    Ponderomotive scattering of an electron-bunch before injection into a laser wakefield

    For the purpose of laser wakefield acceleration, it turned out that also the injection of electron bunches longer than a plasma wavelength can generate accelerated femtosecond bunches with relatively low energy spread. This is of high interest because such injecting bunches can be provided, e.g., by state-of-the-art photo cathode RF guns. Here we point out that when an e-bunch is injected in the wakefield it is important to take into account the ponderomotive scattering of the injecting bunch by the laser pulse in the vacuum region located in front of the plasma. At low energies of the injected bunch this scattering results in a significant drop of the collection efficiency. Larger collection efficiency can by reached with lower intensity laser pulses and relatively high injection energies. We also estimate the minimum trapping energy for the injected electrons and the length of the trapped bunch.Comment: 12 pages, 5 figures, submitted to Phys. Rev.

    Electromagnetic radiation from a laser wakefield accelerator

    Coherent and incoherent electromagnetic radiation emitted from a laser wakefield accelerator is calculated based on Lienard-Wiechert potentials. It is found that at wavelengths longer than the bunch length, the radiation is coherent. The coherent radiation, which typically lies in the infrared range, shows features that reveal details of the acceleration process and properties of the electron bunch, such as its duration, charge, energy, and offset with respect to the wakefield axis. The incoherent range of the spectrum, which extends to the X-ray frequency range, consists of rather broad peaks, which are caused by the acceleration.Comment: 14 pages, incl. 6 figures, submitted for publicatio

    Dynamics of Femtosecond Electron Bunches

    In the laser wakefield accelerator (LWFA) a short intense laser pulse, with a duration of the order of a plasma wave period, excites an unusually strong plasma wake wave (laser wakefield). Recent experiments on laser wakefield acceleration [Nature (London) 431, p.535, p.538, p.541 (2004)] demonstrated generation of ultra-short (with a duration of a few femtoseconds) relativistic electron bunches with relatively low energy spread of the order of a few percent. We have studied the dynamics of such bunches in vacuum and in laser wakefield. The results show strong bunch dynamics already on a few millimeters propagation distance in both cases. In vacuum, the bunch radius and emittance quickly grow. The latter worsens the focusability of the bunch. We found that when a femtosecond bunch is accelerated in a channel-guided laser wakefield, for realistic bunch lengths, the bunch length is approximately conserved. However, the spread in betatron frequencies leads to fast betatron phase mixing in the bunch envelope for on-axis injection. When bunch is injected in a laser wakefield off-axis, the bunch decoherence results in considerable increase in the normalized bunch emittance, and, in some cases, in increase in the energy spread, after acceleration.     Generation of stable ultra-relativistic attosecond electron bunches via the laser wakefield acceleration mechanism

    In recent experiments ultra-relativistic femtosecond electron bunches were generated by a Laser Wakefield Accelerator (LWFA) in different regimes. Here we predict that even attosecond bunches can be generated by an LWFA due to the fast betatron phase mixing within a femtosecond electron bunch. The attosecond bunches are stable outside the LWFA and can propagate in vacuum many tens of centimeter without significant change in their duration. Our calculations show that evidence for the formation of attosecond bunches can be found in the spectrum of coherent betatron radiation from LWFA's

    Plasma Citrate Levels Are Associated with an Increased Risk of Cardiovascular Mortality in Patients with Type 2 Diabetes (Zodiac-64)

    Circulating citrate may represent a proxy of mitochondrial dysfunction which plays a role in the development of vascular complications in type 2 diabetes (T2D). Here, we determined the associations between plasma citrate levels and cardiovascular (CV) mortality in T2D patients. In this prospective cohort study, 601 patients were included who participated in the Zwolle Outpatient Diabetes project Integrating Available Care (ZODIAC). Plasma citrate levels were measured by nuclear magnetic resonance spectroscopy. Cox proportional hazards regression models were used to evaluate the associations between plasma citrate and the risk of CV mortality. Over a median follow-up of 11.4 years, 119 (19.8%) of the 601 patients died from a CV cause. In multivariable Cox proportional hazards regression models, adjusting for conventional risk factors, plasma citrate was associated with an increased risk of CV mortality (the hazard ratio (HR) per 1-SD increment was 1.19 (95%CI: 1.00–1.40), p = 0.048). This association was prominent in males (n = 49 with CV mortality) (HR 1.52 (95%CI: 1.14–2.03), p = 0.005), but not in females (n = 70 with CV mortality) (HR 1.11 (95%CI: 0.90–1.37), p = 0.319) (age-adjusted Pinteraction = 0.044). In conclusion, higher plasma citrate levels are associated with an increased risk of CV mortality in patients with established T2D. Future studies are warranted to unravel the potential role of citrate-related pathways in the pathogenesis of T2D-related vascular complications

    The Systemic Redox Status Is Maintained in Non-Smoking Type 2 Diabetic Subjects Without Cardiovascular Disease:Association with Elevated Triglycerides and Large VLDL

