    Analisa Tegangan Dan Regangan Pada Perkerasan Porus Dengan Skala Semi Lapangan Dan Software Ansys

    Perkerasan porus merupakan perkerasan yang mengijinkan air masuk kedalam lapisan perkerasan untuk dialirkan kedalam tanah karena memiliki porositas dan permeabilitas yang tinggi. Namun, hal ini menyebabkan ikatan agregat perkerasan lebih lemah sehingga kekuatannya pun berkurang. Maka, perlu dilakukan penelitian terkait perilaku perkerasan porus yaitu tegangan dan regangan aspal porus dalam menerima beban. Untuk itu, penulis mengadakan penelitian dengan tujuan mengukur regangan pada struktur perkerasan porus skala semi lapangan yang diberi beban dinamis dan beban statis, mengukur tegangan dan regangan struktur perkerasan porus dengan ANSYS dan membandingkan hasil regangan antara pengukuran skala semi lapangan dengan pengukuran menggunakan software ANSYS. Pada penelitian ini, digunakan kriteria perkerasan yang menyerupai kondisi sesungguhnya baik pada beban, material, maupun ketebalannya. Beban yang digunakan merupakan beban satu roda sebesar 17 kg/cm2. Material subbase berupa batu pecah dengan gradasi kelas B, base berupa batu pecah gradasi kelas A, dan surface berupa aspal porus dengan tambahan 8% additive gilsonite. Metodologi penelitian pada pengukuran regangan perkerasan porus skala semi lapangan yaitu regangan dan tegangan yang diukur hanya pada bagian bawah aspal porus. Pertama, perkerasan porus diberikan beban dinamis lalu nilai regangan dibaca setiap 50 kali lintasan hingga 1000 lintasan. Selanjutnya, regangan juga diukur menggunakan beban statis. Sedangkan untuk Finite Element Method dilakukan pengukuran tegangan dan regangan dengan beban statis. Regangan dari pengukuran dengan skala semi lapangan menggunakan beban dinamis, pengukuran dengan skala semi lapangan menggunakan beban statis, dan regangan dari ANSYS dibandingkan satu sama lain. Hasil dari penelitian menunjukkan bahwa regangan yang terjadi pada bagian bawah aspal adalah regangan tarik. Namun, pemberian beban dinamis dan beban statis menyebabkan nilai regangan yang berbeda. Pada analisis ANSYS, tegangan berupa tarik dimana nilai maksimum berada tepat dibawah roda dan akan semakin berkurang seiring dengan jaraknya terhadap beban roda. Regangan dari analisis ANSYS dengan beban statis mendapat nilai yang berbeda dengan hasil regangan dari pengukuran skala semi lapangan yang diberi beban statis dan beban dinamis karena pengaruh perlakuan beban dan karakteristik material.

    Patient engagement with antibiotic messaging in secondary care: a qualitative feasibility study of the ‘review & revise’ experience

    Background: We aimed to investigate and optimise the acceptability and usefulness of a patient leaflet about antibiotic prescribing decisions made during hospitalisation, and to explore individual patient experiences and preferences regarding the process of antibiotic prescription ‘review & revise’ which is a key strategy to minimise antibiotic overuse in hospitals. Methods: In this qualitative study, run within the feasibility study of a large, cluster-randomised stepped wedge trial of 36 hospital organisations, a series of semi-structured, think-aloud telephone interviews were conducted and data were analysed using thematic analysis. Fifteen adult patients who had experienced a recent acute medical hospital admission during which they had been prescribed antimicrobials and offered a patient leaflet about antibiotic prescribing were recruited to the study. Results: Participants reacted positively to the leaflet, reporting that it was both an accessible and important source of information which struck the appropriate balance between informing and reassuring. Participants all valued open communication with clinicians, and were keen to be involved in antibiotic prescribing decisions, with individuals reporting positive experiences regarding antibiotic prescription changes or stopping. Many participants had prior experience or knowledge of antibiotics and resistance, and generally welcomed efforts to reduce antibiotic usage. Overall, there was a feeling that healthcare professionals (HCPs) are trusted experts providing the most appropriate treatment for individual patient conditions. Conclusions: This study offers novel insights into how patients within secondary care are likely to respond to messages advocating a reduction in the use of antibiotics through the ‘review & revise’ approach. Due to the level of trust that patients place in their care provider, encouraging HCPs within secondary care to engage patients with greater communication and information provision could provide great advantages in the drive to reduce antibiotic use. It may also be beneficial for HCPs to view patient experiences as cumulative events that have the potential to impact future behaviour around antibiotic use. Finally, pre-testing messages about antibiotic prescribing and resistance is vital to dispelling any misconceptions either around effectiveness of treatment for patients, or perceptions of how messages may be received

