A distinct sortase SrtB anchors and processes a streptococcal adhesin AbpA with a novel structural property.
Surface display of proteins by sortases in Gram-positive bacteria is crucial for bacterial fitness and virulence. We found a unique gene locus encoding an amylase-binding adhesin AbpA and a sortase B in oral streptococci. AbpA possesses a new distinct C-terminal cell wall sorting signal. We demonstrated that this C-terminal motif is required for anchoring AbpA to cell wall. In vitro and in vivo studies revealed that SrtB has dual functions, anchoring AbpA to the cell wall and processing AbpA into a ladder profile. Solution structure of AbpA determined by NMR reveals a novel structure comprising a small globular α/β domain and an extended coiled-coil heliacal domain. Structural and biochemical studies identified key residues that are crucial for amylase binding. Taken together, our studies document a unique sortase/adhesion substrate system in streptococci adapted to the oral environment rich in salivary amylase
A wideband F-shaped patch antenna for S-band CubeSats communications
A wideband S-band F-shaped patch antenna is proposed for CubeSats communications. To broaden bandwidth, it uses two arms with different lengths to generate a second resonant frequency. The effect of the arm length and width on the return loss, resonant frequency and impedance bandwidth on a 3U CubeSat is studied. The simulation results show that the antenna achieves a wideband of 1121 MHz (1.606-2.727 GHz) with a return loss below −10 dB over the entire frequency band from 1.606 to 2.727 GHz. The antenna has a high gain of 8.51 dB and a small return loss of −32.85 dB at 2.45 GHz

Ras/MAPK pathway signaling is a major participant in neurodevelopment, and evidence suggests that BRAF, a key Ras signal mediator, influences human behavior. We studied the role of the mutation BRAFQ257R, the most common cause of cardiofaciocutaneous syndrome (CFC), in an induced pluripotent stem cell (iPSC)-derived model of human neurodevelopment. In iPSC-derived neuronal cultures from CFC subjects, we observed decreased p-AKT and p-ERK1/2 compared to controls, as well as a depleted neural progenitor pool and rapid neuronal maturation. Pharmacological PI3K/AKT pathway manipulation recapitulated cellular phenotypes in control cells and attenuated them in CFC cells. CFC cultures displayed altered cellular subtype ratios and increased intrinsic excitability. Moreover, in CFC cells, Ras/MAPK pathway activation and morphological abnormalities exhibited cell subtype-specific differences. Our results highlight the importance of exploring specific cellular subtypes and of using iPSC models to reveal relevant human-specific neurodevelopmental events
GPT vs Human for Scientific Reviews: A Dual Source Review on Applications of ChatGPT in Science
The new polymath Large Language Models (LLMs) can speed-up greatly scientific
reviews, possibly using more unbiased quantitative metrics, facilitating
cross-disciplinary connections, and identifying emerging trends and research
gaps by analyzing large volumes of data. However, at the present time, they
lack the required deep understanding of complex methodologies, they have
difficulty in evaluating innovative claims, and they are unable to assess
ethical issues and conflicts of interest. Herein, we consider 13 GPT-related
papers across different scientific domains, reviewed by a human reviewer and
SciSpace, a large language model, with the reviews evaluated by three distinct
types of evaluators, namely GPT-3.5, a crowd panel, and GPT-4. We found that
50% of SciSpace's responses to objective questions align with those of a human
reviewer, with GPT-4 (informed evaluator) often rating the human reviewer
higher in accuracy, and SciSpace higher in structure, clarity, and
completeness. In subjective questions, the uninformed evaluators (GPT-3.5 and
crowd panel) showed varying preferences between SciSpace and human responses,
with the crowd panel showing a preference for the human responses. However,
GPT-4 rated them equally in accuracy and structure but favored SciSpace for
completeness
Verifying Policy Enforcers
Policy enforcers are sophisticated runtime components that can prevent
failures by enforcing the correct behavior of the software. While a single
enforcer can be easily designed focusing only on the behavior of the
application that must be monitored, the effect of multiple enforcers that
enforce different policies might be hard to predict. So far, mechanisms to
resolve interferences between enforcers have been based on priority mechanisms
and heuristics. Although these methods provide a mechanism to take decisions
