    Soil Microbial Responses to Elevated CO2 and O3 in a Nitrogen-Aggrading Agroecosystem

    Climate change factors such as elevated atmospheric carbon dioxide (CO2) and ozone (O3) can exert significant impacts on soil microbes and the ecosystem level processes they mediate. However, the underlying mechanisms by which soil microbes respond to these environmental changes remain poorly understood. The prevailing hypothesis, which states that CO2- or O3-induced changes in carbon (C) availability dominate microbial responses, is primarily based on results from nitrogen (N)-limiting forests and grasslands. It remains largely unexplored how soil microbes respond to elevated CO2 and O3 in N-rich or N-aggrading systems, which severely hinders our ability to predict the long-term soil C dynamics in agroecosystems. Using a long-term field study conducted in a no-till wheat-soybean rotation system with open-top chambers, we showed that elevated CO2 but not O3 had a potent influence on soil microbes. Elevated CO2 (1.5×ambient) significantly increased, while O3 (1.4×ambient) reduced, aboveground (and presumably belowground) plant residue C and N inputs to soil. However, only elevated CO2 significantly affected soil microbial biomass, activities (namely heterotrophic respiration) and community composition. The enhancement of microbial biomass and activities by elevated CO2 largely occurred in the third and fourth years of the experiment and coincided with increased soil N availability, likely due to CO2-stimulation of symbiotic N2 fixation in soybean. Fungal biomass and the fungi∶bacteria ratio decreased under both ambient and elevated CO2 by the third year and also coincided with increased soil N availability; but they were significantly higher under elevated than ambient CO2. These results suggest that more attention should be directed towards assessing the impact of N availability on microbial activities and decomposition in projections of soil organic C balance in N-rich systems under future CO2 scenarios

    Instrumentation for the measurement of autofluorescence in the human skin

    A setup to measure skin autofluorescence was developed to assess accumulation of advanced glycation endproducts (AGE) in patients noninvasively. The method applies direct blacklight tube illumination of the skin of the lower arm, and spectrometry. The setup displays skin autofluorescence (AF) as a ratio of mean intensities detected from the skin between 420-600 nm and 300-420 nm, respectively. In an early clinical application in 46 and control subjects matched for age and gender, AF was significantly increased in the patients (p = 0.015), and highly correlated with skin AGE's that were determined from skin biopsies in both groups. A large follow-up study on type 2 diabetes mellitus, ongoing since 2001 with more than 1000 subjects, aims to assess the value of the instrument in predicting chronic complications of diabetes. At baseline, a relation with age, glycemic status and with complications present was found. In a study in patients with end stage renal disease on dialysis AF was a strong and independent predictor of total and cardiovascular mortality. A commercial version of this AGE-reader is now under development and becomes available early 2005 (DiagnOptics B.V., Groningen, The Netherlands).One of the remaining questions, that will be answered by measuring so-called Exciation-Emission Matrices (EEM's) of the skin tissue in vivo, is whether a more selective choice of wavelengths is more strongly related to clinical characteristics. An experimental instrument to measure these EEM's was, therefore, developed as well. Clinical measurements are underway of EEM's in patient groups with diabetes mellitus and in healthy volunteers.</p

    Problem Based Learning (PBL) for Engineering Education in India: Need and Recommendations

    Historical footprint of problem based learning (PBL) could be traced back at McMasters University, Canada 1968. Since then, PBL is enriched with development in the literature and multiple PBL models across the world. PBL, has gained importance in modern era of higher education as it has been implemented in many universities across the world in higher education. Although, world is weighing up for PBL and its positive outcomes, it could not extend much in Indian higher education. An objective of this manuscript is to give an overview of PBL across the world and in India. In addition to this, Indian higher education landscape and scope of PBL to Indian engineering education is discussed. Article ends with some recommendations to ensure growth of PBL in India.</p
