Nontargeted Lipidomic Characterization of Porcine Organs Using Hydrophilic Interaction Liquid Chromatography and Off-Line Two-Dimensional Liquid Chromatography-Electrospray Ionization Mass Spectrometry
Abstract Lipids form a significant part of animal organs and they are responsible for important biological functions, such as semi-permeability and fluidity of membranes, signaling activity, anti-inflammatory processes, etc. We have performed a comprehensive nontargeted lipidomic characterization of porcine brain, heart, kidney, liver, lung, spinal cord, spleen, and stomach using hydrophilic interaction liquid chromatography (HILIC) coupled to electrospray ionization mass spectrometry (ESI/MS) to describe the representation of individual lipid classes in these organs. Detailed information on identified lipid species inside classes are obtained based on relative abundances of deprotonated molecules [M-H] -in the negative-ion ESI mass spectra, which provides important knowledge on phosphatidylethanolamines and their different forms of fatty acyl linkage (ethers and plasmalogens), phosphatidylinositols, and hexosylceramides containing nonhydroxy-and hydroxy-fatty acyls. The detailed analysis of identified lipid classes using reversed-phase liquid chromatography in the second dimension was performed for porcine brain to determine more than 160 individual lipid species containing attached fatty acyls of different acyl chain length, double-bond number, and positions on the glycerol skeleton. Lipidomic analysis of plant and animal tissues using HPLC/MS
Lipidy jsou důležitou složkou všech biologických tkání s řadou důležitých rolí
souvisejících se správnou funkcí organismu. Základním požadavkem lipidomických studií
je identifikace a kvantifikace velkého množství lipidů ve složitých biologických vzorcích.
Spojení vysokoúčinné kapalinové chromatografie s hmotnostní spektrometrií má velký
potenciál získat detailní informace o celém lipidomu, ale spolehlivá identifikace a
kvantifikace všech tříd lipidů je stále náročný úkol kvůli velké komplexitě vzorků lipidů a
jejich amfipatickému charakteru.
V první dimenzi byl celkový extrakt lipidů separován pomocí kapalinové
chromatografie hydrofilních interakcí (HILIC) na jednotlivé třídy lipidů. Podmínky HILIC
separace byly optimalizovány za účelem dosáhnout co největšího počtu separovaných tříd
lipidů. Byl testován vliv náplně kolony, složení mobilní fáze, teploty kolony a sklonu
gradientu na účinnost separace. Bylo zjištěno, že množství vody v mobilní fázi má zásadní
vliv na selektivitu a účinnost separace. Optimalizovaná HILIC metoda umožnila separaci
16 tříd lipidů a 3 regioisomerních párů v jedné analýze. Jednotlivé třídy lipidů byly
identifikovány pomocí hmotnostní spektrometrie s ionizací elektrosprejem (ESI-MS). Pro
získání detailních informací byly frakce separovaných tříd lipidů v první dimenzi
preparativně izolovány a separovány v druhé dimenzi pomocí vysokoúčinné kapalinové
chromatografie v systémech s obrácenými fázemi (RP-HPLC) na jednotlivé lipidy. Stejně
jako pro HILIC separaci, zvláštní pozornost byla věnována optimalizaci podmínek za
účelem dosažení maximálního počtu separovaných lipidů. Jednotlivé polární lipidy byly
identifikovány pomocí ESI-MS, zatímco nepolární lipidy pomocí chemické ionizace za
atmosférického tlaku (APCI).
Druhá část práce byla věnována necílené metodě kvantifikace celých tříd lipidů
separovaných HILIC-HPLC/ESI-MS metodou při záznamu kladných iontů pomocí
jednoho interního standardu a odezvových faktorů daných tříd lipidů. Odezvové faktory
byly vypočteny z parametrů kalibračních závislostí interního standardu a standardů
jednotlivých tříd lipidů. Koncentrace dané třídy lipidů byla vypočtena jako součin plochy
píku v HILIC chromatogramu a odezvového faktoru dané třídy vztažené k internímu
standardu. Přesnost a správnost našich výsledků byla potvrzena srovnáním s běžně
používanou metodou kvantitativní analýzy s využitím skenu vybraných iontových reakcí
(SRM) pomocí trojitého kvadrupólu a také kvantifikací pomocí nukleární magnetické resonance. Možnost použití různých typů biologických vzorků byla testována na lidské
referentní plazmě a vaječném žloutku.
