    Analysis of gut microbiota in rheumatoid arthritis patients. Disease-related dysbiosis and modifications induced by etanercept

    A certain number of studies were carried out to address the question of how dysbiosis could affect the onset and development of rheumatoid arthritis (RA), but little is known about the reciprocal influence between microbiota composition and immunosuppressive drugs, and how this interaction may have an impact on the clinical outcome. The aim of this study was to characterize the intestinal microbiota in a groups of RA patients treatment-naïve, under methotrexate, and/or etanercept (ETN). Correlations between the gut microbiota composition and validated immunological and clinical parameters of disease activity were also evaluated. In the current study, a 16S analysis was employed to explore the gut microbiota of 42 patients affected by RA and 10 healthy controls. Disease activity score on 28 joints (DAS-28), erythrocyte sedimentation rate, C-reactive protein, rheumatoid factor, anti-cyclic citrullinated peptides, and dietary and smoking habits were assessed. The composition of the gut microbiota in RA patients free of therapy is characterized by several abnormalities compared to healthy controls. Gut dysbiosis in RA patients is associated with different serological and clinical parameters; in particular, the phylum of Euryarchaeota was directly correlated to DAS and emerged as an independent risk factor. Patients under treatment with ETN present a partial restoration of a beneficial microbiota. The results of our study confirm that gut dysbiosis is a hallmark of the disease, and shows, for the first time, that the anti-tumor necrosis factor alpha (TNF-α) ETN is able to modify microbial communities, at least partially restoring a beneficial microbiota

    Ripening-induced changes in microbial groups of artisanal Sicilian goats' milk cheese.

    Changes in the microbial flora of "Caprino dei Nebrodi", a raw goat's milk cheese produced in Sicily, were studied during ripening. From 2 batches of cheese, 4 samples were taken at day 0, 2, 15, and 30 of ripening. Also, samples of curd and milk used in the manufacturing process were analyzed. By the end of the ripening process (day 30), high log10 cfu/g were found for Lactobacilli (7.20), Lattococci (7.10), and Enterococci (7.00), whereas counts of Enterobacteriaceae (3.91), Escherichia coli (3.30), and Staphylococcus (3.89) were found to be lower. The study provides useful information on the microbiological properties of "Caprino dei Nebrodi" cheese, and the results obtained suggest that in order to increase the quality of this artisanal product, it is necessary to improve the sanitary conditions of milking and cheese-making. The study was intended as a preliminary step towards the isolation and identification of bacterial species found in this type of goat's cheese

    Use of peas in organic buffalo farming: effects on nutrient digestibility and milk production

    Twenty lactating buffalo cows, organically farmed, were used to examine the effects of including peas in total mixed ration. Two concentrates were formulated to contain, as the main protein sources, either 350 g/kg of soybean cake (CC) or 450 g/kg of peas (ExpC). Cows were blocked into two groups according to parity and previous milk yield and were assigned to one of two dietary treatments: one group was fed a diet with 6 kg/d of CC, whereas the treatment group was fed diet in which 3 kg/d of CC were replaced by an equal quantity of ExpC. Digestibility of the diets and milk yield of the cows were measured. The experimental period covered the whole lactation period. No differences were observed between groups for milk yield and composition, and for digestibility. The main hypothesis tested, that the replacement of soybean cake with peas in buffalo diet would not affect milk yield and composition,was confirmed. This suggested that the partial substitution of soybean cake with peas in diet for buffalo cows can be possible without affect performances

    In vitro fermentation and chemical characteristics of mediterranean by-products for swine nutrition

    The purpose of the study is to determine the nutritional characteristics of some by-products derived from fruit juice and olive oil production to evaluate their use in pig nutrition. Five by-products of citrus fruit (three citrus fruit pulp and two molasses) and three by-products of olive oil (olive cake) obtained by different varieties are analysed for chemical composition. The fermentation characteristics are evaluated in vitro using the gas production technique with swine faecal inoculum. All the citrus by-products are highly fermentable, producing gas and a high amount of short-chain fatty acids. The fermentation kinetics vary when comparing pulps and molasses. Citrus fruit pulps show lower and slower fermentation rates than molasses. The olive oil by-products, compared to citrus fruits ones, are richer in NDF and ADL. These characteristics negatively affect all the fermentation parameters. Therefore, the high concentration of fiber and lipids represents a key aspect in the nutrition of fattening pigs. The preliminary results obtained in this study confirm that the use of by-products in pig nutrition could represent a valid opportunity the reduce the livestock economic cost and environmental impact

    Evidence for tidal interaction and merger as the origin of galaxy morphology evolution in compact groups

    We present the results of a morphological study based on NIR images of 25 galaxies, with different levels of nuclear activity, in 8 Compact Groups of Galaxies (CGs). We perform independently two different analysis: a isophotal study and a study of morphological asymmetries. The results yielded by the two analysis are highly consistent. For the first time, it is possible to show that deviations from pure ellipses are produced by inhomogeneous stellar mass distributions related to galaxy interactions and mergers. We find evidence of mass asymmetries in 74% of the galaxies in our sample. In 59% of these cases, the asymmetries come in pairs, and are consistent with tidal effects produced by the proximity of companion galaxies. The symmetric galaxies are generally small in size or mass, inactive, and have an early-type morphology. In 20% of the galaxies we find evidence for cannibalism. In 36% of the early-type galaxies the color gradient is positive (blue nucleus) or flat. Summing up these results, as much as 52% of the galaxies in our sample could show evidence of an on going or past mergers. Our observations suggest that galaxies in CGs merge more frequently under ``dry'' conditions. The high frequency of interacting and merging galaxies observed in our study is consistent with the bias of our sample towards CGs of type B, which represents the most active phase in the evolution of the groups. In these groups we also find a strong correlation between asymmetries and nuclear activity in early-type galaxies.     Inhibition of interleukin-6-induced matrix metalloproteinase-2 expression and invasive ability of lemon peel polyphenol extract in human primary colon cancer cells

