    Investigating intra-host and intra-herd sequence diversity of foot-and-mouth disease virus

    Due to the poor-fidelity of the enzymes involved in RNA genome replication, foot-and-mouth disease (FMD) virus samples comprise of unique polymorphic populations. In this study, deep sequencing was utilised to characterise the diversity of FMD virus (FMDV) populations in 6 infected cattle present on a single farm during the series of outbreaks in the UK in 2007. A novel RT–PCR method was developed to amplify a 7.6 kb nucleotide fragment encompassing the polyprotein coding region of the FMDV genome. Illumina sequencing of each sample identified the fine polymorphic structures at each nucleotide position, from consensus level changes to variants present at a 0.24% frequency. These data were used to investigate population dynamics of FMDV at both herd and host levels, evaluate the impact of host on the viral swarm structure and to identify transmission links with viruses recovered from other farms in the same series of outbreaks. In 7 samples, from 6 different animals, a total of 5 consensus level variants were identified, in addition to 104 sub-consensus variants of which 22 were shared between 2 or more animals. Further analysis revealed differences in swarm structures from samples derived from the same animal suggesting the presence of distinct viral populations evolving independently at different lesion sites within the same infected animal

    Using a fuzzy inference system to control a pumped storage hydro plant

    The paper discusses the development of a fuzzy inference system (FIS) based governor control for a pumped storage hydroelectric plant. The First Hydro Company's plant at Dinorwig in North Wales is the largest of its kind in Europe and is mainly used for frequency control of the UK electrical grid. In previous investigations, a detailed model of the plant was developed using MATLAB(R)/SIMULINK(R) and this is now being used to compare FIS governor operation with the proportional-integral-derivative (PID) controller currently used. The paper describes the development of an FIS governor, and shows that its response to a step increase in load is superior to the PID under certain conditions of load. The paper proceeds to discuss the implications of these results in view of the possible practical application of an FIS governor at the Dinorwig plant

    Military service and crime: new evidence

    BACKGROUND: Evidence indicates that a substantial proportion of military personnel are involved in high-risk and antisocial behaviors that place them at jeopardy for criminal justice system involvement. However, prior research on military service and crime has disproportionately focused on veterans from the Vietnam War era (1955–1975), and has tended to focus on either current or former military members. METHODS: This study employed data from a population-based study (i.e., National Study on Drug Use and Health [NSDUH] between 2002 and 2014). It systematically examines the prevalence of self-reported antisocial behaviors, criminal justice system involvement, and substance abuse among the US civilian population and military service members, including reservists (n = 2206) and those who reported having been separated or retired from military service (n = 20,551). These factors are further examined across the developmental spectrum of adulthood (ages 18–34, 35–49, and 50–64). RESULTS: Results showed that military members were more prone to lifetime arrests and overall substance misuse. However, additional findings emerged suggesting that, while the military population overall seems to be positively associated with higher criminal activity than that found in the civilian population, these findings were based on a specific subgroup of the veteran population. This subgroup is comprised of individuals who likely did not fit in with the military culture and were discharged from the military early in their careers. CONCLUSION: Additional research on identifying this subgroup of military members is encouraged to better concentrate on prevention and treatment measures

    Electrophoretic Pattern of Human Hairbulb Tyrosinase

    A small gel electrophoretic system has been developed for the study of tryosinase from human hairbulb melanocytes. Enzyme is released from the hairbulb with Triton X-100. Electrophoresis is run in 60mm microcapillary pipettes with an internal diameter of 1.5mm. With this technique, tyrosinase from normally pigmented brown, black, blond, and red hairbulbs gives a single band when the gel is stained with L-dopa for enzyme activity. Tryosinase from tryosinase-positive oculocutaneous albino and Hermansky-Pudlak syndrome hairbulbs gives a single L-dopa band. Neuraminidase pretreatment of the tyrosinase from normal and albino hairbulbs changes the migration of the tyrosinase band to a less anodal position. Trypsin pretreatment has no effect. We conclude that human hairbulb tyrosinase is a glycoprotein and that tyrosinase-positive albino tyrosinase has no major electrophoretic difference from normal enzyme

    A to Z of the Muon anomalous magnetic moment in the MSSM with Pati-Salam at the GUT scale

    We analyse the low energy predictions of the minimal supersymmetric standard model (MSSM) arising from a GUT scale Pati-Salam gauge group further constrained by an A4 × Z5 family symmetry, resulting in four soft scalar masses at the GUT scale: one left-handed soft mass m0 and three right-handed soft masses m1, m2, m3, one for each generation. We demonstrate that this model, which was initially developed to describe the neutrino sector, can explain collider and non-collider measurements such as the dark matter relic density, the Higgs boson mass and, in particular, the anomalous magnetic moment of the muon (g − 2)ÎŒ. Since about two decades, (g − 2)ÎŒ suffers a puzzling about 3σ excessoftheexperimentallymeasuredvalueoverthetheoreticalprediction,whichour model is able to fully resolve. As the consequence of this resolution, our model predicts specific regions of the parameter space with the specific properties including light smuons and neutralinos, which could also potentially explain di-lepton excesses observed by CMS and ATLAS

    BACKGROUND: Acute upper respiratory tract infections (URTIs), including the common cold and rhinosinusitis, are common afflictions that cause discomfort and debilitation and contribute significantly to workplace absenteeism. Treatment is generally by antipyretic and decongestant drugs and sometimes antibiotics, even though most infections are viral. Nasal irrigation with saline is often employed as an adjunct treatment for URTI symptoms despite a relative lack of evidence for benefit in this clinical setting. This review is an update of the Cochrane review by Kassel et al, which found that saline was probably effective in reducing the severity of some symptoms associated with acute URTIs