V poslední části práce byla provedena detailní charakterizace jednotlivých tříd
lipidů v orgánech prasete (mozek, srdce, ledvina, játra, plíce, mícha, slezina a žaludek)
pomocí naší necílené kvantitativní analýzy. Jednotlivé lipidy uvnitř těchto tříd byly
identifikovány pomocí relativních intenzit deprotonovaných molekul [M-H]- při záznamu
záporných iontů ESI spekter a pomocí off-line dvoudimenzionální HILIC x RPHPLC/
ESI-MS. Průměrné hodnoty počtu atomů uhlíku a počtu dvojných vazeb
analyzovaných orgánů prasete byly vypočteny na základě složení jednotlivých lipidů a
byly použity pro jejich charakterizaci. Složení mastných kyselin v orgánech prasete bylo
po transesterifikaci methanolátem sodným určeno pomocí plynové chromatografie s
plamenově-ionizační detekcí.Lipids are important components in all biological tissues having many essential
roles associated with the proper function of the organism. The identification and
quantitation of a wide range of lipids in complex biological samples is an essential
requirement for the lipidomic studies. High-performance liquid chromatography ? mass
spectrometry coupling has the highest potential to obtain detailed information on the whole
lipidome, but the reliable identification and quantitation of multiple lipid classes is still a
challenging task due to the extreme sample complexity of lipids and their amphipathic
character.
Lipid extracts were separated using hydrophilic interaction liquid chromatography
(HILIC) into individual lipid classes in the first dimension. The HILIC separation
conditions were optimized to achieve the highest number of separated lipid classes. The
influence of column packing, mobile phase composition, separation temperature and
gradient steepness were tested to achieve the best chromatographic resolution. The crucial
influence of water contents in the mobile phase on the separation selectivity and
chromatographic resolution was observed. Optimized HILIC method enabled separate 16
lipid classes and 3 regioisomeric pairs in one analytical run. Individual lipid classes were
identified using electrospray ionization mass spectrometry (ESI-MS). For the detailed
information, the fractions of lipid classes separated in the first dimension were manually
collected and further separated in the second dimension into individual lipid species using
reversed-phase high-performance liquid chromatography (RP-HPLC). As for the HILIC
method, the special attention was paid to the optimization of chromatographic conditions
to achieve the highest number of separated species. ESI-MS was used for the identification
of polar lipid species, while atmospheric pressure chemical ionization (APCI) was prefered
for non-polar lipid species.
The second part of this work was devoted to the nontargeted quantitation of lipid
classes separated by HILIC-HPLC/ESI-MS method in the positive-ion mode using a single
internal standard and lipid class response factors. Response factors were calculated from
parameters of calibration curves measured for the internal standard and standards of
individual lipid classes. The concentration of lipid class was obtained by the peak area in
the HILIC chromatogram multiplied by RF related to the single internal standard. The
accuracy and precision of our results were confirmed by the comparison with established
selected reaction monitoring (SRM) quantitation approach on triple quadrupole mass spectrometer and also nuclear magnetic resonance quantitation. The applicability for
various types of biological samples was tested on human reference plasma and egg yolk.
