    The selection of comparators for randomized controlled trials of health-related behavioral interventions: recommendations of an NIH expert panel

    Objectives: To provide recommendations for the selection of comparators for randomized controlled trials of health-related behavioral interventions. Study Design and Setting: The National Institutes of Health Office of Behavioral and Social Science Research convened an expert panel to critically review the literature on control or comparison groups for behavioral trials and to develop strategies for improving comparator choices and for resolving controversies and disagreements about comparators. Results: The panel developed a Pragmatic Model for Comparator Selection in Health-Related Behavioral Trials. The model indicates that the optimal comparator is the one that best serves the primary purpose of the trial but that the optimal comparator's limitations and barriers to its use must also be taken into account. Conclusion: We developed best practice recommendations for the selection of comparators for health-related behavioral trials. Use of the Pragmatic Model for Comparator Selection in Health-Related Behavioral Trials can improve the comparator selection process and help resolve disagreements about comparator choices

    Isoprenylcysteine-O-carboxyl methyltransferase regulates aldosterone- sensitive Na+ reabsorption

    The Xenopus laevis distal tubule epithelial cell line A6 was used as a model epithelia to study the role of isoprenylcysteine-O-carboxyl methyltransferase (pcMTase) in aldosterone-mediated stimulation of Na+ transport. Polyclonal antibodies raised against X. laevis pcMTase were immunoreactive with a 33-kDa protein in whole cell lysate. These antibodies were also reactive with a 33-kDa product from in vitro translation of the pcMTase cDNA. Aldosterone application increased pcMTase activity resulting in elevation of total protein methyl esterification in vivo, but pcMTase protein levels were not affected by steroid, suggesting that aldosterone increased activity independent of enzyme number. Inhibition of pcMTase resulted in a reduction of aldosterone-induced Na+ transport demonstrating the necessity of pcMTase-mediated transmethylation for steroid induced Na+ reabsorption. Transfection with an eukaryotic expression construct containing pcMTase cDNA increased pcMTase protein level and activity. This resulted in potentiation of the natriferic actions of aldosterone. However, overexpression did not change Na+ reabsorption in the absence of steroid, suggesting that pcMTase activity is not limiting Na+ transport in the absence of steroid, but that subsequent to aldosterone addition, pcMTase activity becomes limiting. These results suggest that a critical transmethylation is necessary for aldosterone-induction of Na+ transport. It is likely that the protein catalyzing this methylation is isoprenylcysteine-O-carboxyl methyltransferase and that aldosterone activates pcMTase without affecting transferase expression.SCOPUS: ar.jinfo:eu-repo/semantics/publishe

    Fine sediment trapping in two mangrove-fringed estuaries exposed to contrasting land-use intensity, Palau, Micronesia

    A comparative study was undertaken of the fate of fine sediment in the Ngerikiil and Ngerdorch mangrovefringed\ud estuaries in Babeldaob Island, Palau, Micronesia, in 2002. The mangroves comprised 3.8% of each catchment area, and in both systems, they trapped about 30% of the riverine sediment. Mangroves are important buffers protecting fringing coral reefs from excessive sedimentation. The sediment yield was significantly higher in the Ngerikiil River catchment (150 tons km2 yr1) that has been extensively cleared and farmed, than in Ngerdorch River catchment (1.9 tons km2 yr1) that was still relatively pristine during the study period