    This paper details a comprehensive study undertaken by the National Renewable Energy Laboratory, Electric Power Research Institute, and the University of Colorado to understand how the contribution of wind power providing active power control (APC) can benefit the total power system economics, increase revenue streams, improve the reliability and security of the power system, and provide superior and efficient response while reducing any structural and loading impacts that may reduce the life of the wind turbine or its components. The study includes power system simulations, control simulations, and actual field tests using turbines at NREL's National Wind Technology Center (NWTC). The study focuses on synthetic inertial control, primary frequency control, and automatic generation control, and analyzes timeframes ranging from milliseconds to minutes to the lifetime of wind turbines, locational scope ranging from components of turbines to large wind plants to entire synchronous interconnections, and additional topics ranging from economics to power system engineering to control design

    DIFFERENT TOPICAL FORMULATIONS OF KETOROLAC TROMETHAMINE FOR ANTI-INFLAMMATORY APPLICATION AND CLINICAL EFFICACY

    Ketorolac tromethamine (KT) is considered as a member of NSAIDs that used in treatment of rheumatoid arthritis. The main problems associated with the frequent administration of KT orally could be overcome by alternative routes as topical application. KT was formulated in different topical formulations such as gels, emulgels and creams. Sodium carboxymethylcellulose, carbopol 934 and pluronic F127 were used as polymers. In vitro permeation study through rat skin was carried out. The effect of different KT concentrations and the effect of skin penetration enhancer on the amount of KT permeated were investigated. Anti-inflammatory activity using commercial piroxicam gel for comparison was evaluated. The effectiveness and tolerability of the selected KT gel and piroxicam gel in osteoarthritis patients was also studied. The results obtained showed that, the flux of the drug increased with increasing its initial concentration. Using sodium lauryl sulfate as enhancer resulted in an improvement of KT permeation through rat skin. All formulations had the potential for local applications of KT as anti-inflammatory drug as compared to the control group. There was no significant difference in the efficacy between the selected KT gel and piroxicam gel for osteoarthritis patients. So KT gel may be used as another therapeutic option for the treatment of patients with osteoarthritis

    The Assembled Genome of the Stroke-Prone Spontaneously Hypertensive Rat

    BACKGROUND: We report the creation and evaluation of a de novo assembly of the genome of the spontaneously hypertensive rat, the most widely used model of human cardiovascular disease. METHODS: The genome is assembled from long read sequencing (PacBio HiFi and continuous long read data [CLR]) and scaffolded with long-range structural information obtained from Bionano optical maps and proximity ligation sequencing proximity analysis of the genome. The genome assembly was polished with Illumina short reads. Completeness of the assembly was investigated using Benchmarking Universal Single Copy Orthologs analysis. The genome assembly was also evaluated with the rat reference gene set, using NCBI automated protocols. We also generated orthogonal single molecule transcript sequence reads (Iso-Seq) from 8 tissues and used them to validate the coding assembly, to annotate the assembly with RNA transcripts representing unique full length transcript isoforms for each gene and to determine whether divergences between RefSeq sequences and the assembly were attributable to assembly errors or polymorphisms. RESULTS: The assembly analysis indicates that this assembly is comparable in contiguity and completeness to the current rat reference assembly, while the use of HiFi sequencing yields an assembly that is more correct at the single base level. Synteny analysis was performed to uncover the extent of synteny and the presence and distribution of chromosomal rearrangements between the reference and this assembly. CONCLUSION: The resulting genome assembly is reference quality and captures significant structural variation