In the last part of this work, the comprehensive lipidomic characterizations of
porcine organs (brain, heart, kidney, liver, lung, spinal cord, spleen and stomach) were
performed using our developed nontargeted quantitative method of lipid classes. Individual
lipid species inside these classes were determined based on relative abundances of
deprotonated molecules [M-H]- in the negative-ion ESI mode and off-line HILIC x RPHPLC/
ESI-MS. The average values of numbers of carbon atoms and double bonds of
analyzed porcine organs were calculated based on the lipid species composition and used
for their characterization. The fatty acid composition of porcine organs was determined by
gas chromatography with the flame ionization detection after the transesterification with

Analysis of eicosanoids in biological samples using liquid chromatography and mass spectrometry
Eikosanoidy hrají důležitou roli při probíhajícím zánětu v lidském těle. Vznikají enzymatickou i neenzymatickou oxidací arachidonové kyseliny a jiných polynenasycených mastných kyselin. Pro analýzu eikosanoidů byla použita ultravysokoúčinná kapalinová chromatografie ve spojení s hmotnostní spektrometrií (UHPLC/MS) pracující v negativním módu elektrosprejové ionizace a s použitím trojitého kvadrupólového analyzátoru. Analyty byly separovány na koloně Acquity UPLC BEH C18 (2.1x150 mm, 1.7 μm). Všechny důležité parametry iontového zdroje, jako teplota iontového zdroje a průtoky plynů, byly optimalizovány tak, aby byla získána největší citlivost pro následnou SRM (Selected reaction monitoring) metodu. MS/MS spektra eikosanoidů vykazují více produktových iontů, avšak nejdůležitější produktový ion pro následné SRM byl ten s největší intenzitou. Kolizní energie byla optimalizována pro každý prekurzor/produkt iontový pár využitím SRM detekce. Tato metoda bude následně validovaná a aplikovaná na vzorky lidské plazmy.Eicosanoids play an important role in the ongoing inflammation in the human body. Eicosanoids are formed by the enzymatic or non-enzymatic oxidation of arachidonic acid or polyunsaturated fatty acids. For the analysis of eikosanoids, we used an ultrahigh-performance liquid chromatography (UHPLC) coupled to negative-ion electrospray ionization mass spectrometry using a triple quadrupole analyzer. Analytes were separated in reversed-phase mode using Acquity UPLC BEH C18 (2.1x150 mm, 1.7 μm) column. All important parameters, such as turbo ion spray source temperature, nebulizer, and turbo gas flow have been optimized to achieve the highest sensitivity for subsequent selected reaction monitoring (SRM) transitions. MS/MS spectra of eicosanoids show multiple product ions. The collision energy was optimized for each precursor / product ion pair used for SRM detection. This method will be validated and applied to samples of human plasma
Lipidomic analysis of porcine organs using hydrophilic interaction liquid chromatography and mass spectroscopy
The identification and quantitation of all lipids in complex biological tissues is the
first step towards the understanding how lipids function in a biological system and
the elucidation of the mechanism of lipid-related diseases including obesity,
atherosclerosis, cancer, cardiovascular diseases, etc. Our optimized nontargeted
method using hydrophilic interaction liquid chromatography (HILIC) coupled to
electrospray ionization mass spectrometry (ESI-MS) was used for the
characterization of the lipidome, mainly glycerophospholipids and sphingolipids
in porcine brain, heart, kidney, liver, lung, spinal cord, spleen and stomach.
Individual lipid classes are quantified based on the peak integration of individual
lipid classes separated in the HILIC mode multiplied by their response factors and
correlated by sphingosylethanolamine (d17:1/12:0) as a single internal standard.
Subsequently, relative abundances of deprotonated molecules [M-H]– in negativeion
ESI mass spectra are used for determination of lipid species concentration
inside individual classes. The approach using [M-H]– ions enables to obtain detailed information about phosphatidylethanolamines and their different forms
of fatty acyl linkage on the glycerol skeleton (plasmalogens and ether analogs),
phosphatidylinositols and hexosylceramides in studied organs. Our results
provide important knowledge about lipid representations in vital porcine organs
and can be applied for future metabolic studies on human to investigate serious
lipid-related human disorders
Hydrofilní interakční chromatografie ve spojen s hmotnostní spektrometrií pro stanovení kyseliny fosfatidové, fosfatidylserinu a jejich lysoderivátů.
