    Rapid, quantitative determination of bacteria in water

    A bioluminescent assay for ATP in water borne bacteria is made by adding nitric acid to a water sample with concentrated bacteria to rupture the bacterial cells. The sample is diluted with sterile, deionized water, then mixed with a luciferase-luciferin mixture and the resulting light output of the bioluminescent reaction is measured and correlated with bacteria present. A standard and a blank also are presented so that the light output can be correlated to bacteria in the sample and system noise can be substracted from the readings. A chemiluminescent assay for iron porphyrins in water borne bacteria is made by adding luminol reagent to a water sample with concentrated bacteria and measuring the resulting light output of the chemiluminescent reaction

    The Importance of Phase in Nulling Interferometry and a Three Telescope Closure-Phase Nulling Interferometer Concept

    We discuss the theory of the Bracewell nulling interferometer and explicitly demonstrate that the phase of the "white light" null fringe is the same as the phase of the bright output from an ordinary stellar interferometer. As a consequence a "closure phase" exists for a nulling interferometer with three or more telescopes. We calculate the phase offset as a function of baseline length for an Earth-like planet around the Sun at 10 pc, with a contrast ratio of 10610^{-6} at 10 μ\mum. The magnitude of the phase due to the planet is 106\sim 10^{-6} radians, assuming the star is at the phase center of the array. Although this is small, this phase may be observable in a three-telescope nulling interferometer that measures the closure phase. We propose a simple non-redundant three-telescope nulling interferometer that can perform this measurement. This configuration is expected to have improved characteristics compared to other nulling interferometer concepts, such as a relaxation of pathlength tolerances, through the use of the "ratio of wavelengths" technique, a closure phase, and better discrimination between exodiacal dust and planets

    THESIS is a concept for a medium class mission designed for spectroscopic characterization of extrasolar planets between 2-14 microns. The concept leverages off the recent first-steps made by Spitzer and Hubble in characterizing the atmospheres of alien gas giants. Under favourable circumstances, THESIS is capable of identifying biogenic molecules in habitable-zone planets, thereby determining conditions on worlds where life might exist. By systematically characterizing many worlds, from rocky planets to gas-giants, THESIS would deliver transformational science of profound interest to astronomers and the general public

    C-myc amplification in breast cancer: a meta-analysis of its occurrence and prognostic relevance

    Data from basic research suggests that amplification of the proto-oncogene c-myc is important in breast cancer pathogenesis, but its frequency of amplification and prognostic relevance in human studies have been inconsistent. In an effort to clarify the clinical significance of c-myc amplification in breast cancer, we conducted a comprehensive literature search and a meta-analysis in which 29 studies were evaluated. The weighted average frequency of c-myc amplification in breast tumours was 15.7% (95% CI = 12.5–18.8%), although estimates in individual studies exhibited significant heterogeneity, P< 0.0001. C-myc amplification exhibited significant but weak associations with tumour grade (RR = 1.61), lymph-node metastasis (RR = 1.24), negative progesterone receptor status (RR = 1.27), and postmenopausal status (RR = 0.82). Amplification was significantly associated with risk of relapse and death, with pooled estimates RR = 2.05 (95% CI = 1.51–2.78) and RR = 1.74 (95% CI = 1.27–2.39), respectively. This effect did not appear to be merely a surrogate for other prognostic factors. These results suggest that c-myc amplification is relatively common in breast cancer and may provide independent prognostic information. More rigorous studies with consistent methodology are required to validate this association, and to investigate its potential as a molecular predictor of specific therapy response. © 2000 Cancer Research Campaign http://www.bjcancer.co

    The interaction of lean and building information modeling in construction

    Lean construction and Building Information Modeling are quite different initiatives, but both are having profound impacts on the construction industry. A rigorous analysis of the myriad specific interactions between them indicates that a synergy exists which, if properly understood in theoretical terms, can be exploited to improve construction processes beyond the degree to which it might be improved by application of either of these paradigms independently. Using a matrix that juxtaposes BIM functionalities with prescriptive lean construction principles, fifty-six interactions have been identified, all but four of which represent constructive interaction. Although evidence for the majority of these has been found, the matrix is not considered complete, but rather a framework for research to explore the degree of validity of the interactions. Construction executives, managers, designers and developers of IT systems for construction can also benefit from the framework as an aid to recognizing the potential synergies when planning their lean and BIM adoption strategies

    A new Search for Carbon Monoxide Absorption in the Transmission Spectrum of the Extrasolar Planet HD 209458b

    We have revisited the search for carbon monoxide absorption features in transmission during the transit of the extrasolar planet HD 209458b. We acquired 1077 high resolution spectra at 2 microns using NIRSPEC on Keck II during three transits. Our sensitivity is sufficient to test the degree of CO absorption in the first overtone bands during transit, based on plausible models of the planetary atmosphere. We compare to theoretical tangent geometry absorption spectra, computed by adding height-invariant ad hoc temperature pertubations to the model atmosphere of Sudarsky et al., and by treating cloud height as an adjustable parameter. We do not detect CO absorption. Our analysis indicates a weakening similar to the case of sodium, suggesting that a general masking mechanism is at work in the planetary atmosphere. If this masking is provided by high clouds, our analysis defines the maximum cloud top pressure (i.e., minimum height) as a function of the model atmospheric temperature. For the relatively hot model used by Charbonneau et al. to interpret their sodium detection, our CO limit requires cloud tops at or above 3.3 mbar, and these clouds must be opaque at a wavelength of 2 microns. High clouds comprised of submicron-sized particles are already present in some models, but may not provide sufficient opacity to account for our CO result. Cooler model atmospheres, having smaller atmospheric scale heights and lower CO mixing ratios, may alleviate this problem to some extent. However, even models 500K cooler that the Sudarsky et al. model require clouds above the 100 mbar level to be consistent with our observations. Our null result therefore requires that clouds exist at an observable level in the atmosphere of HD 209458b, unless this planet is dramatically colder than current belief.Comment: 29 pages, 8 figures, accepted for Ap
