    Understanding the mechanisms of zinc bacitracin and avilamycin on animal production: linking gut microbiota and growth performance in chickens

    Unravelling the mechanisms of how antibiotics influence growth performance through changes in gut microbiota can lead to the identification of highly productive microbiota in animal production. Here we investigated the effect of zinc bacitracin and avilamycin on growth performance and caecal microbiota in chickens and analysed associations between individual bacteria and growth performance. Two trials were undertaken; each used 96 individually caged 15-day-old Cobb broilers. Trial 1 had a control group (n = 48) and a zinc bacitracin (50 ppm) treatment group (n = 48). Trial 2 had a control group (n = 48) and an avilamycin (15 ppm) treatment group (n = 48). Chicken growth performance was evaluated over a 10-day period, and caecal microbiota was characterised by sequencing of bacterial 16S rRNA gene amplicons. Avilamycin produced no effect on growth performance and exhibited little significant disturbance of the microbiota structure. However, zinc bacitracin reduced the feed conversion ratio (FCR) in treated birds, changed the composition and increased the diversity of their caecal microbiota by reducing dominant species. Avilamycin only produced minor reductions in the abundance of two microbial taxa, whereas zinc bacitracin produced relatively large shifts in a number of taxa, primarily Lactobacillus species. Also, a number of phylotypes closely related to lactobacilli species were positively or negatively correlated with FCR values, suggesting contrasting effects of Lactobacillus spp. on chicken growth performance. By harnessing such bacteria, it may be possible to develop high-productivity strategies in poultry that rely on the use of probiotics and less on in-feed antibiotics.Eduardo Crisol-Martínez, Dragana Stanley, Mark S.     © 2016 Ecological Society of Australia Microbats perform important ecological services in agro-ecosystems, but several species are globally threatened by loss of roosting and breeding habitats. The successful conservation of bats in agricultural land requires adequate knowledge of their ecology. Using ultrasonic recorders, we studied the activity of insectivorous bats in areas of macadamia production in eastern Australia at two spatial scales: across woodland-orchard transects at the local scale and across three levels of fragmentation at the landscape scale. At the local scale, activity patterns of ‘clutter’ and ‘edge’ specialists were consistently higher in woodland patches, gradually decreasing towards isolated orchards, where only a few ‘open’ specialists were active. At the landscape scale, bat community activity was affected by the level of fragmentation, partly because three of the most recorded taxa (Austronomus australis, Saccolaimus flaviventris and Miniopterus australis) had their highest activity in less-fragmented areas. A distance-based model explained 24% of the bat community activity based on a combination of six environmental variables. Canonical correspondence analysis showed that a number of bat taxa were associated with open areas of macadamia, whereas other taxa were associated with increasing values of landscape composition, and arthropod and water availability. In addition, total bat activity was highly correlated with foraging rate. These results suggest that most bat taxa were influenced by proximity to woodland and the degree of fragmentation, and only few taxa were able to exploit isolated orchards. Environmental factors that promote bat activity could be exploited to strengthen conservation efforts. Preserving remnant woodland and promoting habitat heterogeneity will benefit several bat species. In particular, the foraging activity of ‘edge’ specialists could be fostered by increasing landscape connectivity and maintaining unobstructed water bodies near macadamia orchards. Considering that bats forage as they navigate these areas, conservation efforts could also bring benefits to farmers through pest-reduction services

    Susceptibility and tolerance in hybrid and pure-line rice varieties to herbivore attack: biomass partitioning and resource-based compensation in response to damage

    © 2016 Association of Applied Biologists Hybrid rice has been noted for its susceptibility to insects and diseases compared to pure-line (conventional) rice varieties. We investigated herbivory by Nilaparvata lugens, Sogatella furcifera and Scirpophaga incertulas on replicated three-line hybrid sets (parental and hybrid lines) in field and greenhouse experiments. In a field experiment, caterpillar densities and stemborer damage was similar among hybrid and parental lines. In field and greenhouse experiments, the cytoplasmic male sterile (CMS)-lines and maintainer lines had higher densities of planthoppers (including N. lugens and S. furcifera) than restorer or hybrid lines likely because of their wild abortive CMS-lineage. High nitrogen levels increased plant mortality due to N. lugens, but often reduced mortality from S. furcifera and S. incertulas: this was similar between hybrid and pure-line varieties. The hybrids were generally more tolerant of herbivory (lower biomass reductions per unit weight of insect) than the inbred parental lines. The addition of nitrogen to both the hybrid and pure-line varieties had contrasting effects on tolerance depending on the nature of the attacking insect: fertiliser increased tolerance to S. furcifera (lower losses of yield and shoot biomass per mg insect) and S. incertulas (lower yield, shoot and root biomass loss) but fertiliser reduced tolerance to N. lugens (higher loss of root biomass and no effects on yield and shoot biomass loss). Our results indicate that hybrid rice is not physiologically more susceptible to herbivores than are pure-line varieties even under high nitrogen conditions, but does have higher tolerance to insect damage

