    Unusual localization of pennella sp. In swordfish (xiphias gladius) hearts

    The genus Pennella comprises hematophagous parasites of marine aquatic species, including cephalopods, marine mammals, and pelagic fish. Nine species have been officially included in the genus Pennella plus another six species inquirendae. They are most often found in the host’s musculature, without penetrating internal organs. For the present study, 83 hearts from swordfish (Xiphias gladius) caught in the Mediterranean Sea were sampled and immediately fixed in formalin for histopathological analysis. In total, 10 (12.05%) hearts were found to be parasitized by copepods of the genus Pennella. Macroscopically, there was mild-to-severe fibrinous pericarditis with atrial wall thickening and multiple parasitic nodules. Histologically, the parasitic nodules were surrounded by an inflammatory-necrotizing reaction. Parasitic infestation by Pennella spp. is common in pelagic fish and in swordfish, in particular. Here, however, we report atypical cardiac localization. A future area of focus is the evaluation of cardiac Pennella spp. infestation by histopathology and genetic identification of the parasites

    Underdiagnosis of osteoporotic vertebral fractures in patients with fragility fractures: retrospective analysis of over 300 patients

    Osteoporosis (OP) is a silent disease unless a fracture occurs; it is a major health problem, mainly due to fragility fractures, that occur at vertebral and peripheral sites. Vertebral fractures (VF) are probably the most common fragility fractures, but they go often unrecognized. The main clinical symptoms of VF are acute and chronic back pain, spinal deformity, reduced mobility and impaired quality of life. They are frequently associated with other fragility fractures. We examined 478 patients at our outpatient clinic, who were referred for fragility fracture occurrence. The most common fragility fractures was hip fractures. However, after execution of spine X-rays in patients who had sustained hip fracture, we found that a large proportion of them had VF, which had not been reported in their medical history

    Deep Energy Retrofit of Residential Buildings in the Mediterranean Area: The MedZEB Approach

    Publicado en : Green Energy and TechnologyMediterranean deep retrofit markets are characterized by common barriers and bottlenecks, which barely have been identified as shared challenges, and this has led to a lack of dedicated solutions and to a substantial delay in achieving the 2020 EU policy targets. This situation is addressed by the H2020 HAPPEN project by proposing a new MedZEB approach characterized by the following features: Holistic, i.e., aimed at integrating the most relevant aspects of the retrofitting supply chain;Transparent, i.e., aimed at putting on the market novel tools for enhancing investors’ trust;Adaptive, i.e., aimed at enhancing “added values” of the retrofitting such as flexibility, well-being, etc. Having arrived at its midpoint, HAPPEN has produced an advanced version of its main outputs, among which are: the HAPPEN cost-optimal technical solutions, developed according to a step-by-step logic;the HAPPEN financial solution, fully integrated with the step-by-step logic, and aimed at funding the retrofitting process by relying on the energy savings achieved;the MedZEB protocol conceived as a guarantee scheme for the achievement of retrofit targets;the HAPPEN platform, an assisted digital marketplace aimed at matching demand and offer according to a one-stop-shop logic, at defragmenting the retrofit value chain, and at supporting actors with dedicated tools. These outputs have been developed also thanks to an extensive living laboratory and pilot-building program, carried out within ten pilot sites across seven EU Med countries; this has made it possible for a large engagement of potential users, which resulted in the ideation of the HAPPEN program, an overall framework aimed at integrating project outputs into an exploitable renovation procedure powered by the HAPPEN platform. This paper describes the first simulation of such a procedure in its entirety based on a real case study. After characterizing the building according to the HAPPEN reference buildings and climates lists, a step-by-step cost-optimal package of solutions was calculated, followed by the application of the HAPPEN financial solution, and by the draft issue of the MedZEB protocol. Results provide the first evidence of the effectiveness of the MedZEB approach in potentially unlocking the deep retrofit market in the Med area, with special attention to the possibility of funding the interventions by relying on the economies generated by the energy savings achieved.     Enalapril reduces proliferation and hyaluronic acid release in orbital fibroblasts

    BACKGROUND: Orbital fibroblast proliferation and hyaluronic acid (HA) release are responsible for some of the clinical features of Graves' ophthalmopathy (GO). Thus, inhibition of these processes may be a possible therapeutic approach to this syndrome. Enalapril, a widely used antihypertensive drug, was found to have some inhibitory actions on fibroblast proliferation in cheloid scars in vivo, based on which we investigated its effects in primary cultures of orbital fibroblasts from GO patients and control subjects. METHODS: Primary cultures of GO and control fibroblasts were treated with enalapril or with a control compound (lisinopril). Cell proliferation assays, lactate dehydrogenase release assays (as a measure of cell necrosis), apoptosis assays, and measurement of HA in the cell media were performed. RESULTS: Enalapril significantly reduced cell proliferation in both GO and control fibroblasts. Because enalapril did not affect cell necrosis and apoptosis, we concluded that its effects on proliferation reflected an inhibition of cell growth and/or a delay in cell cycle. Enalapril significantly reduced HA concentrations in the media from both GO and control fibroblasts. CONCLUSIONS: Enalapril has antiproliferative and HA suppressing actions in both GO and control fibroblasts. Clinical studies are needed to investigate whether enalapril has any effects in vivo in patients with GO

    Novel associations for hypothyroidism include known autoimmune risk loci

    Hypothyroidism is the most common thyroid disorder, affecting about 5% of the general population. Here we present the first large genome-wide association study of hypothyroidism, in 2,564 cases and 24,448 controls from the customer base of 23andMe, Inc., a personal genetics company. We identify four genome-wide significant associations, two of which are well known to be involved with a large spectrum of autoimmune diseases: rs6679677 near _PTPN22_ and rs3184504 in _SH2B3_ (p-values 3.5e-13 and 3.0e-11, respectively). We also report associations with rs4915077 near _VAV3_ (p-value 8.3e-11), another gene involved in immune function, and rs965513 near _FOXE1_ (p-value 3.1e-14). Of these, the association with _PTPN22_ confirms a recent small candidate gene study, and _FOXE1_ was previously known to be associated with thyroid-stimulating hormone (TSH) levels. Although _SH2B3_ has been previously linked with a number of autoimmune diseases, this is the first report of its association with thyroid disease. The _VAV3_ association is novel. These results suggest heterogeneity in the genetic etiology of hypothyroidism, implicating genes involved in both autoimmune disorders and thyroid function. Using a genetic risk profile score based on the top association from each of the four genome-wide significant regions in our study, the relative risk between the highest and lowest deciles of genetic risk is 2.1
