    Current challenges in a nutritional therapy of hypercholesterolaemia

    Hipercholesterolemia, określana jako nadmier ne stężenie lipidów w surowicy kr wi, nie jest stanem chorobowym, natomiast należy do grupy zaburzeń metabolicznych i predysponuje do rozwoju chorób sercowo-naczyniowych. Terapią pierwszego rzutu w leczeniu hipercholesterolemii jest modyfikacja diety. Postępowanie dietetyczne polega na ograniczeniu podaży kalorii, tłuszczu, nasyconych kwasów tłuszczowych oraz cholesterolu. W diecie osób z hipercholesterolemią należy z kolei zwiększyć podaż rozpuszczalnej frakcji włókna pokarmowego, steroli/ /stanoli roślinnych (ok. 2 g/dzień) oraz niezbędnych kwasów tłuszczowych z rodziny omega-3 (EPA, DHA). We wspomagającym leczeniu hipercholesterolemii zastosowanie znajdują również suplementy żywności o działaniu hipolipemizującym, takie jak: rozpuszczalna frakcja włókna pokarmowego (b-glukan, psyllium, pektyny, guma guar), probiotyki (Lactobacillus coagulans, Rhodospirillum rubrum, grzyby z rodzaju Monascus), białko soi (izoflawony: genisteina, daidzeina, gliceteina) oraz fitoestrogeny.Hypercholesterolaemia is characterized by raised blood choleste rol level. It is not a disease itself, but rather metabolic disorder which can lead to development of cardiovascular diseases. The first line therapy is lifestyle modification including diet. The intake of calories, fat, saturated fatty acids and cholesterol should be minimized. On the other hand, it is advisable to increase consumption of soluble fiber, plant sterols/stanols (2 g/day) and omega-3 essential fatty acids (EPA, DHA). It is also recommended to use adjuvant therapy and incl ude in a diet food pro - ducts which decrease cholesterol level: soluble fiber (b-glucan, psyllium, pectins, guar gum), probiotics (Lactobacillus coagulans, Rhodospirillum rubrum, fungus Monascus), soy protein (isoflavones: genistein, daidzein, glycitein) and fitoestrogens

    Modeling Oncogenic Signaling in Colon Tumors by Multidirectional Analyses of Microarray Data Directed for Maximization of Analytical Reliability

    Clinical progression of colorectal cancers (CRC) may occur in parallel with distinctive signaling alterations. We designed multidirectional analyses integrating microarray-based data with biostatistics and bioinformatics to elucidate the signaling and metabolic alterations underlying CRC development in the adenoma-carcinoma sequence.Studies were performed on normal mucosa, adenoma, and carcinoma samples obtained during surgery or colonoscopy. Collections of cryostat sections prepared from the tissue samples were evaluated by a pathologist to control the relative cell type content. The measurements were done using Affymetrix GeneChip HG-U133plus2, and probe set data was generated using two normalization algorithms: MAS5.0 and GCRMA with least-variant set (LVS). The data was evaluated using pair-wise comparisons and data decomposition into singular value decomposition (SVD) modes. The method selected for the functional analysis used the Kolmogorov-Smirnov test. Expressional profiles obtained in 105 samples of whole tissue sections were used to establish oncogenic signaling alterations in progression of CRC, while those representing 40 microdissected specimens were used to select differences in KEGG pathways between epithelium and mucosa. Based on a consensus of the results obtained by two normalization algorithms, and two probe set sorting criteria, we identified 14 and 17 KEGG signaling and metabolic pathways that are significantly altered between normal and tumor samples and between benign and malignant tumors, respectively. Several of them were also selected from the raw microarray data of 2 recently published studies (GSE4183 and GSE8671).Although the proposed strategy is computationally complex and labor–intensive, it may reduce the number of false results

    Functional features of gene expression profiles differentiating gastrointestinal stromal tumours according to KIT mutations and expression