The goal of this work is a systematic optimization of hydrophilic interaction liquid chromatography (HILIC) separation of acidic lipid classes (namely phosphatidic acids-PA, lysophosphatidic acids-LPA, phosphatidylserines-PS and lysophosphatidylserines-LPS) and other lipid classes under mass spectrometry (MS) compatible conditions. The main parameters included in this optimization are the type of stationary phases used in HILIC, pH of the mobile phase, the type and concentration of mobile phase additives. Nine HILIC columns with different chemistries (unmodified silica, modified silica using diol, 2-picolylamine, diethylamine and 1-aminoanthracene and hydride silica) are compared with the emphasis on peak shapes of acidic lipid classes. The optimization of pH is correlated with the theoretical calculation of acidobasic equilibria of studied lipid classes. The final method using the hydride column, pH 4 adjusted by formic acid and the gradient of acetonitrile and 40 mmol/L of aqueous ammonium formate provides good peak shapes for all analyzed lipid classes including acidic lipids. This method is applied for the identification of lipids in real samples of porcine brain and kidney extracts.Cílem této práce byla optimalizace separace polárních lipidů zejména kyseliny fosfatidové, fosfatidylserinu a jejich lysoderivátů. Retenční chování lipidů s použitím kapalinové chromatografie s reverzními fázemi ve spojení s hmotnostní spekltrometrií s elektrosprejovou ionizací
Tato práce se zabývá retenčním chování lipidů v biologických vzorcích jako je lidská plasma a moč a prasečí mozek. Pro práci byla použita kapalinová chromatografie s reverzními fázemi ve spojení s hmotnostní spektrometrií. Identifikace jednotlivých lipidů byla provedena v negativním a pozitivním módu s přesností lepší než 5 ppm. Bylo identifikováno 14 polárních a 5 nepolárních tříd a více než 400 jednotlivých lipidů
HILIC/MS analysis of (lyso)phosphatidic acids, (lyso)phosphatidylserines and other lipidclasses
Lipidomika je obor zabývající se studiem biochemických drah lipidů v biologických systémech. Cílem lipidomiky je pochopení funkce lipidů v biologickém metabolismu, vysvětlení mechanismu vzniku a průběhu některých metabolických chorob. Cílem této práce byla optimalizace separace co největšího počtu tříd lipidů s využitím kapalinové chromatografie hydrofilních interakcí (HILIC) za podmínek vhodných pro spojení s hmotnostní spektrometrií. Zvláštní důraz byl při optimalizaci HILIC podmínek kladen na ty třídy polárních lipidů, které se dosud touto technikou nedařilo analyzovat, fosfatidylseriny, kyseliny fosfatidové a jejich lysoderiváty. V této práci byly studovány vlivy typu náplně kolony, složení mobilní fáze, aditiva, pH mobilní fáze, teploty kolony a sklonu gradientu na účinnost separace. Největší vliv na separaci těchto tříd lipidů má typ náplně kolony a pH mobilní fáze. Jednotlivé třídy lipidů byly identifikovány díky spojení s hmotnostní spektrometrií (MS) s využitím ionizace elektrosprejem v kladném i záporném módu záznamu iontů. Optimalizovaná metoda byla použita pro charakterizaci jednotlivých lipidů v extraktu prasečího mozku a ledviny.Lipidomics is a discipline dealing with studies of biochemical pathways of lipids in biological systems. The goal of lipidomics is to understand the function of lipids in biological metabolism, to explain the mechanism of the development and progress of some metabolic diseases. The aim of this work was the optimization of hydrophilic interaction liquid chromatography (HILIC) separation of the greatest number of classes of lipids using conditions compatible with the mass spectrometry. A special attention of this optimization has been paid to lipid classes not yet analyzed by HILIC so far, i.e. phosphatidylserines, phosphatidic acids and their lyso-derivatives. LIPIDOMIC ANALYSIS OF BIOLOGICAL SAMPLES USING DIRECT- INFUSION MASS SPECTROMETRY
Lipidomic analysis of biological samples in clinical research represents a challenging task due to the extreme complexity of samples and their large number. One of the ways to get complex information on the composition of sample is to use direct infusion mass spectrometry (DI-MS). The DI-MS technique was used to measure plasma samples of patients with renal cancer and healthy volunteers.Lipidomická analýza biologických vzorků v klinickém výzkumu představuje náročný úkol, který je dán vysokou komplexností a velkým počtem zpracovávaných vzorků. Identifikace gangliosidů v biologických vzorcích pomocí kapalinové chromatografie hydrofilních interakcí- hmotnostní spektrometrii.
