    Implications of a Changing Agro-Ecosystem on the Nascent Wine Grape Production Industry in Idaho

    Wine-grape growing in the state of Idaho is just now coming into its own. Unlike traditional and established row crops, sustainable wine grape production requires the encouragement and development of new agricultural capacity in the state. Concurrently, creating a sustainable wine grape production culture in the state may be constrained and impacted by a changing climate and related stresses on the ecosystem of the semi-arid inland northwest. An understanding of the effect that a changing ecosystem will have on the viability of growing wine grapes, as well as an examination of the challenges that both farmers and policymakers will face in developing and maintaining wine grape production is critical to the successful establishment of a vibrant viticulture in Idaho

    Twenty-Three Unsolved Problems in Hydrology (UPH): A Community Perspective

    This paper is the outcome of a community initiative to identify major unsolved scientific problems in hydrology motivated by a need for stronger harmonisation of research efforts. The procedure involved a public consultation through online media, followed by two workshops through which a large number of potential science questions were collated, prioritised, and synthesised. In spite of the diversity of the participants (230 scientists in total), the process revealed much about community priorities and the state of our science: a preference for continuity in research questions rather than radical departures or redirections from past and current work. Questions remain focused on the process-based understanding of hydrological variability and causality at all space and time scales. Increased attention to environmental change drives a new emphasis on understanding how change propagates across interfaces within the hydrological system and across disciplinary boundaries. In particular, the expansion of the human footprint raises a new set of questions related to human interactions with nature and water cycle feedbacks in the context of complex water management problems. We hope that this reflection and synthesis of the 23 unsolved problems in hydrology will help guide research efforts for some years to come

    Reflections on Recent Recommendations on the Use of Science in Disaster Risk Reduction Using Case Studies from Bangladesh and the Western United States

    The valuable role that science has to play in disaster preparedness and risk reduction is widely recognized and was highlighted during the development of the successor to the Hyogo Framework for Action for disaster risk reduction that was adopted in March 2015. However, there are many factors that limit how effectively science can inform both disaster risk reduction policy and practice. Understanding these factors and taking steps to overcome them require a broad view, and a comparative approach can be instructive. We focus on two projects that were independently completed by the authors: earthquake risk management in Bangladesh and flooding and wildfires management in the United States. We use each case to reflect on the implications of recent recommendations made by the Science and Technology Advisory Group (STAG) of the United Nations Office for Disaster Risk Reduction that attempt to increase the integration of science in disaster risk reduction policy making. We then use the STAG recommendations as a framework for integrating our independent case study fi ndings. Despite the differences in the geographic contexts and hazards being considered, these examples broadly support the STAG recommendations. However, the fine details of the way in which science is used in decision making need to be given careful consideration if science is to fully support disaster risk reduction. Although our collective observations suggest that science is an important part of the disaster risk reduction (DRR) process, suggesting that it is “key to post-2015 DRR efforts” as the STAG recommendations do, may perhaps overstate the role that science is able to play

    Dominant gain-of-function mutations in Hsp104p reveal crucial roles for the middle region

    Heat-shock protein 104 (Hsp104p) is a protein-remodeling factor that promotes survival after extreme stress by disassembling aggregated proteins and can either promote or prevent the propagation of prions (protein-based genetic elements). Hsp104p can be greatly overexpressed without slowing growth, suggesting tight control of its powerful protein-remodeling activities. We isolated point mutations in Hsp104p that interfere with this control and block cell growth. Each mutant contained alterations in the middle region (MR). Each of the three MR point mutations analyzed in detail had distinct phenotypes. In combination with nucleotide binding site mutations, Hsp104p(T499I) altered bud morphology and caused septin mislocalization, colocalizing with the misplaced septins. Point mutations in the septin Cdc12p suppressed this phenotype, suggesting that it is due to direct Hsp104p–septin interactions. Hsp104p(A503V) did not perturb morphology but stopped cell growth. Remarkably, when expressed transiently, the mutant protein promoted survival after extreme stress as effectively as did wild-type Hsp104p. Hsp104p(A509D) had no deleterious effects on growth or morphology but had a greatly reduced ability to promote thermotolerance. That mutations in an 11-amino acid stretch of the MR have such profound and diverse effects suggests the MR plays a central role in regulating Hsp104p function

    This quantitative correlational research study examined if a relationship existed between perceived servant leadership behaviors of fire and emergency service leaders and employee job satisfaction in fire and emergency services personnel. The study involved n= 205 participants who completed the Organizational Leadership Assessment and the data were analyzed using Spearman’s correlation. The results showed that there is a statistically significant relationship between servant leadership and employee job satisfaction in fire and emergency services personnel

    Subunit interactions influence the biochemical and biological properties of Hsp104

