    Navigating the Rough Waters of Change : The New OhioLINK ETD Service

    On January 14, 2013, the OhioLINK community was informed that the Electronic Theses & Dissertations (ETD) service was down for emergency maintenance. Few in the community could have predicted that single event would trigger a tidal wave of changes that ultimately resulted in a complete system rebuild. Panel participants will discuss the situation from their perspectives as OhioLINK ETD stakeholders. Implementing a new consortial system is a tricky undertaking even with adequate time to plan for the development. What additional pressures come into play when an emergency necessitates rapid development and implementation? What is the impact of changing to a new system in the middle of the academic year during the weeks leading up to the community’s busiest ETD season? How will the hundreds of papers submitted but not approved before the system went down be handled? What adjustments will stakeholders need to make in the short term to realize the long term benefit of an improved consortial ETD submission and approval system? What improvements have been realized with the new system? Communication is vitally important during times of transition. What successes and/or failures of communication can be identified in hindsight? How would the situation be different if the Ohio ETD community had not gathered in the summer of 2012 to discuss their needs and desires for a better system that provided a foundation for functional requirements? What are these new functions and how are they working so far? What future enhancements are on the horizon for the new system? The panelists will discuss these and other issues in a session designed for active audience participation

    Relationship between personality change and the onset and course of alcohol dependence in young adulthood

    Aims  To examine the reciprocal effects between the onset and course of alcohol use disorder (AUD) and normative changes in personality traits of behavioral disinhibition and negative emotionality during the transition between adolescence and young adulthood. Design  Longitudinal–epidemiological study assessing AUD and personality at ages 17 and 24 years. Setting  Participants were recruited from the community and took part in a day‐long, in‐person assessment. Participants  Male ( n  = 1161) and female ( n  = 1022) twins participating in the Minnesota Twin Family Study. Measurements  The effects of onset (adolescent versus young adult) and course (persistent versus desistent) of AUD on change in personality traits of behavioral disinhibition and negative emotionality from ages 17 to 24 years. Findings  Onset and course of AUD moderated personality change from ages 17 to 24 years. Adolescent onset AUD was associated with greater decreases in behavioral disinhibition. Those with an adolescent onset and persistent course failed to exhibit normative declines in negative emotionality. Desistence was associated with a ‘recovery’ towards psychological maturity in young adulthood, while persistence was associated with continued personality dysfunction. Personality traits at age 11 predicted onset and course of AUD, indicating personality differences were not due to active substance abuse. Conclusions  Personality differences present prior to initiation of alcohol use increase risk for alcohol use disorder, but the course of alcohol use disorder affects the rate of personality change during emerging adulthood.     Affected Family Members\u27 Communicative Management of Opioid Misuse Stigma: Applying and Rethinking the Stigma Management Communication Typology

    Opioid misuse is a prevalent health problem in the United States with consequences extending past the person who misuses opioids to affected family members (AFM) through courtesy stigma. The goals of this study were to understand the stigma management communication (SMC) strategies employed by AFMs when they experience courtesy stigma and changes in strategies used over time. The findings from interviews with 34 AFMs suggest the SMC strategies they employ range from those that indicate acceptance of stigma and avoidance of stigma situations to strategies where AFMs actively challenge opioid misuse stigma. However, strategy use depended on the social context and AFMs’ perceptions of opioid misuse stigma at a given moment in time. Further, findings suggest changes in AFMs’ SMC strategies over time are related to changes in their perceptions of opioid misuse stigma. Theoretical and practical implications of how families manage stigma are discussed

    Personality and Psychopathology: A Stagnant Field in Need of Development

    A dominant paradigm in psychopathology research proposes that individual differences in personality are centrally involved in the origins and manifestations of psychopathology, and structural models of personality and psychopathology have been extremely useful in helping to organize associations among many traits and disorders. However, these models merely describe patterns of covariation; they do not explain the processes by which these patterns emerge. We argue that the field is stagnated, as it is overly focused on the demonstration of concurrent associations and on confirming a spectrum model that proposes traits and disorders are manifestations of the same underlying constructs. We contend that if the field is to move towards an understanding of causal processes, it must integrate knowledge and principles of personality development and developmental psychopathology. To begin this integration, we review (i) normative trends in personality change, (ii) age‐related changes in the prevalence of disorders, and (iii) the impact of onset and chronicity on the severity of disorders. We propose several developmental processes that may contribute to the co‐development of personality and psychopathology. We then present novel empirical findings to illustrate how a developmental perspective on traits and disorders can inform new hypotheses and propose principles and hypotheses that should guide future research.     Codevelopment Between Key Personality Traits and Alcohol Use Disorder From Adolescence Through Young Adulthood