    Decreased circulating levels of free thiols (R-SH, sulfhydryl groups) reflect enhanced oxidative stress, which plays an important role in the pathogenesis of cardiometabolic diseases. Since hyperglycemia causes oxidative stress, we questioned whether plasma free thiols are altered in patients with type 2 diabetes mellitus (T2DM) without cardiovascular disease or renal function impairment. We also determined their relationship with elevated triglycerides and very low density lipoproteins (VLDL), a central feature of diabetic dyslipidemia. Fasting plasma free thiols (colorimetric method), lipoproteins, VLDL (nuclear magnetic resonance spectrometry), free fatty acids (FFA), phospholipid transfer protein (PLTP) activity and adiponectin were measured in 79 adult non-smoking T2DM subjects (HbA1c 51 ± 8 mmol/mol, no use of insulin or lipid lowering drugs), and in 89 non-smoking subjects without T2DM. Plasma free thiols were univariately correlated with glucose (r = 0.196, p < 0.05), but were not decreased in T2DM subjects versus non-diabetic subjects (p = 0.31). Free thiols were higher in subjects with (663 ± 84 µmol/L) versus subjects without elevated triglycerides (619 ± 91 µmol/L; p = 0.002). Age- and sex-adjusted multivariable linear regression analysis demonstrated that plasma triglycerides were positively and independently associated with free thiols (β = 0.215, p = 0.004), FFA (β = 0.168, p = 0.029) and PLTP activity (β = 0.228, p = 0.002), inversely with adiponectin (β = -0.308, p < 0.001) but not with glucose (β = 0.052, p = 0.51). Notably, the positive association of free thiols with (elevated) triglycerides appeared to be particularly evident in men. Additionally, large VLDL were independently associated with free thiols (β = 0.188, p = 0.029). In conclusion, circulating free thiols are not decreased in this cohort of non-smoking and generally well-controlled T2DM subjects. Paradoxically, higher triglycerides and more large VLDL particles are likely associated with higher plasma levels of thiols, reflecting lower systemic oxidative stress

    Prolonged Organ Extraction Time Negatively Impacts Kidney Transplantation Outcome

    Main Problem: Following cold aortic flush in a deceased organ donation procedure, kidneys never reach the intended 0–4°C and stay ischemic at around 20°C in the donor’s body until actual surgical retrieval. Therefore, organ extraction time could have a detrimental influence on kidney transplant outcome. Materials and Methods: We analyzed the association between extraction time and kidney transplant outcome in multicenter data of 5,426 transplant procedures from the Dutch Organ Transplantation Registry (NOTR) and 15,849 transplant procedures from the United Network for Organ Sharing (UNOS). Results: Extraction time was grouped per 10-min increment. In the NOTR database, extraction time was independently associated with graft loss [HR 1.027 (1.004–1.050); p = 0.022] and with DGF [OR 1.043 (1.021–1.066); p 80 min was associated with a 27.4% higher hazard rate of graft failure [HR 1.274 (1.080–1.502); p = 0.004] and such kidneys had 43.8% higher odds of developing DGF [OR 1.438, (1.236–1.673); p 30 min was associated with 14.5% higher odds of developing DGF [OR 1.145 (1.063–1.233); p < 0.005]. Discussion: Prolonged kidney extraction time negatively influenced graft survival in Dutch donors and increased DGF risk in all deceased donor recipients

    Maternal Anxiety, Infant Stress, and the Role of Live-Performed Music Therapy during NICU Stay in The Netherlands

    Having an infant in the neonatal intensive care unit (NICU) elicits maternal anxiety, which may hamper parent−child bonding. We performed a prospective cohort study to describe anxiety in mothers of infants born before 30 weeks of gestation during NICU stay in The Netherlands, and investigated the influence of infant stress and gestational age. Second, we performed a randomized-controlled live-performed music therapy trial (LPMT trial) to investigate whether music therapy applied to the infant alleviated maternal anxiety. The relation between infant stress, gestational age, and maternal anxiety was measured in 45 mother−infant dyads, using the Neonatal Infant Stressor Scale and the State-Trait Anxiety Inventory (STAI). The effect of LPMT on anxiety was assessed in 21 mothers whose infants were assigned to either LPMT (n = 12) or waitlist (n = 9). Mothers completed the STAI before and after this period. Maternal anxiety decreased over time in all mothers, and was strongly related with infant stress (r = 0.706, p < 0.001), but not with gestational age. Anxiety scores decreased by 12% after LMPT, and increased by 1% after a waitlist period (p = 0.30). Our results indicate that LPMT in the weeks after birth may accelerate the reduction of maternal anxiety. Further research should focus on the effects on mother−child bonding

    Mild Coronavirus Disease 2019 (COVID-19) Is Marked by Systemic Oxidative Stress:A Pilot Study

    Oxidative stress has been implicated to play a critical role in the pathophysiology of coronavirus disease 2019 (COVID-19) and may therefore be considered as a relevant therapeutic target. Serum free thiols (R-SH, sulfhydryl groups) comprise a robust marker of systemic oxidative stress, since they are readily oxidized by reactive oxygen species (ROS). In this study, serum free thiol concentrations were measured in hospitalized and non-hospitalized patients with COVID-19 and healthy controls and their associations with relevant clinical parameters were examined. Serum free thiol concentrations were measured colorimetrically (Ellman’s method) in 29 non-hospitalized COVID-19 subjects and 30 age-, sex-, and body-mass index (BMI)-matched healthy controls and analyzed for associations with clinical and biochemical disease parameters. Additional free thiol measurements were performed on seven serum samples from COVID-19 subjects who required hospitalization to examine their correlation with disease severity. Non-hospitalized subjects with COVID-19 had significantly lower concentrations of serum free thiols compared to healthy controls (p = 0.014), indicating oxidative stress. Serum free thiols were positively associated with albumin (St. β = 0.710, p < 0.001) and inversely associated with CRP (St. β = −0.434, p = 0.027), and showed significant discriminative ability to differentiate subjects with COVID-19 from healthy controls (AUC = 0.69, p = 0.011), which was slightly higher than the discriminative performance of CRP concentrations regarding COVID-19 diagnosis (AUC = 0.66, p = 0.042). This study concludes that systemic oxidative stress is increased in patients with COVID-19 compared with healthy controls. This opens an avenue of treatment options since free thiols are amenable to therapeutic modulation
