    Zanthoxylum ailanthoides

    Zanthoxylum ailanthoides (ZA) has been used as folk medicines in East Asian and recently reported to have several bioactivity; however, the studies of ZA on the regulation of triacylglycerol (TG) biosynthesis have not been elucidated yet. In this study, we examined whether the methanol extract of ZA (ZA-M) could reduce oleic acid- (OA-) induced intracellular lipid accumulation and confirmed its mode of action in HepG2 cells. ZA-M was shown to promote the phosphorylation of AMPK and its upstream LKB1, followed by reduction of lipogenic gene expressions. As a result, treatment of ZA-M blocked de novo TG biosynthesis and subsequently mitigated intracellular neutral lipid accumulation in HepG2 cells. ZA-M also inhibited OA-induced production of reactive oxygen species (ROS) and TNF-α, suggesting that ZA-M possess the anti-inflammatory feature in fatty acid over accumulated condition. Taken together, these results suggest that ZA-M attenuates OA-induced lipid accumulation and inflammation through the activation of LKB1/AMPK signaling pathway in HepG2 cells

    Pollen Application Methods Affecting Fruit Quality and Seed Formation in Artificial Pollination of Yellow-Fleshed Kiwifruit

    This study investigated pollen application methods for artificial pollination in tetraploid kiwifruit cultivars ‘Halla Gold’ and ‘Sweet Gold’ grown in a nonheated plastic-film house in Jeju, Korea. Pollen of the hexaploid cultivar ‘Bohwa’ (A. chinensis var. deliciosa) bred in Korea was used for artificial pollination. We examined the effect of repeated pollination, pretreatment of stigma with wetting materials, application of dry pollen or pollen in suspension on fruit quality, and seed formation. With repeated pollination, pollen tubes in the pistil reached and penetrated the ovule three days after artificial pollination, although the pattern varied depending on the number of dry pollen applications. In both cultivars, the number of pollen tubes was clearly higher following repeated pollination than following single pollination, and fruit weight, dry matter (DM), number of seeds, and 100-seed weight were also higher. When pistillate flowers were pollinated with dry pollen immediately after water sprinkle, both cultivars showed the lowest fruit weight, DM, firmness, number of seeds, and 100-seed weight, whereas there were no significant differences in fruit quality or seed formation for dry pollen application 1 h after water sprinkle, or immediately or 1 h after suspension medium sprinkle. For pollination using a pollen suspension, the fruit weight was lower in both cultivars. There were no significant differences in fruit quality and seed formation following application of dry pollen or a pollen suspension, except for fruit weight in ‘Sweet Gold’. It could be seen from the results of this study that raindrops or dewdrops on the stigma might reduce the efficiency of artificial pollination using dry pollen. Still, the application of repeated pollination enhanced the efficiency of artificial pollination

    Hip Muscle Strength and Ratio Differences in Delivery Workers With and Without Iliotibial Band Syndrome

    Background: Delivery workers repeatedly get in and out of trucks and walk or run to deliver packages during work. Iliotibial band syndrome (ITBS) is a well-known non-traumatic overuse injury of the lateral side of the knee caused by frequent knee flexion and extension. Hip muscle strength is among the factors that prevent lower extremity injuries. Although many studies have examined the relationship between ITBS and hip muscle strengths, there was no study comparing hip muscle strength and ratio between delivery workers with and without ITBS. Objects: This study aimed to compare hip muscle strength and hip internal/external rotator and adductor/abductor strength ratios between delivery workers with and without ITBS. Methods: Fourteen delivery workers with ITBS matched inclusion criteria in the present study among 20 participants. Because total sample size was required 28 subjects by G*power program (ver. 3.1.9.4; University of Trier), 14 delivery workers without ITBS were recruited. Hip muscle strengths were measured in a side-lying position using a Smart KEMA pulling sensor (KOREATECH Co. Ltd.). An independent t-test was used to compare hip muscle strengths and hip internal/external rotator and hip adductor/abductor strength ratios between delivery workers with and without ITBS. Results: The adductor/abductor strength ratio was significantly greater in delivery workers without ITBS than in those with ITBS (p < 0.05). The strengths of the hip abductor, hip adductor, hip internal rotator, hip external rotator, and the ratio of internal/external rotator strengths were not significantly different between the delivery workers with and without ITBS (p > 0.05). Conclusion: This study’s findings showed that delivery workers with ITBS had significantly lesser adductor/abductor strength ratio, while the strengths of the hip abductor and adductor muscles did not differ significantly. These results suggest that adductor/abductor strength ratio should be considered when evaluating and treating ITBS in delivery workers

