shape, the dispersion strength and the intrinsic frequency on the dielectric dispersion, DEP and ER spectra. Furthermore, we include the local-field effects due to the mutual interactions between cells in a dense suspension, and study the dependence of co-field or anti-field dispersion peaks on the volume fractions.Comment: accepted by Phys.     Hygienic practices and diarrheal illness among persons living in at-risk settings in Kabul, Afghanistan: a cross-sectional study

    Abstract Background Sustained civil and military conflict, resulting in large numbers of internally displaced persons (IDP), in combination with rapid urbanization has strained public health and sanitation within cities in Afghanistan. In order to examine the association between preventive sanitary behaviors and diarrhea within two high risk settings located within Kabul, Afghanistan, this study aimed to evaluate the prevalence of hygienic practices and diarrheal illness in an IDP camp and an urban slum. Methods In this cross sectional study, a convenience sample of residents of an IDP camp and an urban slum in Kabul, Afghanistan, was used. Participants were asked to describe their hygienic practices and interviewers independently documented household sanitation. The knowledge and attitudes about and practice of hygienic activities to prevent diarrhea were compared between the two settings. Results Two hundred participants, 100 from each setting, were enrolled. Knowledge, attitudes, and practices regarding hygienic activities to prevent diarrhea were greater among the slum dwellers than the IDP. Fewer than half of participants washed their hands with soap before eating or after eating: 31 % of slum dwellers washed before eating compared to 11 % of IDPs (P = 0.0050), and 25 % of slum dwellers washed after defecating compared to 4 % of IDPs (P = 0.0020). The IDPs were more likely to share a latrine (P = 0.0144) and less likely to disinfect their latrine than slum dwellers. Diarrhea in the household within the past 3 months was more common in the IDP camp (54 %) than the slum (20 %) (P = 0.0020). Conclusions Even though certain sanitary and hygienic practices were more common among slum dwellers than IDPs, the lack of hygienic activities in both setting indicates that interventions to change behavior, like increasing the availability of soap and encouraging hand washing, are needed.     Gauging the spectator equations

    We show how to derive relativistic, unitary, gauge invariant, and charge conserving three-dimensional scattering equations for a system of hadrons interacting with an electromagnetic field. In the method proposed, the spectator equations describing the strong interactions of the hadrons are gauged using our recently introduced gauging of equations method. A key ingredient in our model is the on-mass-shell particle propagator. We discuss how to gauge this on-mass-shell propagator so that both the Ward-Takahashi and Ward identities are satisfied. We then demonstrate our gauging procedure by deriving the gauge-invariant three-dimensional expression for the deuteron photodisintegration amplitude within the spectator approach.Comment: 17 pages, REVTeX, epsf, 1 Postscript figur

    Switchable genome editing via genetic code expansion

    Multiple applications of genome editing by CRISPR-Cas9 necessitate stringent regulation and Cas9 variants have accordingly been generated whose activity responds to small ligands, temperature or light. However, these approaches are often impracticable, for example in clinical therapeutic genome editing in situ or gene drives in which environmentally-compatible control is paramount. With this in mind, we have developed heritable Cas9-mediated mammalian genome editing that is acutely controlled by the cheap lysine derivative, Lys(Boc) (BOC). Genetic code expansion permitted non-physiological BOC incorporation such that Cas9 (Cas9BOC) was expressed in a full-length, active form in cultured somatic cells only after BOC exposure. Stringently BOC-dependent, heritable editing of transgenic and native genomic loci occurred when Cas9BOC was expressed at the onset of mouse embryonic development from cRNA or Cas9BOC transgenic females. The tightly controlled Cas9 editing system reported here promises to have broad applications and is a first step towards purposed, spatiotemporal gene drive regulation over large geographical ranges

    Asymmetric parental genome engineering by Cas9 during mouse meiotic exit

    Mammalian genomes can be edited by injecting pronuclear embryos with Cas9 cRNA and guide RNA (gRNA) but it is unknown whether editing can also occur during the onset of embryonic development, prior to pronuclear embryogenesis. We here report Cas9-mediated editing during sperm-induced meiotic exit and the initiation of development. Injection of unfertilized, mouse metaphase II (mII) oocytes with Cas9 cRNA, gRNA and sperm enabled efficient editing of transgenic and native alleles. Pre-loading oocytes with Cas9 increased sensitivity to gRNA ~100-fold. Paternal allelic editing occurred as an early event: single embryo genome analysis revealed editing within 3 h of sperm injection, coinciding with sperm chromatin decondensation during the gamete-to-embryo transition but prior to pronucleus formation. Maternal alleles underwent editing after the first round of DNA replication, resulting in mosaicism. Asymmetric editing of maternal and paternal alleles suggests a novel strategy for discriminatory targeting of parental genomes

    Development of Thick-foil and Fine-pitch GEMs with a Laser Etching Technique

    We have produced thick-foil and fine-pitch gas electron multipliers (GEMs) using a laser etching technique. To improve production yield we have employed a new material, Liquid Crystal Polymer, instead of polyimide as an insulator layer. The effective gain of the thick-foil GEM with a hole pitch of 140 um, a hole diameter of 70 um, and a thickness of 100 um reached a value of 10^4 at an applied voltage of 720 V. The measured effective gain of the thick-foil and fine-pitch GEM (80 um pitch, 40 um diameter, and 100 um thick) was similar to that of the thick-foil GEM. The gain stability was measured for the thick-foil and fine-pitch GEM, showing no significant increase or decrease as a function of elapsed time from applying the high voltage. The gain stability over 3 h of operation was about 0.5%. Gain mapping across the GEM showed a good uniformity with a standard deviation of about 4%. The distribution of hole diameters across the GEM was homogeneous with a standard deviation of about 3%. There was no clear correlation between the gain and hole diameter maps.Comment: 21 pages, 9 figure

    Development of Resistive Electrode Gas Electron Multiplier (RE-GEM)

    We successfully produced Resistive-Electrode Gas Electron Multiplier (RE-GEM) which has resistive electrodes instead of the metal ones which are employed for the standard GEM foils. RE-GEM has a resistive electrode of 25 micron-thick and an insulator layer of 100 micron-thick. The hole structure of RE-GEM is a single conical with the wider and narrower hole diameters of 80 micron and 60 micron, respectively. A hole pitch of RE-GEM is 140 micron. We obtained the maximum gain of about 600 and the typical energy resolution of about 20% (FWHM) at an applied voltage between the resistive electrodes of 620 V, using a collimated 8 keV X-rays from a generator in a gas mixture of 70% Ar and 30% CO2 by volume at the atmospheric pressure. We measured the effective gain as a function of the electric field of the drift region and obtained the maximum gain at an drift field of 0.5 kV/cm

    Bethe-Salpeter equation and a nonperturbative quark-gluon vertex

    A Ward-Takahashi identity preserving Bethe-Salpeter kernel can always be calculated explicitly from a dressed-quark-gluon vertex whose diagrammatic content is enumerable. We illustrate that fact using a vertex obtained via the complete resummation of dressed-gluon ladders. While this vertex is planar, the vertex-consistent kernel is nonplanar and that is true for any dressed vertex. In an exemplifying model the rainbow-ladder truncation of the gap and Bethe-Salpeter equations yields many results; e.g., pi- and rho-meson masses, that are changed little by including higher-order corrections. Repulsion generated by nonplanar diagrams in the vertex-consistent Bethe-Salpeter kernel for quark-quark scattering is sufficient to guarantee that diquark bound states do not exist.Comment: 16 pages, 12 figures, REVTEX