    <p>Abstract</p> <p>Background</p> <p>Gastrointestinal stromal tumours (GISTs) represent a heterogeneous group of tumours of mesenchymal origin characterized by gain-of-function mutations in <it>KIT </it>or <it>PDGFRA </it>of the type III receptor tyrosine kinase family. Although mutations in either receptor are thought to drive an early oncogenic event through similar pathways, two previous studies reported the mutation-specific gene expression profiles. However, their further conclusions were rather discordant. To clarify the molecular characteristics of differentially expressed genes according to GIST receptor mutations, we combined microarray-based analysis with detailed functional annotations.</p> <p>Methods</p> <p>Total RNA was isolated from 29 frozen gastric GISTs and processed for hybridization on GENECHIP<sup>® </sup>HG-U133 Plus 2.0 microarrays (Affymetrix). <it>KIT </it>and <it>PDGFRA </it>were analyzed by sequencing, while related mRNA levels were analyzed by quantitative RT-PCR.</p> <p>Results</p> <p>Fifteen and eleven tumours possessed mutations in <it>KIT </it>and <it>PDGFRA</it>, respectively; no mutation was found in three tumours. Gene expression analysis identified no discriminative profiles associated with clinical or pathological parameters, even though expression of hundreds of genes differentiated tumour receptor mutation and expression status. Functional features of genes differentially expressed between the two groups of GISTs suggested alterations in angiogenesis and G-protein-related and calcium signalling.</p> <p>Conclusion</p> <p>Our study has identified novel molecular elements likely to be involved in receptor-dependent GIST development and allowed confirmation of previously published results. These elements may be potential therapeutic targets and novel markers of <it>KIT </it>mutation status.</p

    Analysis of the Sustainable Development Index in the Communes of the Podkarpackie Voivodeship: A Polish Case Study

    Intense global urbanization, including spatial planning development, is an essential area that determines sustainable development. It is known that urban development is typically tied to an increase in socioeconomic productivity while also creating considerable inequalities. Despite mounting evidence of intense urban area development, little is known about its consequences on the sustainable development of territories adjacent to said areas. Despite the positive and negative consequences of urbanization and their impact on sustainable development often being highlighted, there is little understanding of and a dearth of analyses on sustainability processes that include spatial planning development. To fill this gap, it must be assessed where sustainable development is actually taking place. Such analyses should not only be confined to the four essential areas: economic development, social development, environmental development, and institutional development, that are tied to sustainable development index calculations. They should also determine the transformations experienced by the areas and factor in a fifth analysis area: spatial planning development. In this paper, detailed data sourced from the Statistics Poland were used to formulate sustainable development indices for urban, rural&ndash;urban, and rural communes of the Podkarpackie region of Poland. The data concerned the five areas listed above. Using data standardization and the averaged index method, sustainable development index values were quantified to demonstrate that they displayed various levels of inequalities for the two reference periods of 2015 and 2020. These statistics indicate the key role of spatial planning development in assessing sustainability indices. The findings show that it is not only possible to enhance standard calculation methods to include other data and use them in time and space to create a simple and general quantitative rating of sustainable development, but urbanization can also be factored in that includes spatial planning development. The findings show that a modified computation approach is a reliable and relatively complete index of sustainable development that compensates for the deficiencies of current metrics

    Renal regulation of sodium, potassium and chloride balance in single- and twin-pregnant goats

    The aim of these studies was to analyse and compare changes in selected parameters of renal function in terms of water-electrolyte balance regulation in single- and twin-pregnant goats. Clearance analyses were carried out on 16 pregnant White Improved goats (8 in single and 8 in twin gestation). Blood plasma and urine samples were analysed for the concentration of inulin, endogenous creatinine, sodium, potassium, and chlorides. It has been demonstrated that glomerular filtration rate (GFR) in the goat kidney does not change significantly during gestation. GFR recorded from the 1st week until the 20th week of gestation in twin-pregnant goats was only slightly higher compared to those observed in single-pregnant does. Blood plasma concentrations of major electrolytes, i.e. sodium, potassium and chloride ions, did not differ significantly in pregnant and non-pregnant goats, and remained within the reference values. From the very beginning of gestation, the single-pregnant goats showed increased renal potassium clearance; however, the level of sodium clearance remained stable. On the other hand, sodium clearance increased from the 2nd month of gestation in the twin-pregnant goats, while the load of excreted potassium did not change. These changes had probably resulted from varied levels of aldosterone and progesterone and their mutual proportions differing between the groups

    The Role of Cluster C19MC in Pre-Eclampsia Development

    Pre-eclampsia is a placenta-related complication occurring in 2&ndash;10% of all pregnancies. miRNAs are a group of non-coding RNAs regulating gene expression. There is evidence that C19MC miRNAs are involved in the development of the placenta. Deregulation of chromosome 19 microRNA cluster (C19MC) miRNAs expression leads to impaired cell differentiation, abnormal trophoblast invasion and pathological angiogenesis, which can lead to the development of pre-eclampsia. Information was obtained through a review of articles available in PubMed Medline. Articles on the role of the C19MC miRNA in the development of pre-eclampsia published in 2009&ndash;2022 were analyzed. This review article summarizes the current data on the role of the C19MC miRNA in the development of pre-eclampsia. They indicate a significant increase in the expression of most C19MC miRNAs in placental tissue and a high level of circulating fractions in serum and plasma, both in the first and/or third trimester in women with PE. Only for miR-525-5p, low levels of plasma expression were noted in the first trimester, and in the placenta in the third trimester. The search for molecular factors indicating the development of pre-eclampsia before the onset of clinical symptoms seems to be a promising diagnostic route. Identifying women at risk of developing pre-eclampsia at the pre-symptomatic stage would avoid serious complications in both mothers and fetuses. We believe that miRNAs belonging to cluster C19MC could be promising biomarkers of pre-eclampsia development