    ObjectivePersonality traits related to negative emotionality and low constraint are strong correlates of alcohol use disorder (AUD), but few studies have evaluated the prospective interplay between these traits and AUD symptoms from adolescence to young adulthood.MethodThe Minnesota Twin Family Study (N = 2,769) was used to examine the developmental interplay between AUD symptoms and three personality measures of constraint, negative emotionality, and aggressive undercontrol from ages 17 to 29.ResultsResults from random‐intercept, cross‐lagged panel models showed that low constraint and aggressive undercontrol predicted subsequent rank‐order increases in AUD symptoms from ages 17 to 24. AUD symptoms did not predict rank‐order change in these traits from ages 17 to 24. There was support for both cross‐effects from ages 24 to 29. Biometric analysis of the twin data showed genetic influences accounted for most of the phenotypic correlations over time.ConclusionResults are consistent with the notion that personality traits related to low constraint and aggressive undercontrol are important vulnerability/predisposition factors for the development of early adult AUD. In later young adulthood, there is more evidence for the simultaneous codevelopment of personality and AUD.     Streamlined preparation of genomic DNA in agarose plugs for pulsed-field gel electrophoresis

    Genome analysis using pulsed-field gel electrophoresis (PFGE) has been used in applications ranging from typing bacterial strains to radiobiology to cancer research. While methods for running PFGE have been significantly improved since its invention, the method for preparing chromosomal DNA itself has remained essentially unchanged. This limits the applicability of PFGE, especially when analyses require many samples. We have streamlined sample preparation for routine applications of PFGE through the use of deep-well 48-well plates. Besides saving time, our protocol has the added advantage of reducing the volume of expensive reagents. Our improved protocol enables us to reduce throughput time and simplify the procedure, facilitating wider application of PFGE-based analyses in the laboratory

    The influence of resistance training on neuromuscular function in middle-aged and older adults: a systematic review and meta-analysis of randomised controlled trials.

    Background Deterioration of neuromuscular function is a major mechanism of age-related strength loss. Resistance training (RT) improves muscle strength and mass. However, the effects of RT on neuromuscular adaptations in middle-aged and older adults are unclear. Methods Randomised controlled RT interventions (≄2 weeks) involving adults aged ≄50 years were identified. Primary outcome measures were voluntary activation (VA), electromyographic (EMG) activity during maximal voluntary contraction (MVC), and antagonist coactivation. Data were pooled using a weighted random-effect model. Sub-analyses were conducted by muscle or muscle group and health status of participants. Sensitivity analysis was based on study quality. P < 0.05 indicated statistical significance. Results Twenty-seven studies were included. An effect was found for VA (standardised mean difference [SMD] 0.54, 0.01 to 1.07, P = 0.04), This result remained significant following sensitivity analysis involving only studies that were low risk of bias. Subgroup analyses showed an effect for plantar flexor VA (SMD 1.13, 0.20 to 2.06, P = 0.02) and VA in healthy participants (SMD 1.04, 0.32 to 1.76, P = 0.004). There was no effect for EMG activity or antagonist coactivation of any muscle group (P > 0.05). Discussion Resistance training did not alter EMG activity or antagonist coactivation in older adults. Sensitivity analysis resulted in the effect for VA remaining significant, indicating that this finding was not dependent on study quality. Studies predominantly involved healthy older adults (78%), limiting the generalisability of these findings to clinical cohorts. Future research should determine the effects of RT on neuromuscular function in people with sarcopenia and age-related syndromes

    New Concepts of the Chemistry of Electric-Discharge Oxygen-Iodine Lasers

    ABSTRACT The chemistry of electric discharge driven oxygen iodine lasers (EOIL) has long been believed to have O 2 (a 1 Δ g ) as the sole energy carrier for excitation of the lasing state I( 2 P 1/2 ), and O( 3 P) as the primary quencher of this state. In many sets of experimental measurements over a wide range of conditions, we have observed persistent evidence to the contrary. In this paper, we review our experimental data base in both room-temperature discharge-flow measurements and EOIL reactor results, in comparison to model predictions and kinetics analysis, to identify the missing production and loss terms in the EOIL reaction mechanism. The analysis points to a significantly higher level of understanding of this energetic chemical system, which can support advanced concepts in power scaling investigations

    Hypoxia-Mediated ATF4 Induction Promotes Survival in Detached Conditions in Metastatic Murine Mammary Cancer Cells

    Regions of hypoxia are common in solid tumors and drive changes in gene expression that increase risk of cancer metastasis. Tumor cells must respond to the stress of hypoxia by activating genes to modify cell metabolism and antioxidant response to improve survival. The goal of the current study was to determine the effect of hypoxia on cell metabolism and markers of oxidative stress in metastatic (metM-Wntlung) compared with nonmetastatic (M-Wnt) murine mammary cancer cell lines. We show that hypoxia induced a greater suppression of glutamine to glutamate conversion in metastatic cells (13% in metastatic cells compared to 7% in nonmetastatic cells). We also show that hypoxia increased expression of genes involved in antioxidant response in metastatic compared to nonmetastatic cells, including glutamate cysteine ligase catalytic and modifier subunits and malic enzyme 1. Interestingly, hypoxia increased the mRNA level of the transaminase glutamic pyruvic transaminase 2 (Gpt2, 7.7-fold) only in metM-Wntlung cells. The change in Gpt2 expression was accompanied by transcriptional (4.2-fold) and translational (6.5-fold) induction of the integrated stress response effector protein activating transcription factor 4 (ATF4). Genetic depletion ATF4 demonstrated importance of this molecule for survival of hypoxic metastatic cells in detached conditions. These findings indicate that more aggressive, metastatic cancer cells utilize hypoxia for metabolic reprogramming and induction of antioxidant defense, including activation of ATF4, for survival in detached conditions
