    Decreased expression of heat shock protein HSP90α after exposure to doxorubicin in breast cancer cell lines (MCF-7 and MDA-MB-231)

    Background and purpose: Incidence of breast cancer is increasing day by day. Scientists are interested in the effects of inhibition of breast cancer cell on treatment of this cancer. The aim of this study was to determine IC50 of doxorubicin in 24 hours on cell lines MCF-7 and MDA-MB-231 and the expression of heat shock protein HSP90α as a factor in the cell before and after 24 hours exposure to doxorubicin in both breast cancer cell lines. Materials and methods: A descriptive interventional study was done in two cell lines MCF-7 and MDA-MB-231` after 24 hours exposure to doxorubicin. Sensitivity of cells to doxorubicin was determined using MTT Assay in excel software. HSP90α heat shock protein expression were qualitatively compared in both cell lines before and after exposure to doxorubicin using immunofluorescent techniques (Immunocytochemistry). Results: MTT Assay showed that IC50 value in MDA-MB-231 and MCF7 cells after 24 hours exposure to doxorubicin (the dose that kills 50% of cells) were 14.521 and 16.3315µM, respectively. Immunocytochemistry revealed that HSP90α protein expression in both cell lines after exposure to doxorubicin decreased compared to the control group. Conclusion: Cell density in cell lines (ER-) MDA-MB-231 and line MCF-7 (ER+) after exposure to doxorubicin and increasing the dose of medication, decreased indicating a dose dependent effect. Also, apoptosis occurred in both cell lines and expression of HSP90α decreased but MDA-MB-231 cells were found to be more sensitive. © 2017, Mazandaran University of Medical Sciences.     Differential expression of HSP90β in MDA-MB-231 and MCF-7 cell lines after treatment with doxorubicin

    Breast cancer is a complex, heterogeneous disease and one of the most common malignancies in women worldwide. The efficacy of chemotherapy as an important breast cancer treatment option has been severely limited because of the inherent or acquired resistance of cancer cells. The molecular chaperone heat shock protein 90 (HSP90) upregulated in response to cellular stress is required for functions such as conformational maturation, activation and stability in more than 200 client proteins, mostly of the signaling type. In this study, the expression of HSP90 isoforms including HSP90α and HSP90β in breast cancer cell lines before and after treatment with doxorubicin (DOX) was assessed. Material and Methods: The cell cytotoxicity of DOX in MDA-MB-231 and MCF-7 cell lines was determined using the MTT assay. Immunofluorescence and western blotting techniques were used to determine the expression of HSP90β in the cell lines before and after DOX treatment. Immunofluorescence was also conducted to ascertain the expression of HSP90α. Results: The MTT assay results showed that the MDAMB- 231 cells (IC50=14.521 μM) were more sensitive than the MCF-7 cells (IC50=16.3315 μM) to DOX. The immunofluorescence results indicated that the expression of HSP90α in both cell lines decreased after exposure to DOX. The western blot and immunofluorescence analyses showed that HSP90β expression decreased in the MCF-7 cells but increased in the MDAMB- 231 cells after DOX treatment. Conclusion: The obtained results suggested that HSP90α and HSP90β expression levels were reduced in the MCF-7 cells after exposure to DOX. In the MDA-MB-231 cells, HSP90α expression was reduced while HSP90β was found to be overexpressed following DOX treatment. © 2019 Korean Pharmacopuncture Institute

    Outcomes of chronic total occlusion percutaneous coronary intervention from the RAIAN (RAjaie - Iran) registry

    Objective: While most of the evidence in CTO interventions emerge from Western and Japanese studies, few data have been published up today from the Middle East. Objective of this study was to evaluate technical success rates and clinical outcomes of an Iranian population undergoing CTO PCI in a tertiary referral hospital. Moreover, we sought to evaluate the efficacy of our CTO teaching program. Methods: This is a retrospective single-center cohort study including 790 patients who underwent CTO PCI performed by operators with different volumes of CTOs PCI performed per year. According to PCI result, all patients have been divided into successful (n = 555, 70.3 %) and unsuccessful (n = 235, 29.7 %) groups. Study endpoints were Major Adverse Cardiovascular Events and Health Status Improvement evaluated using the Seattle Angina Questionnaire at one year. Results: A global success rate of 70 % for antegrade and 80 % for retrograde approach was shown despite the lack of some CTO-dedicated devices. During the enrollment period, the success rate increased significantly among operators with a lower number of CTO procedures per year. One-year MACE rate was similar in both successful and unsuccessful groups (13.5 % in successful and 10.6 % in unsuccessful group, p = 0.173). One year patients' health status improved significantly only in successful group. Conclusions: No significant differences of in-hospital and one-year MACE were found between the successful and unsuccessful groups. Angina symptoms and quality of life significantly improved after successful CTO PCI. The RAIAN registry confirmed the importance of operator expertise for CTO PCI success