    Analysis of chemical stability of tap water in terms of required level of technological safety

    The main goal of this work is to show the new approach to determining safety technological levels (SLs) in terms of water quality and its chemical stability, as well as issues of water corrosion properties in water distribution systems (WDSs), due to the fact that water supply pipes are prone to corrosion. In the paper the methodology of determining the risk associated with threat to technical infrastructure was considered. The concept was studied on the basis of real operational data from the water treatment plant. The probability of exceeding the individual parameters for WTPI is slightly larger than for WTPII, which means that this water treatment process may cause lack of chemical stability in the water supply network. Operators should anticipate in the process of designing water distribution system, using proper materials, as to ensure an adequate level of safety from the water source to the water recipient. It should be noted that it is necessary to adjust the material of internal installation of water supply networks to the parameters of the water. At present, there are no correlations between the designing step and water parameters. It was concluded that to protect the water supply infrastructure, which belongs to critical infrastructure, water company should put more emphasis on distribution of stable water that has not potentially corrosion properties. Some suggestions were made for the protection of WDS and to ensure safety of system functioning and long-term usability of water pipes

    Melanin—The Éminence Grise of Melanoma and Parkinson’s Disease Development

    A common feature of Parkinson’s disease (PD) and melanoma is their starting points being based on cells capable of converting tyrosine into melanin. Melanocytes produce two types of melanin: eumelanin and pheomelanin. These dyes are designed to protect epidermal cells from the harmful effects of UV radiation. Neurones of the substantia nigra, which degenerate during PD, produce neuromelanin, the physiological role of which is not fully explained. This article discusses the potential role of melanins in the pathogenesis of both diseases. Melanins, due to their ability to accumulate toxic substances, may become their sources over time. The use of glutathione for the synthesis of pheomelanins and neuromelanins may reduce the antioxidant capacity of cells, leading to an excessive synthesis of free radicals. This study also tested the hypothesis that certain drugs used in the treatment of PD (L-DOPA, MAO-B and COMT inhibitors, and amantadine), aimed at increasing dopamine concentration, could potentially contribute to the development of melanoma. The role and properties of melanins should continue to be researched. Whether excessive melanin synthesis or its accumulation in the extracellular space may be factors initiating the development of diseases remains an open question

    The Nitrogen Fixation and Yielding of Pea in Different Soil Tillage Systems

    The field experiment was carried out in 2013 and 2014 as part of a long-term experiment to test the influence of different soil tillage systems (conventional, reduced, direct sowing) on (a) the yield of summer pea (Pisum sativum L.) and (b) the amount of biologically reduced atmospheric nitrogen (N), which was determined using the 15N dilution method (ID15N). Spring barley was used as a reference plant. Climatic conditions did not have a significant influence on the yield of pea seeds (mean value 4.56 t ha−1), post-harvest residues (3.76 t ha−1) and total biomass (8.33 t ha−1). Soil tillage system was found to have a significant impact on all components of the pea yield in years of experiment, with the highest average seed yield values observed with the conventional system (5.19 t ha−1) and significantly lower values observed with reduced tillage (4.34 t ha−1) and direct sowing (4.17 t ha−1). The content of total N was greatest in the pea biomass that was harvested in the conventional system (202 kg ha−1) and lowest with direct sowing (155 kg ha−1). Nitrogen accumulated in the pea seeds from three sources: atmosphere (mean value 35.2%), fertilizers (6.8%) and soil (57.9%), equating to 48.6, 9.9, and 85.4 kg ha−1, respectively. Soil tillage system was found to have a significant impact on the amount of N that was fixed from the atmosphere by the peas: 17.7% in conventional tillage, 37.9 in reduced system and 50.2% in direct sowing, which equates to a harvested seed yield of 28.9, 52.0 and 64.4 kg ha−1, respectively. In the post-harvest residues, the amount of N fixed from the atmosphere by the pea crop was also modified by the soil tillage system in range: 20.2% in conventional tillage and 32.6% in direct sowing (which equates to 7.4 and 8.5 kg N ha−1, respectively), but the difference was not significant
