A summary is presented of results concerning design approaches, estimated costs (ROM), critical technology, associated ground and orbital test programs with emphasis on dc to rf conversion, transmitting antenna, phase control, mechanical systems, flight operations, ground power receiving-rectifying antenna with systems analysis, and evaluation. Recommendations for early further in-depth studies complementing the technology program are included

    Introduction, organization, analyses, conclusions, and recommendations for each of the spaceborne subsystems are presented. Environmental effects - propagation analyses are presented with appendices covering radio wave diffraction by random ionospheric irregularities, self-focusing plasma instabilities and ohmic heating of the D-region. Analyses of dc to rf conversion subsystems and system considerations for both the amplitron and the klystron are included with appendices for the klystron covering cavity circuit calculations, output power of the solenoid-focused klystron, thermal control system, and confined flow focusing of a relativistic beam. The photovoltaic power source characteristics are discussed as they apply to interfacing with the power distribution flow paths, magnetic field interaction, dc to rf converter protection, power distribution including estimates for the power budget, weights, and costs. Analyses for the transmitting antenna consider the aperture illumination and size, with associated efficiencies and ground power distributions. Analyses of subarray types and dimensions, attitude error, flatness, phase error, subarray layout, frequency tolerance, attenuation, waveguide dimensional tolerances, mechanical including thermal considerations are included. Implications associated with transportation, assembly and packaging, attitude control and alignment are discussed.     Analysis for co-occurring sequence features identifies link between common synonymous variant and an early-terminated NPC1 isoform

    Direct assessment of allelic phase for DNA and RNA features of diploid genomes has been challenging for Sanger sequencing, due to its allele-conflating base-calling signal. Massively parallel sequencing technologies are based on the generation of a continuous copy of a single strand sequence segments, thus preserving the allelic relation between the features of the original molecules. We have performed a transcriptome-wide search for co-occurrence of variant nucleotides and exon-intron boundaries positioned within the length of a single sequencing read. Analysis of 75 human transcriptomes from retinal pigment epithelia (RPE), glioblastoma, low-grade brain tumor, breast cancer and colon cancer, have identified an association between the synonymous variant rs1140458 and an early-terminated NPC1 isoform lacking exons 19–25. Higher proportion of molecules bearing the variant nucleotide (versus the reference) incorporates the intron (P \u3c0.0001), which turns the last codon of exon 18 into a stop codon. The significance is highest in RPE cells (P = 3.88 × 10−12). NPC1 protein is involved in the control of the cholesterol trafficking. NPC1 mutations lead, in an autosomal recessive manner, to the neurological disorder Niemann-Pick syndrome type C (NP-C), and, ablation of NPC1 causes age-progressive retinal degeneration in mice and drosophila. The vast majority of the NP-C causative variants consist of missense/nonsense substitutions, small indels, and, intronic splice variants. Rs1140458 is a common exonic synonymous substitution that has never been linked to alternative splicing or pathogenicity. Our analysis suggests that rs1140458 may affect the levels of the functional NPC1 protein, and to contribute to some of the cholesterol-implicated cellular phenotype

    Severe Brain Injury, Disability, and the Law: Achieving Justice for a Marginalized Population

    Thousands of persons with severe brain injury who are minimally conscious or locked in are wrongly treated as if they are unconscious. Such individuals are unable to advocate for themselves and are typically segregated from society in hospitals or nursing homes. As a result, they constitute a class of persons who often lack access to adequate medical care, rehabilitation, and assistive devices that could aid them in communication and recovery. While this problem is often approached from a medical or scientific point of view, here we frame it as a legal issue amenable to legal remedies. This Article comprehensively explores and analyzes sources of federal, state, and international human rights law that can be leveraged- both in traditional and novel ways-to improve the lives and protect the rights of persons with severe brain injury. We argue that state laws may be the most promising basis for legal action to ameliorate the clinical marginalization and societal neglect faced by persons with severe brain injury, and to promote their recovery and reintegration into their communities