    Serum Krebs von den Lungen-6 level predicts disease progression in interstitial lung disease.

    Disease progression (DP) in interstitial lung disease (ILD) is variable and difficult to predict. In previous reports, serum Krebs von den Lungen-6 (KL-6) was suggested to be useful in diagnosing and predicting survival in ILD. The aim of our study was to investigate the usefulness of serum KL-6 as a predictor of DP in ILD. Clinical data of 199 patients with ILD (idiopathic pulmonary fibrosis: 22.8%) were prospectively collected and serum KL-6 levels were measured. DP was defined as a relative decline in forced vital capacity (FVC) ≥ 10%, acute exacerbation, or death during follow-up. The median follow-up period was 11.1 months. The mean age of the subjects was 62.2 years, and 59.8% were male. DP occurred in 21.6% of patients. The progressed group showed lower FVC, lower diffusing capacity for carbon monoxide, lower the minimum oxygen saturation during the 6-minute walk test, higher fibrosis scores on high-resolution computed tomography, and higher KL-6 levels (826.3 vs. 629.0 U/mL; p < 0.001) than those of the non-progressed group. In receiver operating characteristic curve analysis, serum KL-6 levels were a significant predictor of DP in ILD (area under the curve = 0.629, p = 0.009, and the optimal cut-off level was 811 U/mL). In multivariable Cox analysis, high serum KL-6 levels (≥ 800 U/mL) were only independently associated with DP in ILD (HR 2.689, 95% CI 1.445-5.004, P = 0.002). Serum KL-6 levels might be useful to predict DP in patients with ILD

    ‘Minihyang’ mandarin bears fruits with small size and high sugar content. ‘Minihyang’ mandarin grafted on trifoliate orange (TO) tends to be vigorous and develops water sprout open. It is associated with insufficient floral differentiation and fruit set. Recently, the use of Flying Dragon (FD) as rootstock with a high dwarf effect has been proposed to improve this situation. Therefore, this study was conducted to evaluate the effect of two different rootstock genotypes on tree growth, fruit yield, and fruit quality at the physiological, biochemical, and molecular levels. As a result of the study, in FD, tree vigor was stably maintained, fruit size was large, and the sugar content was high compared to the TO. Fructose, glucose, and sucrose of fruit continued to increase from development to maturity. In particular, fructose and sucrose were significantly higher in the fruits of the FD than those in TO at 150 and 220 days after anthesis. The total sugar content was also significantly higher in the fruit of the FD. The activities of SPS and SS associated with sucrose synthesis tended to be increased during the fruit maturity season, but there was no significant difference between the two rootstocks. On the other hand, the activities of SS and AI breaking down sucrose were high in FD at 150 and 220 days after anthesis. These results suggest that the unloading of sucrose might be increased and affect the sugar content. However, the results of real-time PCR analysis of gene expression related to sucrose metabolism did not show an association with changes in enzyme activity affecting sugar content. Therefore, further detailed studies on the process after the regulation of gene expression are likely to be needed

