    Poly(ADP-Ribose) Polymerase 1 Accelerates Single-Strand Break Repair in Concert with Poly(ADP-Ribose) Glycohydrolase

    Single-strand breaks are the commonest lesions arising in cells, and defects in their repair are implicated in neurodegenerative disease. One of the earliest events during single-strand break repair (SSBR) is the rapid synthesis of poly(ADP-ribose) (PAR) by poly(ADP-ribose) polymerase (PARP), followed by its rapid degradation by poly(ADP-ribose) glycohydrolase (PARG). While the synthesis of poly(ADP-ribose) is important for rapid rates of chromosomal SSBR, the relative importance of poly(ADP-ribose) polymerase 1 (PARP-1) and PARP-2 and of the subsequent degradation of PAR by PARG is unclear. Here we have quantified SSBR rates in human A549 cells depleted of PARP-1, PARP-2, and PARG, both separately and in combination. We report that whereas PARP-1 is critical for rapid global rates of SSBR in human A549 cells, depletion of PARP-2 has only a minor impact, even in the presence of depleted levels of PARP-1. Moreover, we identify PARG as a novel and critical component of SSBR that accelerates this process in concert with PARP-1

    AbstractAs a decision aid for discrete multicriteria decision problems, this paper proposes a multilevel graph of alternatives to represent the ranking, to the extent that this is possible when incomplete information on weights is available under the assumption of the additive value function. To construct it, the nested decomposition of the set of alternatives is established along the lines of data envelopment analysis (DEA). A numerical example is given to illustrate a multilevel graph based on the nested decomposition and compare it with the hierarchical dominance graph based on dominance relations proposed by Park and Kim

    Probing the parameter space of HD 49933: a comparison between global and local methods

    We present two independent methods for studying the global stellar parameter space (mass M, age, initial chemical composition X_0, Z_0) of HD 49933 with seismic data. Using a local minimization and an MCMC algorithm, we obtain consistent results for the determination of the stellar properties: M = 1.1 - 1.2 M_solar, Age ~ 3.0 Gyr, Z_0 ~ 0.008. A description of the error ellipses can be defined using Singular Value Decomposition techniques, and this is validated by comparing the errors with those from the MCMC method.Comment: to be published in JPC

    The Effect of Environment on the X-Ray Emission from Early-Type Galaxies

    In order to help understand the phenomena of X-ray emission from early-type galaxies, we obtained an optically flux-limited sample of 34 early-type galaxies, observed with ROSAT. A previous analysis of this sample suggested that the most X-ray luminous galaxies were in rich environments. Here we investigate environmental influences quantitatively, and find a positive correlation between L_B/L_X and the local galaxy density. We suggest that this correlation occurs because the X-ray luminosity is enhanced either through accretion of the intergalactic gas or because the ambient medium stifles galactic winds. When the ambient medium is unimportant, partial or global galactic winds can occur, reducing L_B/L_X. These effects lead to the large observed dispersion in L_X at fixed L_B. We argue that the transition from global winds to partial winds is one of the principle reasons for the steep relationship between L_X and L_B. We discuss details of the data reduction not previously presented, and examine the dependence of L_X on the choice of outer source radius and background location. Effects of Malmquist bias are shown not to be important for the issues addressed.     The survival and destruction of X-ray coronae of early-type galaxies in the rich cluster environments: a case study of Abell 1367

    A new Chandra observation of the northwest region of the galaxy cluster A1367 reveals four cool galaxy coronae (0.4 - 1.0 keV) embedded in the hot intracluster medium (ICM) (5 - 6 keV). While the large coronae of NGC 3842 and NGC 3837 appear symmetric and relaxed, the galaxy coronae of the \lsim L* galaxies (NGC 3841 and CGCG 97090) are disturbed and being stripped. Massive galaxies, with dense cooling cores, are better able to resist ram pressure stripping and survive in rich environments than \lsim L* galaxies whose galactic coronae are much less dense. The survival of these cool coronae implies that thermal conduction from the hot surrounding ICM has to be suppressed by a factor of at least 60, at the corona boundary. Within the galaxy coronae of NGC 3842 and NGC 3837, stellar mass loss or heat conduction with the Spitzer value may be sufficient to balance radiative cooling. Energy deposition at the ends of collimated jets may heat the outer coronae, but allow the survival of a small, dense gas core (e.g., NGC 3842 in A1367 and NGC 4874 in Coma).     We use the Quantum Hypernetted Chain Approximation (QHNC) to calculate the ion-ion and electron-ion correlations for liquid metallic Li, Be, Na, Mg, Al, K, Ca, and Ga. We discuss trends in electron-ion structure factors and radial distribution functions, and also calculate the free-atom and metallic-atom form-factors, focusing on how bonding effects affect the interpretation of X-ray scattering experiments, especially experimental measurements of the ion-ion structure factor in the liquid metallic phase.Comment: RevTeX, 19 pages, 7 figure

    Deficiency in clonogenic endometrial mesenchymal stem cells in obese women with reproductive failure – a pilot study

    The mechanisms of obesity associated reproductive complications remain poorly understood. Endometrial mesenchymal stem-cells are critical for cyclic renewal and uterine function. Recently, W5C5+ cells, with high clonogenicity, capable of producing endometrial stroma in vivo, have been described. We sought to investigate the abundance and cloning efficiency of W5C5+ and W5C5− endometrial cells in relation to Body Mass Index, age and reproductive outcome. Design W5C5+ and W5C5− cells were purified from mid-luteal endometrial biopsies (n = 54) by magnetic bead separation and subjected to in vitro colony-forming assays. Results First trimester pregnancy losses were significantly higher in obese subjects (n = 12) compared to overweight (n = 20) and subjects with normal Body Mass Index (n = 22) (P0.05). Conclusions Our observations suggest that the regenerative capacity and plasticity of the endometrium of obese women is suboptimal, which in turn may account for the increased risk of reproductive complications associated with obesity

    Statistical mechanical analysis of the linear vector channel in digital communication

    A statistical mechanical framework to analyze linear vector channel models in digital wireless communication is proposed for a large system. The framework is a generalization of that proposed for code-division multiple-access systems in Europhys. Lett. 76 (2006) 1193 and enables the analysis of the system in which the elements of the channel transfer matrix are statistically correlated with each other. The significance of the proposed scheme is demonstrated by assessing the performance of an existing model of multi-input multi-output communication systems.Comment: 15 pages, 2 figure
