    Fermentation, Isolation, Structure, and antidiabetic activity of NFAT-133 produced by Streptomyces strain PM0324667

    Type-2 diabetes is mediated by defects in either insulin secretion or insulin action. In an effort to identify extracts that may stimulate glucose uptake, similar to insulin, a high throughput-screening assay for measuring glucose uptake in skeletal muscle cells was established. During the screening studies to discover novel antidiabetic compounds from microbial resources a Streptomyces strain PM0324667 (MTCC 5543, the Strain accession number at Institute of Microbial Technology, Chandigarh, India), an isolate from arid soil was identified which expressed a secondary metabolite that induced glucose uptake in L6 skeletal muscle cells. By employing bioactivity guided fractionation techniques, a tri-substituted simple aromatic compound with anti-diabetic potential was isolated. It was characterized based on MS and 2D NMR spectral data and identified as NFAT-133 which is a known immunosuppressive agent that inhibits NFAT-dependent transcription in vitro. Our investigations revealed the antidiabetic potential of NFAT-133. The compound induced glucose uptake in differentiated L6 myotubes with an EC50 of 6.3 ± 1.8 μM without activating the peroxisome proliferator-activated receptor-γ. Further, NFAT-133 was also efficacious in vivo in diabetic animals and reduced systemic glucose levels. Thus it is a potential lead compound which can be considered for development as a therapeutic for the treatment of type-2 diabetes.     Various skin problems such as wrinkles, pimples, acne, and dark circles can all be reduced with the use of herbal face packs. Additionally, they are helpful in improving the fairness and smoothness of the skin. The face packs from natural origin do contain some important ingredients such as vitamins, which are beneficial for skin's health. They act on skin by increasing blood circulation within the veins of the face. The objective of this work is to formulate and evaluate a polyherbal face pack for cosmetic purpose containing herbal ingredients such as, hibiscus, sandalwood, multani mitti, orange peel powder, turmeric and neem.     Synthesis and Antimicrobial Activity of New Carbohydrazide Bearing Quinoline Scaffolds in Silico ADMET and Molecular Docking Studies

    In search of a more potent and new series of fluorine-containing quinoline, hybrid Schiff bases (6a–o) analogues were synthesized by a facile and efficient conventional method. They were developed via condensation of quinoline-4-carbohydrazide intermediate and aromatic aldehydes in presence of ethanol. All compounds viz., 6a–o were efficiently synthesized in good yields in ranges of 76–84%, respectively. All synthesized compounds were well characterized by using various spectroscopic techniques such as FT-IR, 1H NMR, 13C NMR, Mass spectroscopy. Moreover, all newly synthesized hybrid Schiff bases (6a–o) have been screened for their antifungal and antibacterial activity. Among these compounds (6a–d) shows good antibacterial activity, while compound 6b was found to be effective against a fungal pathogen Aspergillus niger and compound 6a was found to inhibit the visible growth of Staphylococcus aureus ATCC 6538 at low concentration with MIC 340 µg/.</p
