    Loss of Sugar Detection by GLUT2 Affects Glucose Homeostasis in Mice

    International audienceBACKGROUND: Mammals must sense the amount of sugar available to them and respond appropriately. For many years attention has focused on intracellular glucose sensing derived from glucose metabolism. Here, we studied the detection of extracellular glucose concentrations in vivo by invalidating the transduction pathway downstream from the transporter-detector GLUT2 and measured the physiological impact of this pathway. METHODOLOGY/PRINCIPAL FINDINGS: We produced mice that ubiquitously express the largest cytoplasmic loop of GLUT2, blocking glucose-mediated gene expression in vitro without affecting glucose metabolism. Impairment of GLUT2-mediated sugar detection transiently protected transgenic mice against starvation and streptozotocin-induced diabetes, suggesting that both low- and high-glucose concentrations were not detected. Transgenic mice favored lipid oxidation, and oral glucose was slowly cleared from blood due to low insulin production, despite massive urinary glucose excretion. Kidney adaptation was characterized by a lower rate of glucose reabsorption, whereas pancreatic adaptation was associated with a larger number of small islets. CONCLUSIONS/SIGNIFICANCE: Molecular invalidation of sugar sensing in GLUT2-loop transgenic mice changed multiple aspects of glucose homeostasis, highlighting by a top-down approach, the role of membrane glucose receptors as potential therapeutic targets

    Vertical profiles of arsenic and arsenic species transformations in deep-sea sediment, Nankai Trough, offshore Japan

    Abstract Concentrations of arsenic (As) and its chemical forms were determined on deep-sea sediments drilled at three sites of Nankai Trough, off the Kii Peninsula, Japan. Those sediments were analyzed to document the behavior of As in relation to methane hydrate formation and the deep biosphere. The analytical results showed the total As concentration of interstitial water (IW) and squeezed cake (SC) ranged from 0.9 to 380 ppb and from 3 to 14 ppm (average, 6.4 ppm), respectively. The sediments from Site C0002, of which sediment column was the longest down to 2200 m below the seafloor (mbsf) among the studied three drilling sites, were analyzed for the host phase transformation of As. The total concentration of As of IW and SC from 200 to 500 mbsf, where methane hydrate zone was included, was higher than those from the uppermost 200 m. Concentration of As was ultimately controlled by pH. Also, organoarsenicals, such as methylarsonic acid (MMA) and arsenobetaine (AsB), were detected in the sediment column, implying that these organoarsenicals appeared in relation to the in situ microbial activities. These observations suggest that As becomes mobilized directly or indirectly as a result of microbial activity in deep-sea sediments

    Postoperative complications:an observational study of trends in the United States from 2012 to 2018

    BACKGROUND: Postoperative complications continue to constitute a major issue for both the healthcare system and the individual patient and are associated with inferior outcomes and higher healthcare costs. The objective of this study was to evaluate the trends of postoperative complication rates over a 7-year period. METHODS: The NSQIP datasets from 2012 to 2018 were used to assess 30-day complication incidence rates including mortality rate following surgical procedures within ten surgical subspecialties. Multivariable logistic regression was used to associate complication rates with dataset year, while adjusting for relevant confounders. RESULTS: A total of 5,880,829 patients undergoing major surgery were included. Particularly the incidence rates of four complications were found to be decreasing: superficial SSI (1.9 to 1.3%), deep SSI (0.6 to 0.4%), urinary tract infection (1.6 to 1.2%) and patient unplanned return to the operating room (3.1 to 2.7%). Incidence rate for organ/space SSI exhibited an increase (1.1 to 1.5%). When adjusted, regression analyses indicated decreased odds ratios (OR) through the study period years for particularly deep SSI OR 0.92 [0.92–0.93], superficial SSI OR 0.94 [0.94–0.94] and acute renal failure OR 0.96 [0.95–0.96] as the predictor variable (study year) increased (p < 0.01). However, OR’s for organ/space SSI 1.05 [1.05–1.06], myocardial infarction 1.01 [1.01–1.02] and sepsis 1.01 [1.01–1.02] increased slightly over time (all p < 0.01). CONCLUSIONS: Incidence rates for the complications exhibited a stable trend over the study period, with minor in or decreases observed.     Geochemical distribution and fate of arsenic in water and sediments of rivers from the Hokusetsu area, Japan

    Study region: Hokusetsu Region in Osaka, Japan. Study focus: The As contamination was investigated through the geochemical mapping and analysis of river waters and bed sediments over an area of 440 km2. Sulfur from sulfides in rocks and dissolved sulfates in water were compared via isotopic analysis to assess their origin and the subsequently released As. The fate of As (transport, binding on settling particles) was evaluated through the total and dissolved fractions of As and trace metals Fe, Mn and Al in river waters. New hydrological insights for the region: The geochemical mapping showed that As in river water exceeded the maximum limit concentration of 10 ppb in several places. The highest As levels (waters and sediments) correlated well with the surface geologies, concentrating in a halo around granitic intrusion and nearby faults. The isotopic analysis of sulfur revealed the occurrence of two kinds of sulfide mineralizations responsible for As contamination: one from Late Paleozoic submarine volcanism in sedimentary rocks, and one from Late Cretaceous igneous activities in contact-metamorphosed rocks disseminated with sulfides. The transport of As along river courses occurred mainly as a dissolved species rather than absorbed on Fe/Mn/Al particles, signifying the least role of iron oxy-hydroxides in As adsorption