when multiple enforcers try to affect the execution at a same time, they do not
guarantee the lack of interference on the global behavior of the system. In
this paper we present a verification strategy that can be exploited to discover
interferences between sets of enforcers and thus safely identify a-priori the
enforcers that can co-exist at run-time. In our evaluation, we experimented our
verification method with several policy enforcers for Android and discovered

Multispectral autofluorescence characteristics of reproductive aging in old and young mouse oocytes
Increasing age has a major detrimental impact on female fertility, which, with an ageing population, has major sociological implications. This impact is primarily mediated through deteriorating quality of the oocyte. Deteriorating oocyte quality with biological age is the greatest rate-limiting factor to female fertility. Here we have used label-free, non-invasive multi-spectral imaging to identify unique autofluorescence profiles of oocytes from young and aged animals. Discriminant analysis demonstrated that young oocytes have a distinct autofluorescent profile which accurately distinguishes them from aged oocytes. We recently showed that treatment with the nicotinamide adenine dinucleotide (NAD+) precursor nicotinamide mononucleotide (NMN) restored oocyte quality and fertility in aged animals, and when our analysis was applied to oocytes from aged animals treated with NMN, 85% of these oocytes were classified as having the autofluorescent signature of young animals. Spectral unmixing using the Robust Dependent Component Analysis (RoDECA) algorithm demonstrated that NMN treatment altered the metabolic profile of oocytes, increasing free NAD(P)H, protein bound NAD(P)H, redox ratio and the ratio of bound to free NAD(P)H. The frequency of oocytes with simultaneously high NAD(P)H and flavin content was also significantly increased in mice treated with NMN. Young and Aged + NMN oocytes had a smoother spectral distribution, with the distribution of NAD(P)H in young oocytes specifically differing from that of aged oocytes. Identifying the multispectral profile of oocyte autofluorescence during aging could have utility as a non-invasive and sensitive measure of oocyte quality
Effects of stitching on delamination of satin weave carbon-epoxy laminates under mode I, mode II and mixed-mode I/II loadings
The objective of the present study is to characterize the effect of modified chain stitching on the delamination growth under mixed-mode I/II loading conditions. Delamination toughness under mode I is experimentally determined, for unstitched and stitched laminates, by using untabbed and tabbed double cantilever beam (TDCB) tests. The effect of the reinforcing tabs on mode I toughness is investigated. Stitching improves the energy release rate (ERR) up to 4 times in mode I. Mode II delamination toughness is evaluated in end-notched flexure (ENF) tests. Different geometries of stitched specimens are tested. Crack propagation occurs without any failure of stitching yarns. The final crack length attains the mid-span or it stops before and the specimen breaks in bending. The ERR is initially low and gradually increases with crack length to very high values. The mixedmode delamination behaviour is investigated using a mixed-mode bending (MMB) test. For unstitched specimens, a simple mixed-mode criterion is identified. For stitched specimens, stitching yarns do not break during 25% of mode I ratio tests and the ERR increase is relatively small compared to unstitched values. For 70% and 50% of mode I ratios, failures of yarns are observed during crack propagation and tests are able to capture correctly the effect of the stitching: it clearly improves the ERR for these two mixed modes, as much as threefold
Multi-objective Robust Strategy Synthesis for Interval Markov Decision Processes
Interval Markov decision processes (IMDPs) generalise classical MDPs by
having interval-valued transition probabilities. They provide a powerful
modelling tool for probabilistic systems with an additional variation or
uncertainty that prevents the knowledge of the exact transition probabilities.
In this paper, we consider the problem of multi-objective robust strategy
synthesis for interval MDPs, where the aim is to find a robust strategy that
guarantees the satisfaction of multiple properties at the same time in face of
the transition probability uncertainty. We first show that this problem is
PSPACE-hard. Then, we provide a value iteration-based decision algorithm to
approximate the Pareto set of achievable points. We finally demonstrate the
practical effectiveness of our proposed approaches by applying them on several