    Fanny Copeland and the geographical imagination

    Raised in Scotland, married and divorced in the English south, an adopted Slovene, Fanny Copeland (1872 – 1970) occupied the intersection of a number of complex spatial and temporal conjunctures. A Slavophile, she played a part in the formation of what subsequently became the Kingdom of Yugoslavia that emerged from the First World War. Living in Ljubljana, she facilitated the first ‘foreign visit’ (in 1932) of the newly formed Le Play Society (a precursor of the Institute of British Geographers) and guided its studies of Solčava (a then ‘remote’ Alpine valley system) which, led by Dudley Stamp and commended by Halford Mackinder, were subsequently hailed as a model for regional studies elsewhere. Arrested by the Gestapo and interned in Italy during the Second World War, she eventually returned to a socialist Yugoslavia, a celebrated figure. An accomplished musician, linguist, and mountaineer, she became an authority on (and populist for) the Julian Alps and was instrumental in the establishment of the Triglav National Park. Copeland’s role as participant observer (and protagonist) enriches our understanding of the particularities of her time and place and illuminates some inter-war relationships within G/geography, inside and outside the academy, suggesting their relative autonomy in the production of geographical knowledge

    G-quadruplex formation within the promoter of the KRAS proto-oncogene and its effect on transcription

    In human and mouse, the promoter of the KRAS gene contains a nuclease hypersensitive polypurine–polypyrimidine element (NHPPE) that is essential for transcription. An interesting feature of the polypurine G-rich strand of NHPPE is its ability to assume an unusual DNA structure that, according to circular dichroism (CD) and DMS footprinting experiments, is attributed to an intramolecular parallel G-quadruplex, consisting of three G-tetrads and three loops. The human and mouse KRAS NHPPE G-rich strands display melting temperature of 64 and 73°C, respectively, as well as a K(+)-dependent capacity to arrest DNA polymerase. Photocleavage and CD experiments showed that the cationic porphyrin TMPyP4 stacks to the external G-tetrads of the KRAS quadruplexes, increasing the T(m) by ∼20°C. These findings raise the intriguing question that the G-quadruplex formed within the NHPPE of KRAS may be involved in the regulation of transcription. Indeed, transfection experiments showed that the activity of the mouse KRAS promoter is reduced to 20% of control, in the presence of the quadruplex-stabilizing TMPyP4. In addition, we found that G-rich oligonucleotides mimicking the KRAS quadruplex, but not the corresponding 4-base mutant sequences or oligonucleotides forming quadruplexes with different structures, competed with the NHPPE duplex for binding to nuclear proteins. When vector pKRS-413, containing CAT driven by the mouse KRAS promoter, and KRAS quadruplex oligonucleotides were co-transfected in 293 cells, the expression of CAT was found to be downregulated to 40% of the control. On the basis of these data, we propose that the NHPPE of KRAS exists in equilibrium between a double-stranded form favouring transcription and a folded quadruplex form, which instead inhibits transcription. Such a mechanism, which is probably adopted by other growth-related genes, provides useful hints for the rational design of anticancer drugs against the KRAS oncogene

    Neuroprotective Potential of Biphalin, Multireceptor Opioid Peptide, Against Excitotoxic Injury in Hippocampal Organotypic Culture

    Biphalin is a dimeric opioid peptide that exhibits affinity for three types of opioid receptors (MOP, DOP and KOP). Biphalin is undergoing intensive preclinical study. It was recognized that activation of δ-opioid receptor elicits neuroprotection against brain hypoxia and ischemia. We compare the effect of biphalin and morphine and the inhibition of opioid receptors by naltrexone on survival of neurons in rat organotypic hippocampal cultures challenged with NMDA. Findings: (1) 0.025–0.1 μM biphalin reduces NMDA-induced neuronal damage; (2) biphalin neuroprotection is abolished by naltrexone; (3) reduced number of dead cells is shown even if biphalin is applied with delay after NMDA challenge