    Comparative study of the functional properties of three legume seed isolates: adzuki, pea and soy bean

    The aim of this work was to compare functional properties including solubility, emulsifying and foaming properties of native and thermally treated adzuki, soy and pea protein isolates prepared under the same conditions. These functional properties were tested at four pH values: pH 3.0, pH 5.0, pH 7.0 and pH 8.0. The lowest solubility at all pH values were obtained for isolate of adzuki whereas isolates of soybean had the highest values at almost all pHs. Thermal treatment reduced solubility of soy and pea isolates at all pH values, whereas solubility of adzuki isolate was unchanged, except at pH 8. Native isolate of adzuki had the best emulsifying properties at pH 7.0 whereas at the other pH values some of native pea and soybean protein isolates were superior. After thermal treatment, depending on tested pH and selected variety all of three species could be a good emulsifier. Native soy protein isolates formed the most stable foams at all pHs. Thermal treatment significantly improved foaming properties of adzuki isolate, whereas reduced foaming capacity of soy and pea isolates, but could improve foam stability of these isolates at specific pH. Appropriate selection of legume seed as well as variety could have great importance in achievement of desirable functional properties of final products. All three tested species could find specific application in wide range of food products

    Comparison of water sorption and solubility of Acropars and Meliodent heat cure acrylic resins

    Background and Aim: Water sorption and solubility are important properties of acrylic resins. Denture base acrylic resins have low solubility. This solubility results from the leaching out of unreacted monomer and water soluble additives into the oral fluids. The solubility of denture bases can cause oral soft tissue reactions. In addition, water absorbed into this material acts as a plasticizer and decreases the mechanical properties such as hardness, transverse strength, fatigue limit and also can change the color and dimensional stability. The aim of this study was to compare the water sorption and solubility of Acropars and Meliodent heat cure acrylic resins. Materials and Methods: This experimental study was performed on the basis of ADA specification No.12 and ISO No.1567 and standards NO: 2571 of Institute of Standards & Industrial Research of Iran. Six disc form samples of each acrylic resin were prepared, with the dimension of 50×0.5 mm. After desiccating, the samples were kept in an oven for 24 hours and weighed. Then they were immersed in water, kept in oven for 7 days and weighed again. After this phase, the samples were carried to a dessicator, for 24 hours and kept in an oven for drying and were weighed for the third time. Data were analyzed with Mann Whitney and one sample t-test. P<0.05 was considered as the limit of significance. Results: Water sorption mean values were 30.5±0.1 µg/mm3 or 0.76±0.01 mg/cm2 for Meliodent samples and 30.7±0.87 µg/mm3 or 0.77±0.009 mg/cm2 for Acropars samples. No significant difference was observed in water sorption of these two materials (P=0.9). Meliodent acrylic resin showed lower solubility (1.7±0.097 µg/mm3 or 0.042±0.001 mg/cm2) than Acropars acrylic resin (2.5±0.13 µg/mm3 or 0.062±0.001 mg/cm2) (P=0.002). Conclusion: Acropars heat cure acrylic resin matched well with the requirements of the international standards for water sorption, but its solubility was not favorable. This problem is probably the result of high concentration of residual monomer, the amount and type of cross linking agents and plasticizers as well as the amount of water soluble additives in this material

    The effects of vasectomy on epididymal morphology and sperm parameters in adult male Balb/c mice

    Vasectomy is a common contraceptive procedure in men. The present study is aimed to explore the impact of vasectomy on epididymal morphology and sperm parameters in adult male Balb/c mice. Twenty adult (age: 8 weeks) male Balb/c mice, weighting 20-30 g were used in the experiments. They were divided into 2 groups (vasectomy and sham). The operations were performed under sodium pentobarbital (40 mg/kg body weight, IP) anesthesia via a lower mid-abdominal incision. The left epididymis caput was fixed for histological studies and the right one was used for sperm count and motility. Progressive fast and slow sperm motility were significantly decreased in the vasectomized compared to the sham operation group (P<0.05) and the number of immotile sperm in the vasectomized group was increased in comparison to control group. Sperm granuloma was seen in 60 of epididymis after vasectomy. Also, Histological study showed an increase in tubular lumen diameter, interstitial space and infiltration of immune cells in interstitial tissue in vasectomized group. Vasectomy increases histopathological changes in epididymis and decreases the motility of sperm developing a reduction in fertility rates
