    Research on forming quality of poly-wedge pulley spinning

    As an important power transmission part, pulleys are widely used in automobile industry, agricultural machinery, pumps and machines. A near-net forming process for six-wedge belt pulleys manufacturing was put forward. For this purpose, the required tooth shape and size can be formed directly by spinning without machining. The whole manufacturing procedures include blanking, drawing and spinning. The spinning procedure includes five processes, performing, drumming, thickening, toothing and finishing. The forming defects occurred during each forming processes of poly-wedge pulley spinning, such as the drumming failure, flanged opening-end, folded side-wall, insufficient bottom size, flashed opening-end, cutting-off bottom, are introduced, and the factors influencing the defects are analyzed.     Thermal conductivity of deformed carbon nanotubes

    We investigate the thermal conductivity of four types of deformed carbon nanotubes by using the nonequilibrium molecular dynamics method. It is reported that various deformations have different influence on the thermal properties of carbon nanotubes. For the bending carbon nanotubes, the thermal conductivity is independent on the bending angle. However, the thermal conductivity increases lightly with XY-distortion and decreases rapidly with Z-distortion. The thermal conductivity does not change with the screw ratio before the breaking of carbon nanotubes but decreases sharply after the critical screw ratio.Comment: 6figure

    Seroprevalence and Risk Factors of Chlamydia abortus

    Chlamydia abortus, an important pathogen in a variety of animals, is associated with abortion in sheep. In the present study, 1732 blood samples, collected from Tibetan sheep between June 2013 and April 2014, were examined by the indirect hemagglutination (IHA) test, aiming to evaluate the seroprevalence and risk factors of C. abortus infection in Tibetan sheep. 323 of 1732 (18.65%) samples were seropositive for C. abortus antibodies at the cut-off of 1 : 16. A multivariate logistic regression analysis was used to evaluate the risk factors associated with seroprevalence, which could provide foundation to prevent and control C. abortus infection in Tibetan sheep. Gender of Tibetan sheep was left out of the final model because it is not significant in the logistic regression analysis (P>0.05). Region, season, and age were considered as major risk factors associated with C. abortus infection in Tibetan sheep. Our study revealed a widespread and high prevalence of C. abortus infection in Tibetan sheep in Gansu province, northwest China, with higher exposure risk in different seasons and ages and distinct geographical distribution

    MicroRNA 27b-3p Modulates SYK in Pediatric Asthma Induced by Dust Mites

    The PI3K-AKT pathway is known to regulate cytokines in dust mite-induced pediatric asthma. However, the underlying molecular steps involved are not clear. In order to clarify further the molecular steps, this study investigated the expression of certain genes and the involvement of miRNAs in the PI3K-AKT pathway, which might affect the resultant cytokine-secretion. in-vivo and in-vitro ELISA, qRT-PCR and microarrays analyses were used in this study. A down-expression of miRNA-27b-3p in dust mite induced asthma group (group D) was found by microarray analysis. This was confirmed by qRT-PCR that found the miRNA-27b-3p transcripts that regulated the expression of SYK and EGFR were also significantly decreased (p < 0.01) in group D. The transcript levels of the SYK and PI3K genes were higher, while those of EGFR were lower in the former group. Meanwhile, we found significant differences in plasma concentrations of some cytokines between the dust mite-induced asthma subjects and the healthy controls. On the other hand, this correlated with the finding that the transcripts of SYK and its downstream PI3K were decreased in HBE transfected with miRNA-27b-3p, but were increased in HBE transfected with the inhibitor in vitro. Our results indicate that the differential expression of the miRNAs in dust mite-induced pediatric asthma may regulate their target gene SYK and may have an impact on the PI3K-AKT pathway associated with the production of cytokines. These findings should add new insight into the pathogenesis of pediatric asthma

    Effect of radiation on the Notch signaling pathway in osteoblasts

    Notch signaling has been shown to be important in osteoblast differentiation. Therapeutic radiation has been shown to alter the skeletal system, yet little information is available on the changes in Notch signaling in irradiated osteoblasts. The purpose of this study was to analyze the effect of radiation therapy with 2 and 4 Gy on Notch signaling in osteoblasts. In order to assess the radiation damage on osteoblast differentiation, total RNA and protein were collected three days after exposure to radiation. The effects of radiation on Notch signaling at the early and terminal stages of osteoblastic MC3T3-E1 cell differentiation was analyzed by qRT-PCR and western blot analysis. Our study applied a previously established method to induce MC3T3-E1 cell differentiation into osteoblasts and osteoblast precursors. Our results showed that the expression of Notch receptors (Notch1-4), ligands (Jagged1, Jagged2 and Delta1), target of Notch signaling (Hes1) and markers (ALP, M-CSF, RANKL and OPG) were altered following 2 and 4 Gy of irradiation. The present research did not indicate a strong relationship between Notch1 regulation and suppression of osteoblast differentiation. We found Hes1 may play a role in the radiation effect on osteoblast differentiation. Our results indicate that radiated osteoblast precursors and osteoblasts promoted osteoclast differentiation and proliferation

