    Human NK cells internalize recombinant major stress protein HSP70

    Heat shock proteins 70 kDa (HSP70) protect intracellular proteins from the damaging effects of stress factors of various natures. Moreover, HSP70 play an important role in the vital activity of cells under normal physiological conditions, performing chaperone functions. These functions are realized in the intracellular space; however, in some cases, these proteins are also found on the cell surface and in the extracellular environment. The causes and mechanisms of HSP70 translocation to the cell surface and secretion into the extracellular space have not yet been well understood, but such an unusual localization of HSP70 activates the immune system. The surface HSP70 and their extracellular pool stimulate the cytotoxic activity of NK cells. However, direct experimental evidence for the internalization of HSP70 molecules by NK cells has not yet been demonstrated. This paper presents the results of the interaction of the extracellular HSP70 pool with NK cells from the peripheral blood. The results demonstrated the confirmation of the internalization of exogenous HSP70 molecules by NK cells. To this end, fluorescently labeled recombinant stress-inducible human HSP70 were obtained. The electrophoretic data indicated the absence of protein degradation during the labeling process, the purity and stability of the modified protein. To assess the interaction of HSP70 with NK cells, the fluorescently labeled HSP70 was added to an in vitro culture of NK cells isolated by magnetic separation from the peripheral blood mononuclear fraction and analyzed by confocal microscopy. This analysis indicated that living NK cells internalize extracellular HSP70 with localization both in lysosomes and in phagosomes. Our experiments illustrated for the first time the process of penetration of the extracellular form of HSP70 into these cells. The results suggest that the activation of NK cells under the action of exogenous HSP70 could be associated with the internalization of these protein molecules

    Objectives. To study the hydrogenation of substituted 5-acyl-1,3-dioxanes in the presence of metal-containing catalysts (Pt/Re, Pd/C, Ni/kieselguhr, and Ni/Mo).Methods. In order to determine the qualitative and quantitative composition of the reaction masses, the following analysis methods were used: gas-liquid chromatography (using the Kristall 2000 hardware complex); mass-spectroscopy (using Chromatec-Kristall 5000M device with NIST 2012); nuclear magnetic resonance (NMR) spectrometry (using Bruker AM-500 device with operating frequencies of 500 and 125 MHz).Results. Hydrogenation of substituted 5-acyl-1,3-dioxanes obtained by condensation of carbonyl compounds with paraformaldehyde and sulfuric acid was used to synthesize heterocyclic alcohols in the presence of metal-containing catalysts with a conversion of the initial ketones of 60–90% and a formation selectivity of target products of 70–90%. Substances were analyzed and confirmed by gas-liquid chromatography, mass spectrometry and NMR spectroscopy.Conclusions. The best catalyst for the reduction of substituted 5-acyl-1,3-dioxanes is Pd/C. By using this catalyst, it is possible to achieve a high selectivity for the formation of the corresponding heterocyclic alcohols at a conversion rate of the initial ketones of 60–90%.Цели. Изучить гидрирование замещенных 5-ацил-1,3-диоксанов в присутствии металлосодержащих катализаторов (Pt/Re, Pd/C, «Ni на кизельгуре», Ni/Mo).Методы. Для определения качественного и количественного состава реакционных масс были использованы следующие методы анализа: газожидкостная хроматография (на аппаратно-программном комплексе «Кристалл 2000»), масс-спектроскопия (на приборе «Хроматэк-Кристалл 5000М» с базой NIST 2012), и спектроскопия ядерного магнитного резонанса (ЯМР-спектроскопия) (на приборе «BrukerAM-500» с рабочими частотами 500 и 125 МГц).Результаты. Гидрированием замещенных 5-ацил-1,3-диоксанов, полученных конденсацией карбонильных соединений с параформом с использованием серной кислоты, синтезированы гетероциклические спирты в присутствии металлосодержащих катализаторов с конверсией исходных кетонов 60–90% и селективностью образования целевых продуктов 70–90%. Вещества проанализированы и доказаны методами газожидкостной хроматографии, масс-спектрометрии и ЯМР-спектроскопии.Выводы. Установлено, что лучшим катализатором восстановления замещенных 5-ацил-1,3-диоксанов является Pd/C, позволяющий достичь высокой селективности образования соответствующих гетероциклических спиртов при конверсии исходных кетонов 60–90%

    Raw experimental data on heat transfer from the wall are presented in the form of electric signals (voltage in V). The data were collected from 42 measurement tracks; 1 reference resistor 226 Ω in the temperature measurement circuit with a resistance thermometer UTR; 1 signal from resistance thermometer UT; 4 signals from 4 reference resistors 5 Ω in circuits of measurement elements UR 10 1 - UR 10 4. Measurement elements Ri corresponded to the following channels: - R1 – R10 – channels 32 to 23; - R11 – R26 – channels 48 to 33; - R27 – R42 – channels 16 to 1; - UT – channel 17; - UTR – channel 20; - UR 10 1 – UR 10 4 – channels 18, 19, 21, 22. Elements R1 – R42 were placed on the wall with a streamwise spacing of 9.5 mm. Voltage drop measured across the reference resistors yielded the magnitude of electric current in the respective circuits. Each file contains the data for one regime of air flow. File names specify the volumetric flow rate (Q, m3/h) and forcing frequency (f, Hz). R0i file contains signals for estimation of reference resistances of measurement elements at 23°С
