    THE COMBINED EFFECT OF DEXAMETHASONE AND BACILLUS CALMETTE–GUÉRIN ON THE NEUROBEHAVIORAL ASPECT IN MALE MICE (MUS MUSCULUS)

    Objective: Our study aims to evaluate the combined effect of Bacille Calmette–GuĂ©rin (BCG) and dexamethasone (DEX) on male mice undergoing behavioral testing (forced swimming test, elevated plus maze, tail suspension test, and sucrose intake). An intraperitoneal injection of 0.2 ml/mg of BCG and 0.1 ml/mg of DEX body weight was administered after all behavioral tests were applied. Methods: This study is based on the distribution of 15 male mice in three groups: Controls, DEX, and DEX+BCG. These mice underwent behavioral tests. Results: The result after administration of DEX and BCG was found after 10 days. We observed an increase in locomotors activity with some degree of anxiety. Conclusion: This allowed us to conclude that the combination of DEX+BCG seems to play a depressinogenic effect that will develop to become antidepressive

    Preventive effect of zinc on nickel-induced oxidative liver injury in rats

    This study pertains to the potential ability of zinc, used as nutritional supplements, to alternate oxidative stress induced by nickel. Male rats were randomly divided into four groups of eight each. Group I served as the controls; group II received in their drinking water ZnSO4 (227 mg/l); group III received NiSO4 (2 mg/100b.w/day intraperitoneally); group IV was treated with ZnSO4 and NiSO4. The exposure of rats to nickel sulfate for 21 days resulted in a significant decrease in body weight gain and absolute liver weight, relative liver weight. Nickel treatment also produced oxidative liver injury characterized by increasing serum glucose concentration, glutamate-pyruvate transaminase (GPT), alanine aminotransferase (GOT) and alkaline phosphatase (ALP) activities. Meanwhile nickel supplementation decreased serum total protein and albumin in animals. In addition, liver glutathione level, catalase and glutathione peroxidase (GSH-Px) activities were diminished. The administration of zinc with nickel (Ni + Zn) corrective effects on Ni-induced oxidative stress in liver was observed. In conclusion, this study demonstrates that intraperitoneally injection with Ni caused reduction in enzymes activities in rat’s liver and treatment with zinc offers a relative protection against nickel induced oxidative liver injury and lipid peroxidation probably due to its antioxidant proprieties.Key words: Nickel, zinc, rats, oxidative stress, liver

    The protective effect of Zinc and Magnesium against subchronic Cadmium toxicity in Wistar rats (Biochemical and neurobehavioral effects)

    Objective: The present study shows the beneficial effect of (zinc 10mg / kg + magnesium 10mg / kg) against cadmium poisoning at a dose of 15mg / kg on biochemical parameters and neurobehavioral functions in Wistar rats during a period of 45 days. Methods: The experiment was carried out on 20 Wistar rats, weighing 180 to 232g for an initial weight before starting treatment with cadmium. The rats are grouped in cages at a rate of 5 in groups:Group (1) control: was supplied only with waterGroup (2) contaminated with cadmium (Cd):  water plus oral administration cadmium dose 15mg / Kg / l.Group (3) contaminated with (Cd) dose 15mg / Kg / l in water plus magnesium (Mg) dose 10mg / Kg / l oral administration.Group (4) contaminated with (Cd) dose 15mg / Kg / l in water plus zinc (Zn) dose 10mg / Kg / l oral administration.Results: The rats exposure to cadmium showed a very highly significant decrease in body weight of cadmium-contaminated rats (P <0.001) compared with the control group. Regarding the biochemical parameters, there was a very highly significant increase (P <0.001) in the cadmium group blood glucose level compared to the control group, a highly significant increase (P <0.01) in the group urea level. cadmium compared to the control, a very highly significant increase (P <0.001) in the creatinine level of the cadmium lot compared to the control, a very highly significant increase (P <0.001) of transaminases (GOT, GPT) of the group cadmium relative to the control, complete blood  count (CBC) demonstrated a very highly significant increase (P <0.001) in the white blood cell count, the hematocrit of the cadmium lot compared to the control, a very highly significant decrease (P <0.001). ) the red blood cell and hemoglobin levels of the cadmium group compared to the control. A significant improvement (P <0.001) (P <0.01) (P <0.05) of lots: zinc, magnesium, cadmium + zinc and cadmium + zinc compared to cadmium. However, the results obtained from the neurobehavioral tests reveal a significant elevation (P <0.001) (P <0.01) (P <0.05) in the number of cells crossed, the number of straightening in the test. Open field in the group exposed to cadmium resulting in locomotors hyperactivity compared to the control and it is minimal in other groups treated with zinc and magnesium. On the other hand, the plus maze (labyrinth test) revealed a very highly significant increase (P <0.001) (P <0.01) (P <0.05) of the time spent in the closed arms of the cadmium group compared to the control, a significant improvement (P <0.001) (P <0.01) (P <0.05) of the lots: zinc, magnesium, cadmium + zinc and cadmium + zinc compared with cadmium. Conclusion: Subchronic cadmium toxicity has harmful effects on the biochemical, hematological and neuro-behavioral parameters of  Wistar rats, with the installation of an anxiety that will lead to a depressive state, which will be reduced and improved by the antioxidant effect of zinc + magnesium. Keywords: Cadmium-zinc-magnesium-subchronic toxicity-Wistar-Open field-plus mazeÂ