    Multistage auto-ignition of undiluted methane/air mixtures under engine-relevant condition

    Gas-phase auto-ignition delay times (IDTs) of methane/“air” (21% O2/79% Ar) mixtures were measured behind reflected shock waves, using a kinetic shock tube. Experiments were performed at fixed pressure of 1.8 MPa and equivalence ratios of 0.5 and 1.0, over the temperature range of 800–1000 K. Overall, the effect of equivalence ratio on IDT is negligible at entire temperatures measured in this study. The difference from traditional ignition regime at high temperatures, the undiluted methane/air mixtures present a four-stage ignition process at lower temperatures, namely deflagration delay, deflagration, deflagration-detonation transition, and detonation. Four popular kinetic mechanisms, UBC Mech 2.1, GRI Mech 3.0, Aramco Mech 2.0, and USC Mech 2.0, were used to simulate the new measurements. Only UBC Mech 2.1 showed satisfactory predictions in the reactivity of the undiluted methane mixtures; it was, thus, adopted to perform sensitivity analysis for identifying dominant reactions in the ignition process. The difference in channels contributing ȮH radicals causes a reduced global activation energy with decreasing temperatures.Keywords: Methane; multistage ignition; shock tube; sensitivity analysi

    Magnetic criticality-enhanced hybrid nanodiamond-thermometer under ambient conditions

    Nitrogen vacancy (NV) centres in diamond are attractive as quantum sensors owing to their superb coherence under ambient conditions. However, the NV centre spin resonances are relatively insensitive to some important parameters such as temperature. Here we design and experimentally demonstrate a hybrid nano-thermometer composed of NV centres and a magnetic nanoparticle (MNP), in which the temperature sensitivity is enhanced by the critical magnetization of the MNP near the ferromagnetic-paramagnetic transition temperature. The temperature susceptibility of the NV center spin resonance reached 14 MHz/K, enhanced from the value without the MNP by two orders of magnitude. The sensitivity of a hybrid nano-thermometer composed of a Cu_{1-x}Ni_{x} MNP and a nanodiamond was measured to be 11 mK/Hz^{1/2} under ambient conditions. With such high-sensitivity, we monitored nanometer-scale temperature variation of 0.3 degree with a time resolution of 60 msec. This hybrid nano-thermometer provides a novel approach to studying a broad range of thermal processes at nanoscales such as nano-plasmonics, sub-cellular heat-stimulated processes, thermodynamics of nanostructures, and thermal remanent magnetization of nanoparticles.Comment: 21 pages, 6 figure

    Comparative transcriptome and metabolome profiles of the leaf and fruits of a Xianjinfeng litchi budding mutant and its mother plant

    Background: Litchi (Litchi chinensis) is an important sub-tropical fruit in the horticulture market in China. Breeding for improved fruit characteristics is needed for satisfying consumer demands. Budding is a sustainable method for its propagation. During our ongoing breeding program, we observed a litchi mutant with flat leaves and sharp fruit peel cracking in comparison to the curled leaves and blunt fruit peel cracking fruits of the mother plant.Methods: To understand the possible molecular pathways involved, we performed a combined metabolome and transcriptome analysis.Results: We identified 1,060 metabolites in litchi leaves and fruits, of which 106 and 101 were differentially accumulated between the leaves and fruits, respectively. The mutant leaves were richer in carbohydrates, nucleotides, and phenolic acids, while the mother plant was rich in most of the amino acids and derivatives, flavonoids, lipids and organic acids and derivatives, and vitamins. Contrastingly, mutant fruits had higher levels of amino acids and derivatives, carbohydrates and derivatives, and organic acids and derivatives. However, the mother plant’s fruits contained higher levels of flavonoids, scopoletin, amines, some amino acids and derivatives, benzamidine, carbohydrates and derivatives, and some organic acids and derivatives. The number of differentially expressed genes was consistent with the metabolome profiles. Gene Ontology and Kyoto Encyclopedia of Genes and Genomes pathway-enriched gene expressions showed consistent profiles as of metabolome analysis.Conclusion: These results provide the groundwork for breeding litchi for fruit and leaf traits that are useful for its taste and yield
