    The orthodontic-oral surgery interface. Part one: A service evaluation and overview of the diagnosis and management of common anomalies

    Introduction: Complex dental anomalies often require multidisciplinary management in secondary care. These anomalies may present to general dental practitioners in the first instance, therefore it is important that all clinicians are aware of such anomalies and their management. / Aims: This service evaluation aimed to profile the dental anomalies presenting to the multidisciplinary joint dento-alveolar clinic at a district general hospital and to outline the management of the common anomalies. / Method: This two-part series details a service evaluation and profiles the dental anomalies presenting to the joint dento-alveolar clinic at a district general hospital. In addition, the features and management of the common dental anomalies are outlined. / Results: Fifty-four percent of patients were female, with a mean age of 17 years. The most common anomalies were impacted canines (49%), followed by supernumerary teeth (16%) and impacted premolars (10%). Other anomalies accounted for only a small percentage of referrals. / Conclusion: A wide range of anomalies presented to the joint dento-alveolar clinic. It is important for all clinicians to be aware of such anomalies, their diagnosis and management. Part one and two of this article therefore provide an overview of the common anomalies and highlight their management

    Characterization of Leishmania donovani MCM4: Expression Patterns and Interaction with PCNA

    Events leading to origin firing and fork elongation in eukaryotes involve several proteins which are mostly conserved across the various eukaryotic species. Nuclear DNA replication in trypanosomatids has thus far remained a largely uninvestigated area. While several eukaryotic replication protein orthologs have been annotated, many are missing, suggesting that novel replication mechanisms may apply in this group of organisms. Here, we characterize the expression of Leishmania donovani MCM4, and find that while it broadly resembles other eukaryotes, noteworthy differences exist. MCM4 is constitutively nuclear, signifying that, unlike what is seen in S.cerevisiae, varying subcellular localization of MCM4 is not a mode of replication regulation in Leishmania. Overexpression of MCM4 in Leishmania promastigotes causes progress through S phase faster than usual, implicating a role for MCM4 in the modulation of cell cycle progression. We find for the first time in eukaryotes, an interaction between any of the proteins of the MCM2-7 (MCM4) and PCNA. MCM4 colocalizes with PCNA in S phase cells, in keeping with the MCM2-7 complex being involved not only in replication initiation, but fork elongation as well. Analysis of a LdMCM4 mutant indicates that MCM4 interacts with PCNA via the PIP box motif of MCM4 - perhaps as an integral component of the MCM2-7 complex, although we have no direct evidence that MCM4 harboring a PIP box mutation can still functionally associate with the other members of the MCM2-7 complex- and the PIP box motif is important for cell survival and viability. In Leishmania, MCM4 may possibly help in recruiting PCNA to chromatin, a role assigned to MCM10 in other eukaryotes

    Medical Grade vs Off-the-Shelf Color Displays: Influence on Observer Performance and Visual Search

    The goal of this study was to compare diagnostic accuracy of radiologists viewing clinical images on a top-of-the-line medical-grade vs a top-of-the-line commercial off-the-shelf (COTS) color display with the luminance values set to simulate a display that had been in use for 1 year. A set of 50 digital radiography chest images was selected for use in the study, half containing a solitary pulmonary nodule and half nodule-free. The images were displayed twice to each of six observers, once on each display. Eye position was recorded on a subset of the images. Overall, there was a statistically significant difference (F = 4.1496, p = 0.0471) between the medical-grade color display and the COTS color display in terms of receiver operating characteristic area under the curve values, with the medical-grade display yielding higher diagnostic accuracy. Total viewing time did not differ significantly, but eye position data revealed differences, suggesting better search and decision-making efficiency with the medical-grade display. Medical-grade color displays at 1 year old yield better diagnostic and search efficiency than COTS color displays and thus are recommended for primary reading if color displays are to be used

    We have recently analyzed in mouse embryo NIH3T3 fibroblasts and human fibrosarcoma HT1080 cells the molecular basis and the biological role of AR interaction with the full-length filamin A (FLNa), an actin-cross-linking protein. Here, we describe a procedure revealing the AR/FLNa complex in stromal cells. Upon physiological (10 nM) androgen stimulation of quiescent NIH3T3 cells, FLNa co-immunoprecipitates with AR and co-localizes with the receptor at intermediate actin filaments. The AR/FLNa complex specifically regulates AR extranuclear functions leading to Rac1 activation and consequent cell motility. This complex adds a new and unexpected piece to the growing evidence of the role of signalling effectors, scaffolds, and cytoskeletal proteins in the rapid androgen action and in progression of hormone-dependent cancers