    Effect of Mg contents on the mechanical proprieties and precipitation kinetics in Al–3.3 wt.% Cu alloy

    The effect of additional Mg on the microstructure, mechanical properties, and transformation kinetics during aging in Al–3.3 wt.% Cu alloy was studied. The compositions and microstructure were examined by X-ray diffraction, Differential scanning calorimetry (DSC) and scanning electron microscope (SEM) with energy dispersive X-ray spectroscopy (EDS). The results show that the Mg in the Al–Cu alloy mainly precipitated to the grain boundaries during the process of transformation and formed a ternary Al2CuMg metallic compound and the rate of discontinuous precipitation reaction decreases with increasing concentration of Mg.     Association between erythrocyte Na+K+-ATPase activity and some blood lipids in type 1 diabetic patients from Lagos, Nigeria

    <p>Abstract</p> <p>Background</p> <p>Altered levels of erythrocyte Na<sup>+</sup>K<sup>+</sup>-ATPase, atherogenic and anti-atherogenic lipid metabolites have been implicated in diabetic complications but their pattern of interactions remains poorly understood.</p> <p>This study evaluated this relationship in Nigerian patients with Type 1 diabetes mellitus.</p> <p>Methods</p> <p>A total of 34 consented Type 1 diabetic patients and age -matched 27 non-diabetic controls were enrolled. Fasting plasma levels of total cholesterol, triglycerides and HDL-cholesterol were determined spectrophotometrically and LDL-cholesterol estimated using Friedewald formula. Total protein content and Na+K+-ATPase activity were also determined spectrophotometrically from ghost erythrocyte membrane prepared by osmotic lysis.</p> <p>Results</p> <p>Results indicate significant (P < 0.05) reduction in Na<sup>+</sup>K<sup>+</sup>-ATPase activity in the Type 1 diabetic patients (0.38 ± 0.08 vs. 0.59 ± 0.07 uM Pi/mgprotein/h) compared to the control but with greater reduction in the diabetic subgroup with poor glycemic control (n = 20) and in whom cases of hypercholesterolemia (8.8%), hypertriglyceridemia (2.9%) and elevated LDL-cholesterol (5.9% each) were found. Correlation analyses further revealed significant (P < 0.05) inverse correlations [r = -(0.708-0.797] between all the atherogenic lipid metabolites measured and Na<sup>+</sup>K<sup>+</sup>-ATPase in this subgroup contrary to group with good glycemic control or non-diabetic subjects in which significant (P < 0.05) Na<sup>+</sup>K<sup>+</sup>-ATPase and HDL-C association were found (r = 0.427 - 0.489). The Na<sup>+</sup>K<sup>+</sup>-ATPase from the diabetic patients also exhibited increased sensitivity to digoxin and alterations in kinetic constants Vmax and Km determined by glycemic status of the patients.</p> <p>Conclusion</p> <p>It can be concluded that poor glycemic control evokes greater reduction in erythrocyte Na<sup>+</sup>K<sup>+</sup>-ATPase activity and promote enzyme-blood atherogenic lipid relationships in Type 1 diabetic Nigerian patients.</p

    Iodine status in pre-school children prior to mandatory iodine fortification in Australia

    The iodine status of children between the ages of 5 and 15 years has been routinely assessed in many countries, but few studies have examined iodine status in pre-school children. We conducted a cross-sectional study of pre-school children living in Adelaide, South Australia, between 2005 and 2007. Children 1–5 years old were identified using a unique sampling strategy to ensure that the study population was representative. A 3-day weighed diet record, a blood sample and a urine sample were obtained from each child. The median urinary iodine concentration (UIC) of the children (n = 279) was 129 ”g L⁻Âč, indicating iodine sufficiency (normal range: 100–199 ”g L⁻Âč), but 35% of the children had a UIC < 100 ”g L−1. The median thyroglobulin concentration of children (n = 217) was 24 ”g L⁻Âč and thyroglobulin concentration declined with increasing age (P = 0.024). The mean daily iodine intake was 76 ”g. The intake of iodine was lower than expected and highlights difficulties in accurately assessing iodine intakes. Further studies are needed to monitor dietary changes and iodine status in this age group since the implementation of mandatory fortification of bread with iodised salt in Australia in 2009.Sheila Skeaff, Ying Zhao, Robert Gibson, Maria Makrides, Shao Jia Zho