The hydrophilic interaction liquid chromatography (HILIC) coupled to a negative-ion electrospray ionization tandem mass spectrometry (ESI-MS/MS) method has been developed for the identification of a wide range of gangliosides in biological samples. Gangliosides consist of a backbone of sphingoid base and a polar oligosaccharide chain containing at least one sialic acid. Gangliosides are extracted by chloroform-methanol-water mixture, where an upper aqueous layer containing gangliosides and other polar lipid subclasses is further purified by C18 solid-phase extraction. The optimization of chromatographic conditions includes the column selection, mobile-phase composition, pH value, buffer type, and concentration with the goal to achieve the best chromatographic resolution and MS sensitivity. The identification of gangliosides and other polar lipids is based on accurate m/z values of [M-H]- ions and fragment ions as well measured by high-resolution MS. The detailed interpretation of MS/MS spectra enables the generalization of fragmentation pathways, which is then used for the differentiation of a, b, and c series of gangliosides. The structural assignment is further confirmed by agreement with the predicted retention behavior in HILIC mode on the basis of the correlation among the ganglioside retention, the number of saccharide units, and their sequence. The final HILIC/ESI-MS/MS method is applied for the analysis of porcine brain, human kidney, lungs, plasma, and erythrocytes resulting in unambiguous identification of 145 ganglioside species from 19 subclasses, which represents the highest number of reported gangliosides. Moreover, 71 sulfatides and 59 polar phospholipids (phosphatidylserines, phosphatidylinositols, lysophosphatidylinositols, and phosphatidylglycerols) are detected within a 15 min run.Pro identifikaci široké škály gangliosidů v biologických vzorcích byla vyvinuta metoda založená na chromatografii hydrofilních interakcí (HILIC) ve spojení s tandemovou hmotnostní spektrometrií s ionizací elektrosprejem (ESI-MS/MS). Gangliosidy se skládají ze sfingoidové báze a polárního oligosacharidového řetězce obsahujícího alespoň jednu kyselinu sialovou. Gangliosidy se extrahují směsí chloroformu, methanolu a vody, přičemž horní vodná vrstva obsahující gangliosidy a jiné polární lipidové podtřídy se dále čistí extrakcí C18 v pevné fázi. Optimalizace chromatografických podmínek zahrnuje výběr kolony, složení mobilní fáze, hodnotu pH s cílem dosáhnout nejlepšího chromatografického rozlišení a citlivosti na MS. Identifikace gangliosidů a jiných polárních lipidů je založena na přesných m/z hodnotách iontů [M-H] a iontů fragmentů, měřených MS s vysokým rozlišením. Podrobná interpretace spektra MS/MS umožňuje zobecnění fragmentačních cest, které se pak používají pro diferenciaci gangliosidů řady a, b, c. Strukturní zařazení je dále potvrzeno předpokládaným retenčním chováním v režimu HILIC na základě korelace mezi retencí gangliosidů, počtem sacharidových jednotek a jejich sekvencí. Pro analýzu byly použity vzorky prasečího mozku, lidských ledvin, plic, plazmy a erytrocytů. Použitá metoda HILIC/ESI-MS/MS vede k jednoznačné identifikaci 145 druhů gangliosidů z 19 podtříd, což představuje nejvyšší počet zaznamenaných gangliosidů. Kromě toho bylo v průběhu 15 minut zjištěno 71 sulfatidů a 59 polárních fosfolipidů (fosfatidylserinů, fosfatidylinositolů, lysofosfatidylinositolů a fosfatidylglycerolů)

Gangliosidy jsou třída lipidů obsahující ceramid na kterém jsou navázány cukerné jednotky a alespoň jedna kyselina sialová. Tyto lipidy jsou hojně zastoupeny v centrální nervové soustavě a hrají v buňkách důležitou roli v mnoha fyziologických procesech, jako je kontrola paměti, buněčná signalizace, ochrana neuronů nebo apoptóza. Gangliosidy jsou extrahovány pomocí směsi chloroform, methanol a voda. Vodná fáze obsahující gangliosidy je následně přečištěna na C18 kolonkách. Přečištění se provádí pro odstranění solí. Hlavním cílem této práce je vyvinutí spolehlivé metody pro analýzu gangliosidů. Metoda je založena na HILIC separaci ve spojení s ESI-MS.