    Among matrix metalloproteinases (MMPs), MMP-9/2 are key enzymes involved in the proteolysis of extracellular matrices in the inflammatory process and in cancer. Since MMP-9/2 expression levels, activity, and secretion is up-regulated during inflammation in response to pro-inflammatory cytokines, such as interleukin-6 (IL-6), many efforts have been devoted to identifying factors that could inhibit the IL-6-induced MMP-9/2 expression. Up to now, several reports in-dicated that polyphenols from fruits and vegetables are among the major components of health promotion for their antioxidant properties and also for their anti-inflammatory and anti-cancer agents. Among plant derived polyphenols, lemon (Citrus limon) peel extract (LPE) shows anti-cancer properties in various cancer types. In our previous work, we demonstrated that LPE can reduce IL-6-induced migration/invasiveness and MMP-9/2 up-regulation in some gastric cancer cell lines. This study aims to exploit the anti-cancer properties of LPE using an in vitro system model of inflam-mation, consisting of IL-6-exposed human primary colon cancer cells. We first analyzed the effect of LPE on IL-6-induced cell migration and invasiveness by wound healing and Boyden chamber assay, respectively. The MMP-2 mRNA expression levels and gelatinolytic activity in the cell culture media were determined by q-PCR analysis and gelatin zymography, respectively, and finally, the effects of LPE on IL-6-induced JAK2/STAT3 signaling pathways have been investigated by Western blotting analysis. Our results show that LPE is able to inhibit the IL-6-dependent cell migration and invasiveness associated with the up-regulation of MMP-2 expression levels and that these effects are correlated to the STAT3 phosphorylation in human primary T88 and T93 colon cancer cells

    The use of mobile phone while driving: Behavior and determinant analysis in one of the largest metropolitan area of Italy

    The use of mobile phones while driving is one of the main causes of road accidents and it is a phenomenon in continuous growth. The key aim of this study is to analyse simultaneously knowledge, attitudes, and behavior toward the use of mobile phones while driving in one of the largest and populous metropolitan areas of Italy, Naples. The data acquired from 774 questionnaires - administered to subjects evenly divided by gender and with an average age of 39 years - revealed that 69 % have used their mobile phone while driving at least once in their lifetime. Among those who used the phone, 63.6 % use it to make phone calls while 75.2 % only to answer them; 49.1 % read messages and only 33.3 % write them. It is also notable that 34.1 % do not stop to answer a call and only 10 % do not value the use of headsets while driving as fundamental. The results indicate that cell phone usage while driving is common in the study population, despite many having university-level education and satisfactory risks awareness. The multiple linear regression analysis shows how knowledge is not correlated to the behavior held. On the contrary, attitudes are strongly correlated to knowledge and behavior, meaning that good attitudes bring forth positive behavior. According to the collected data and statistical analysis, it is possible to identify factors that can greatly affect the use of mobile phone while driving and establish targeted prevention programs

    The impact of physical activity on psychological health during Covid-19 pandemic in Italy

    The worldwide spread of COVID-19 has upset the normality of Italian daily life, forcing population to social distancing and self-isolation. Since the containment precautions also concern sport-related activities, home workout remained the only possibility to play sports and stay active during the pandemic. The present study aimed to examine changes in the physical activity levels during self-quarantine in Italy, and the impact of exercise on psychological health. A total of 2974 Italian subjects has completed an online survey, but only 2524 subjects resulted eligible for this study. The questionnaire measured the total weekly physical activity energy expenditure before and during quarantine (i.e. the sum of walking, moderate-intensity physical activities, and vigorous-intensity physical activities) in Metabolic Equivalent Task minutes per week (MET–min/wk) using an adapted version of International Physical Activity Questionnaire and their psychological well-being using the Psychological General Well Being Index. Of the 2524 Italian subjects included in the study, 1426 were females (56.4%) and 1098 males (43.6%). Total physical activity significantly decreased between before and during COVID-19 pandemic (Mean: 2429 vs. 1577 MET–min/wk, ∗∗∗∗p < 0.0001), in all age groups and especially in men (Female, mean: 1994 vs. 1443 MET–min/wk, ∗∗∗∗p < 0.0001; Male, mean: 2998 vs. 1754 MET–min/wk, ∗∗∗∗p < 0.0001). Furthermore, a significant positive correlation was found between the variation of physical activity and mental well-being (r = 0.07541, ∗∗∗p = 0.0002), suggesting that the reduction of total physical activity had a profoundly negative impact on psychological health and well-being of population. Based on this scientific evidence, maintaining a regular exercise routine is a key strategy for physical and mental health during a forced rest period like the current coronavirus emergency

    Neutron star radius-To-mass ratio from partial accretion disk occultation as measured through fe kα line profiles

    We present a new method to measure the radius-To-mass ratio (R/M) of weakly magnetic, disk-Accreting neutron stars by exploiting the occultation of parts of the inner disk by the star itself. This occultation imprints characteristic features on the X-ray line profile that are unique and are expected to be present in low-mass X-ray binary systems seen under inclinations higher than ∼65°. We analyze a Nuclear Spectroscopic Telescope Array observation of a good candidate system, 4U 1636-53, and find that X-ray spectra from current instrumentation are unlikely to single out the occultation features owing to insufficient signal-To-noise. Based on an extensive set of simulations we show that large-Area X-ray detectors of the future generation could measure R/M to ∼2 ÷ 3% precision over a range of inclinations. Such is the precision in radius determination required to derive tight constraints on the equation of state of ultradense matter and it represents the goal that other methods also aim to achieve in the future
