No full textNational audienceUne étude de synthèse totale d’un alcaloïde appelé koumine a conduit à la découverte d’une séquence réactionnelle de type domino par réaction d’un nucléophile lithié avec l’aldéhyde 2-nitroarylcyclohexane-1,3-dione.Alors que nous attendions la formation de l’alcool, la réaction ne s’est pas arrêtée à ce stade et plusieurs liaisons furent formées et cassées au sein même de la molécule dans une séquence dirigée et facilitée par des groupements électroattracteurs, aboutissant au final à un composé lactonique tendu à neuf chainons.Cette séquence réactionnelle est de type domino, illustrant une séquence d’évènements à partir d’un déclencheur, ici l’addition nucléophile. Lors de l’étude, la régiosélectivité et la diastéréosélectivité du processus furent analysées pour aboutir à la synthèse concise de lactones polysubstituées sans étape de lactonisation
Progresses for accessing alpha’-methoxy-y-pyrone heterocycle: Applications to the synthesis of verticipyrone and aureothin
No full textInternational audienceFocusing on the synthesis and reactivity of alpha’-methoxy-y-pyrone scaffold, a heterocycle found in several natural products, this account presents the state of the art and the progresses made to access it. Arecently developed abiotic strategy relied on the desymmetrization of alpha,alpha’-dimethoxy--pyrone, a readily available building block, by conjugate addition with a nucleophile facilitating the construction of the side chain of the target. This transformation was studied with various nucleophiles and the strategy was successfully employed with tributylallylstannane/n-BuLi and 2-lithio-1,3-dithiane to concisely fashion verticipyrone and aureothin, two natural products with interesting biological activities
