    Biomechanical and Neuromuscular Changes in Jump Landings Due to Short or Long Term Ankle Bracing and Fatigue

    In order to prevent ankle sprain, prophylactic ankle bracing is common practice for many sports. The purpose of this dissertation was to investigate changes in loading and neuromuscular activation at the knee and at the hip when the ankle is braced. Additionally, fatigue is a known risk factor for both ankle sprain and anterior cruciate ligament (ACL) tear at the knee. The first two studies examined whether or not there was an interaction between the ankle brace and fatigue. Unwanted adaptation to long term bracing is also a concern. Thus, the third study addressed the question, does long term prophylactic ankle bracing change loading at the knee and hip? Furthermore, jump direction was specifically explored as a factor affecting loading of the hip and knee. Eight video cameras tracked jumping movements, two force platforms measured ground reaction forces, and a wireless electromyography (EMG) system detected muscle electrical activity. Knee and hip joint moments, loading rates, and joint moment impulses were used as kinetic dependent variables. EMG was used to quantify muscle activation as a percentage of maximum voluntary contraction for the vastus lateralis, biceps femoris, and gluteus maximus. Synthesizing the results of the three studies, the most significant findings were the differences in jump direction. They present a consistent picture of what would appear to be a more forceful jump in the forward direction and a more tentative jump in the backward direction. Fatigue did not interact with the braces, nor did it significantly affect the kinetics of the landings. However, it did have an effect on the activation of gluteal muscles, which may indicate a need to train those muscles in order to prevent injury. Results of the first and third studies do not indicate increased loading at the knee as an immediate effect of the ankle brace. Increased hip extension moment and hip extension moment impulse for the habitual bracers in the third study may indicate a proximal shift of shock absorption may be due to long-term bracing, or due to increased athleticism of the habitual brace group, which consisted of an NCAA division one volleyball team

    Expression of three GnRH receptors in specific tissues in male and female sea lampreys Petromyzon marinus at three distinct life stages

    Two recently cloned gonadotropin-releasing hormone (GnRH) receptors (lamprey GnRH-R-2 and lamprey GnRH-R-3) along with lamprey (l) GnRH-R-1 were shown to share similar structural features and amino acid motifs common to other vertebrate receptors. Here we report on our findings of RNA expression of these three GnRH receptors in the three major life stages (larval, parasitic, and adult phases) of the sea lamprey, Petromyzon marinus, a basal vertebrate. For each stage, we examined the expression of messenger RNA encoding the receptors in the brain, pituitary, gonad, heart, muscle, liver, eye, intestine, kidney, skin, thyroid, gill, and endostyle by RT-PCR. In adult lampreys, the spatial expression of the three receptors in the brain and pituitary was investigated by in situ hybridization. In general, the receptors were more widely expressed in adult tissues as compared to parasitic-phase tissues and least widely expressed in the larval tissues. There were noted differences in male and female lampreys in the adult and parasitic phases for all three receptors. The data showed the presence of all three receptor transcripts in brain tissues for adult and parasitic phases and all three receptor transcripts were expressed in the adult pituitaries, but not in the parasitic pituitaries. However, in the larval phase, only lGnRH-R-1 was expressed in the larval brain and pituitary. In situ hybridization revealed that lGnRH-R-2 and -3 were expressed in the pineal tissue of adult female lampreys while lGnRH-R-1 was expressed in the pineal in adult male lampreys, all restricted to the pineal pellucida. In summary, these data provide an initial comparative analysis of expression of three lamprey GnRH receptors suggesting differential regulation within males and females at three different life/reproductive stages

    Dose Duration Effects of 17-α Ethynylestradiol in Zebrafish Toxicology

    Exposure of zebrafish to the synthetic estrogen 17-α ethynylestradiol (EE2) has been shown to cause a number of detrimental effects, including but not limited to feminization of male fish, reduced reproductive capabilities, and impaired embryonic development. This paper systematically reviews the effects of five environmentally relevant concentrations of EE2 on 12 measurements that are commonly selected when studying the effects of EE2 on zebrafish. Concentrations of 0.1 ng EE2/L, 1 ng EE2/L, 3 ng EE2/L, 10 ng EE2/L, 25 ng EE2/L, and 100 ng EE2/L were reviewed for their effects on sex ratio, vitellogenin induction, gonad morphology, spawning success, survival, bodily malformation, length/weight, swim-up success, fecundity, fertilization success, hatching success, and the reversibility of aforementioned effects. A greater occurrence of effects was observed as the dose of EE2 was increased, starting at exposure levels of 1 ng EE2/L. For exposures of 3 and 10 ng EE2/L, negative effects on sex ratio, morphology, and reproductive capabilities were reversible after zebrafish were able to recover in clean water for a period of time. Data for zebrafish exposed to 100 ng EE2/L was limited, as this concentration severely decreased survival