    Chronic obstructive pulmonary disease (COPD) is a chronic inflammatory lung disease which causes breathing problems. YPL-001, consisting of six iridoids, has potent inhibitory efficacy against COPD. Although YPL-001 has completed clinical trial phase 2a as a natural drug for COPD treatment, the most effective iridoid in YPL-001 and its mechanism for reducing airway inflammation remain unclear. To find an iridoid most effectively reducing airway inflammation, we examined the inhibitory effects of the six iridoids in YPL-001 on TNF or PMA-stimulated inflammation (IL-6, IL-8, or MUC5AC) in NCI-H292 cells. Here, we show that verproside among the six iridoids most strongly suppresses inflammation. Both TNF/NF-κB-induced MUC5AC expression and PMA/PKCδ/EGR-1-induced IL-6/-8 expression are successfully reduced by verproside. Verproside also shows anti-inflammatory effects on a broad range of airway stimulants in NCI-H292 cells. The inhibitory effect of verproside on the phosphorylation of PKC enzymes is specific to PKCδ. Finally, in vivo assay using the COPD-mouse model shows that verproside effectively reduces lung inflammation by suppressing PKCδ activation and mucus overproduction. Altogether, we propose YPL-001 and verproside as candidate drugs for treating inflammatory lung diseases that act by inhibiting PKCδ activation and its downstream pathways

    Diplacone Isolated from <i>Paulownia tomentosa</i> Mature Fruit Induces Ferroptosis-Mediated Cell Death through Mitochondrial Ca<sup>2+</sup> Influx and Mitochondrial Permeability Transition

    The recently defined type of cell death ferroptosis has garnered significant attention as a potential new approach to cancer treatment owing to its more immunogenic nature when compared with apoptosis. Ferroptosis is characterized by the depletion of glutathione (GSH)/glutathione peroxidase-4 (GPx4) and iron-dependent lipid peroxidation. Diplacone (DP), a geranylated flavonoid compound found in Paulownia tomentosa fruit, has been identified to have anti-inflammatory and anti-radical activity. In this study, the potential anticancer activity of DP was explored against A549 human lung cancer cells. It was found that DP induced a form of cytotoxicity distinct from apoptosis, which was accompanied by extensive mitochondrial-derived cytoplasmic vacuoles. DP was also shown to increase mitochondrial Ca2+ influx, reactive oxygen species (ROS) production, and mitochondrial permeability transition (MPT) pore-opening. These changes led to decreases in mitochondrial membrane potential and DP-induced cell death. DP also induced lipid peroxidation and ATF3 expression, which are hallmarks of ferroptosis. The ferroptosis inhibitors ferrostatin-1 and liproxstatin-1 were effective in counteracting the DP-mediated ferroptosis-related features. Our results could contribute to the use of DP as a ferroptosis-inducing agent, enabling studies focusing on the relationship between ferroptosis and the immunogenic cell death of cancer cells

    Polyacetylene From Dendropanax morbifera Alleviates Diet-Induced Obesity and Hepatic Steatosis by Activating AMPK Signaling Pathway

    The extract tea of Dendropanax morbifera is popular beverages in Korea, and their preventive and therapeutic roles in metabolic disorders have been reported. However, the molecular mechanism has not been studied despite the known efficacy of D. morbifera. Eleven fractions (fr.1–fr.11) were divided by MPLC to find the active compound. Among them, Fr.5 was superior to others in that the inhibitory efficacy of de novo triglyceride (TG) biosynthesis. NMR analysis revealed that Fr.5 is composed 98% or more (9Z,16S)-16-hydroxy-9,17-octadecadiene-12,14-diynoic acid (HOD). Treatment of HOD diminished oleic acid (OA)-induced TG accumulation in HepG2 hepatocytes and differentiation of 3T3-L1 preadipocytes by activating LKB1/AMPK. In addition, we determined the effect of the oral administration of the extract of D. morbifera on obesity and hepatic steatosis in high-fat diet (HFD)-induced obese mice. This study proved that D. morbifera containing HOD, the active substance, can show preventive or therapeutic efficacy on obesity and hepatic steatosis through the targeting LKB1/AMPK axis