    Sorghum and wheat differentially affect caecal microbiota and associated performance characteristics of meat chickens

    This study compared the effects of wheat-and sorghum-based diets on broiler chickens. The growth performance and caecal microbial community of chickens were measured and correlations between productivity and specific gut microbes were observed. Cobb broilers 15 days of age were individually caged and two dietary treatments were used, one with a wheat-based diet (n D 48) and another one with a sorghum-based diet (n D 48). Growth performance measurements were taken over a 10 day period and samples for microbiota analysis were taken at the end of that period. Caecal microbiota was characterised by sequencing of 16S bacterial rRNA gene amplicons. Overall, the results indicated that a sorghum-based diet produced higher apparent metabolisable energy (AME) and body-weight gain (BWG) values in chickens, compared to a wheatbased diet. Nevertheless, sorghum-fed birds had higher feed conversion ratio (FCR) values than wheat-fed birds, possibly because of some anti-nutritional factors in sorghum. Further analyses showed that caecal microbial community was significantly associated with AME values, but microbiota composition differed between dietary treatments. A number of bacteria were individually correlated with growth performance measurements. Numerous OTUs assigned to strains of Lactobacillus crispatus and Lachnospiraceae, which were prevalent in sorghum-fed chickens, were correlated with high AME and BWG values, respectively. Additionally, a number of OTUs assigned to Clostridiales that were prevalent in wheat-fed chickens were correlated with low FCR values. Overall, these results suggest that between-diet variations in growth performance were partly associated with changes in the caecal microbiota. © 2017 Crisol-Martínez et al

    A low dose of an organophosphate insecticide causes dysbiosis and sex-dependent responses in the intestinal microbiota of the Japanese quail (Coturnix japonica)

In this work we used the Japanese quail (Coturnix japonica) as an avian model to study short-term microbial community responses to a single dose of trichlorfon at low concentration in three sample origins of the gastrointestinal tract (GIT): caecum, large intestine and faeces. Using next-generation sequencing of 16S rRNA gene amplicons as bacterial markers, the study showed that ingestion of insecticide caused significant changes in the GIT microbiome. Specifically, microbiota composition and diversity differed between treated and untreated quail. Insecticide-associated responses in the caecum showed differences between sexes which did not occur with the other sample types. In caecal microbiota, only treated females showed significant shifts in a number of genera within the Lachnospiraceae and the Enterobacteriaceae families. The major responses in the large intestine were a significant reduction in the genus Lactobacillus and increases in abundance of a number of Proteobacteria genera. All microbial shifts in faeces occurred in phylotypes that were represented at low relative abundances. In general, changes in microbiota possibly resulted from contrasting responses towards the insecticide, either positive (e.g., biodegrading bacteria) or negative (e.g., insecticide-susceptible bacteria). This study demonstrates the significant impact that organophosphate insecticides have on the avian gut microbiota; showing that a single small dose of trichlorfon caused dysbiosis in the GIT of the Japanese quail.     Using next-generation sequencing to contrast the diet and explore pest-reduction services of sympatric bird species in macadamia orchards in Australia

    Worldwide, avian communities inhabiting agro-ecosystems are threatened as a consequence of agricultural intensification. Unravelling their ecological role is essential to focus conservation efforts. Dietary analysis can elucidate bird-insect interactions and expose avian pest-reduction services, thus supporting avian conservation. In this study, we used next-generation sequencing to analyse the dietary arthropod contents of 11 sympatric bird species foraging in macadamia orchards in eastern Australia. Across all species and based on arthropod DNA sequence similarities ≄98% with records in the Barcode of Life Database, 257 operational taxonomy units were assigned to 8 orders, 40 families, 90 genera and 89 species. These taxa included 15 insect pests, 5 of which were macadamia pests. Among the latter group, Nezara viridula (Pentatomidae; green vegetable bug), considered a major pest, was present in 23% of all faecal samples collected. Results also showed that resource partitioning in this system is low, as most bird species shared large proportion of their diets by feeding primarily on lepidopteran, dipteran and arachnids. Dietary composition differed between some species, most likely because of differences in foraging behaviour. Overall, this study reached a level of taxonomic resolution never achieved before in the studied species, thus contributing to a significant improvement in the avian ecological knowledge. Our results showed that bird communities prey upon economically important pests in macadamia orchards. This study set a precedent by exploring avian pest-reduction services using next-generation sequencing, which could contribute to the conservation of avian communities and their natural habitats in agricultural systems. © 2016 Crisol-MartĂ­nez et al
