    Microalgae cultivation for lipids and carbohydrates production

    Microalgae are photoautotrophic microorganisms that can produce energy both by using sunlight, water and CO2 (phototrophic metabolism) and by using organic sources such as glucose (heterotrophic metabolism). Heterotrophic growth is a key factor in microalgae research, due to its increased productivity and the lower capital and operative costs compared to photoautotrophic growth in photobioreactors. Carbohydrate production from microalgae is usually investigated for the production of biofuels (e.g. bioethanol) by successive fermentation, but also other applications can be envisaged in biopolymers. In this work an increment in carbohydrate purity after lipid extraction was found. Protein hydrolysis for different microalgae strains (Scenedesmus sp. and Chlorella sp.) was investigated. Microalgae were cultivated under photoautotrophic or heterotrophic conditions, collecting biomass at the end of the growth. Biomass samples were dried or freeze dried and used for carbohydrate and lipid extraction tests. Lipid extraction was achieved using different organic solvents (methanol-chloroform and hexane-2propanol). Basic protein hydrolysis has been carried out testing different temperatures and NaOH concentrations values. Lipids were spectrophotometrically quantified, while residual biomass was saccharificated and the total amount of sugars was measured. Significant differences about the purity of extracted carbohydrates were found comparing dried with freeze dried biomass. However, not a very promising purification of carbohydrates was achieved after protein hydrolysis, asking for further analysis. © Copyright 2017, AIDIC Servizi S.r.l

    An Audience-Centric Approach for Museums Sustainability

    therefore, in order to become sustainable, museums should pay attention to audience and its internal diversity. In this way, a cultural site can plan tailored strategies to increase the number of visits and re-visits and to achieve community support. For this reason it is necessary to understand the cultural needs of visitors, acquiring appropriate monitoring tools, such as qualitative and quantitative ones. Generally, quantitative analyses are more reliable and complete, even if they require a considerable number of observations for the reliability of the results. Moreover, qualitative analysis provides more in depth information, even if their data do not allow us to make generalizations. The qualitative and quantitative methods for the detection of satisfaction are usually used separately, but their integration may bring significant added value in terms of the wealth of information. This study follows the analysis of the potential of the integration of qualitative and quantitative analysis techniques customized with respect to different types of targets. The results of the experimentation performed on ethnographic museums shows a consistency of the results obtained by the two different tools that increase the capacity information of survey instruments

    Identification of the zinc finger 216 (ZNF216) in human carcinoma cells. A potential regulator of EGFR activity

    Epidermal Growth Factor Receptor (EGFR), a member of the ErbB family of receptor tyrosine kinase (RTK) proteins, is aberrantly expressed or deregulated in tumors and plays pivotal roles in cancer onset and metastatic progression. ZNF216 gene has been identified as one of Immediate Early Genes (IEGs) induced by RTKs. Overexpression of ZNF216 protein sensitizes 293 cell line to TNF-α induced apoptosis. However, ZNF216 overexpression has been reported in medulloblastomas and metastatic nasopharyngeal carcinomas. Thus, the role of this protein is still not clearly understood. In this study, the inverse correlation between EGFR and ZNF216 expression was confirmed in various human cancer cell lines differently expressing EGFR. EGF treatment of NIH3T3 cells overexpressing both EGFR and ZNF216 (NIH3T3-EGFR/ZNF216), induced a long lasting activation of EGFR in the cytosolic fraction and an accumulation of phosphorylated EGFR (pEGFR) more in the nuclear than in the cytosolic fraction compared to NIH3T3-EGFR cells. Moreover, EGF was able to stimulate an increased expression of ZNF216 in the cytosolic compartment and its nuclear translocation in a time-dependent manner in NIH3T3-EGFR/ZNF216. A similar trend was observed in A431 cells endogenously expressing the EGFR and transfected with Znf216. The increased levels of pEGFR and ZNF216 in the nuclear fraction of NIH3T3-EGFR/ZNF216 cells were paralleled by increased levels of phospho-MAPK and phospho-Akt. Surprisingly, EGF treatment of NIH3T3-EGFR/ZNF216 cells induced a significant increase of apoptosis thus indicating that ZNF216 could sensitize cells to EGF-induced apoptosis and suggesting that it may be involved in the regulation and effects of EGFR signaling

    Mir-34a-5p Mediates Cross-Talk between M2 Muscarinic Receptors and Notch-1/EGFR Pathways in U87MG Glioblastoma Cells: Implication in Cell Proliferation

    Glioblastoma (GBM) is the most aggressive human brain tumor. The high growth potential and decreased susceptibility to apoptosis of the glioma cells is mainly dependent on genetic amplifications or mutations of oncogenic or pro-apoptotic genes, respectively. We have previously shown that the activation of the M2 acetylcholine muscarinic receptors inhibited cell proliferation and induced apoptosis in two GBM cell lines and cancer stem cells. The aim of this study was to delve into the molecular mechanisms underlying the M2-mediated cell proliferation arrest. Exploiting U87MG and U251MG cell lines as model systems, we evaluated the ability of M2 receptors to interfere with Notch-1 and EGFR pathways, whose activation promotes GBM proliferation. We demonstrated that the activation of M2 receptors, by agonist treatment, counteracted Notch and EGFR signaling, through different regulatory cascades depending, at least in part, on p53 status. Only in U87MG cells, which mimic p53-wild type GBMs, did M2 activation trigger a molecular circuitry involving p53, Notch-1, and the tumor suppressor mir-34a-5p. This regulatory module negatively controls Notch-1, which affects cell proliferation mainly through the Notch-1/EGFR axis. Our data highlighted, for the first time, a molecular circuitry that is deregulated in the p53 wild type GBM, based on the cross-talk between M2 receptor and the Notch-1/EGFR pathways, mediated by mir-34a-5p

    The Relation between Collaborative Consumption and Subjective Well-Being: An Analysis of P2P Accommodation

    The present paper investigates the relationships between Collaborative Consumption (CC) and well-being. Specifically, the study aims to understand the antecedents of subjective well-being (SWB) in peer-to-peer (P2P) accommodation services. The research adopts a mixed-method approach by integrating qualitative (focus groups) and quantitative (survey) methods with sequential logic. A conceptual model was developed and validated through structural equation modelling (SEM). The model confirms that the SWB, which has three indirect antecedents (the social, environmental and economic dimensions), was positively influenced by the attitude towards P2P accommodation services, and the relation between this construct and SWB emphasizes the originality of the study. This research contributes to the current sharing economy debate, unveiling theoretical advances on the link between collaborative services and well-being