    Point mutations in either of the two nucleotide-binding domains (NBD) of Hsp104 (NBD1 and NBD2) eliminate its thermotolerance function in vivo. In vitro, NBD1 mutations virtually eliminate ATP hydrolysis with little effect on hexamerization; analogous NBD2 mutations reduce ATPase activity and severely impair hexamerization. We report that high protein concentrations overcome the assembly defects of NBD2 mutants and increase ATP hydrolysis severalfold, changing V(max) with little effect on K(m). In a complementary fashion, the detergent 3-[(3-cholamidopropyl)dimethylammonio]-1-propanesulfonate inhibits hexamerization of wild-type (WT) Hsp104, lowering V(max) with little effect on K(m). ATP hydrolysis exhibits a Hill coefficient between 1.5 and 2, indicating that it is influenced by cooperative subunit interactions. To further analyze the effects of subunit interactions on Hsp104, we assessed the effects of mutant Hsp104 proteins on WT Hsp104 activities. An NBD1 mutant that hexamerizes but does not hydrolyze ATP reduces the ATPase activity of WT Hsp104 in vitro. In vivo, this mutant is not toxic but specifically inhibits the thermotolerance function of WT Hsp104. Thus, interactions between subunits influence the ATPase activity of Hsp104, play a vital role in its biological functions, and provide a mechanism for conditionally inactivating Hsp104 function in vivo

    Data Associated with A Simplified Approach to Stakeholder Engagement in Natural Resource Management: The Five-Feature Framework

    This paper distills complex frameworks for stakeholder engagement to five main principles that scientists and natural resource managers can use in planning stakeholder engagement efforts. Many natural resource management professionals, including practitioners and scholars, increasingly recognize the need for and potential benefits of engaging stakeholders in complex decision-making processes, yet the implementation of these efforts varies wildly, reflecting great methodological and conceptual diversity. Given the dynamic and diverse natural resource management contexts in which engagement occurs and the often significant stakes involved in making decisions about natural resources, we argue that stakeholder engagement would benefit from a theoretical framework that is both agile and robust. To this end, five essential elements of stakeholder engagement are evaluated and organized to form the Five-Feature Framework, providing a functional and approachable platform with which to consider engagement processes. Aside from introducing and developing the Five-Feature Framework, this paper applies the framework as a measure to evaluate the empirical case-study literature involving SE in natural resource management in an effort to better understand the obstacles facing robust and genuine engagement in natural resource management. Our results suggest that the most basic principles of engagement are often absent from stakeholder engagement projects, confirming the need for a functional framework. The Five-Feature Framework can be used to plan flexible, adaptable, and rigorous engagement projects in a variety of contexts and with teams that have varying backgrounds and experience. By virtue of its simplicity and functionality, the framework demystifies stakeholder engagement in order to help natural resource professionals build opportunities for collaborative decision-making and integrate citizen values and knowledge into complex management issues

    Effect of High In-Situ Stress on Braced Excavations

    The two underground stations and portals of Metro Gold Line\u27s East Los Angeles extension were excavated in heavily over-consolidated alluvium. The excavations were supported with heavy soldier piles with pre-loaded steel-pipe struts. When measured strut loads increased to up to 3 times the design value, and strut-waler connections began to buckle, the contractor was directed to install additional struts. Maintaining that the problem had been caused by inadequate construction means and methods, the owner denied a change-order request for this work. This paper describes the contractor’s investigation into the cause of strut overloading in preparation for a formal hearing by a Dispute Resolution Board. The study concluded that the extremely high bracing loads were caused by high in-situ stresses in the region, which had not been accounted for in the shoring-pressure diagrams provided in the contract drawings

    Worry as a window into the lives of people who use injection drugs: a factor analysis approach

    <p>Abstract</p> <p>Background</p> <p>The concept of risk dominates the HIV/AIDS literature pertaining to People Who Use Injection Drugs (PWUID). In contrast the associated concept of worry is infrequently applied, even though it can produce important perspectives of PWUID's lives. This study asked a sample (n = 105) of PWUID enrolled in a Victoria, British Columbia needle exchange program to evaluate their degree of worry about fourteen factors they may encounter in their daily lives.</p> <p>Methods</p> <p>Exploratory factor analysis was used to analyze their responses.</p> <p>Results</p> <p>Factor analysis delineated three factors: 1) overall personal security, 2) injection drug use-specific risks including overdosing and vein collapse and, 3) contracting infectious diseases associated with injection drug use (e.g. HIV/AIDS and hepatitis C).</p> <p>Conclusion</p> <p>PWUID in this study not only worry about HIV/AIDS but also about stressful factors in their daily life which have been linked to both increased HIV/AIDS risk behaviour and decreased anti-retroviral treatment adherence. The importance PWUID give to this broad range of worry/concerns emphasizes the need to place HIV/AIDS intervention, education, and treatment programs within a broader harm-reduction framework that incorporates their perspectives on both worry and risk.</p
