Physical activity improves sleep quality in women
Objectives: The aim of the study was to check whether women with a higher level of physical activity are less likely to experience sleep problems.Material and methods: 80 women aged 45–65 from Silesia took part in the pilot study. The research tool was a self-survey, the International Physical Activity Questionnaire (IPAQ), the Athens Insomnia Scale (AIS), and the Menopause Rating Scale (MRS). The data was prepared in the STATISTICA 10 program.Results: The mean age of the respondents was 51.75 ± 5.57. The most common symptoms were psychological problems (mean MRS: 4.29 ± 3.25).A sedentary lifestyle was reported in 57.14% of the respondents. There was a significant difference between women suffering from insomnia and women with normal sleep in terms of the level of physical activity (p = 0.025).Conclusions: Physical activity significantly affects the quality of sleep among middle-aged women
Recommendations for the use of simulation methods in a selected area of health sciences based on an example simulation scenario
Over the past two decades, there has been rapid and enthusiastic acceptance of the simulation in medical education on the international stage. Poland compared to other countries has become unique due to the possibility of developing simulation and education by training teachers who teach medical professions, and building simulation centres.One of the most important milestones in the development of the curriculum in medical and health sciences is the introduction of a simulation-based teaching method. Active teaching methods, including medical simulation, create conditions for independent learning,so they should be permanently included in the canon of education methods in the field of health sciences
MCPIP1 functions as a safeguard of early embryonic development
Monocyte chemoattractant protein-induced protein 1 (MCPIP1), also called Regnase-1, is an RNase that has been described as a key negative modulator of inflammation. MCPIP1 also controls numerous tumor-related processes, such as proliferation, apoptosis and differentiation. In this study, we utilized a zebrafish model to investigate the role of Mcpip1 during embryogenic development. Our results demonstrated that during embryogenesis, the expression of the zc3h12a gene encoding Mcpip1 undergoes dynamic changes. Its transcript levels gradually increase from the 2-cell stage to the spherical stage and then decrease rapidly. We further found that ectopic overexpression of wild-type Mcpip1 but not the catalytically inactive mutant form resulted in an embryonic lethal phenotype in zebrafish embryos (24 hpf). At the molecular level, transcriptomic profiling revealed extensive changes in the expression of genes encoding proteins important in the endoplasmic reticulum stress response and in protein folding as well as involved in the formation of primary germ layer, mesendoderm and endoderm development, heart morphogenesis and cell migration. Altogether, our results demonstrate that the expression of zc3h12a must be tightly controlled during the first cell divisions of zebrafish embryos and that a rapid decrease in its mRNA expression is an important factor promoting proper embryo development
Functional gene expression profile underlying methotrexate-induced senescence in human colon cancer cells
Cellular functions accompanying establishment of premature senescence in methotrexate-treated human colon cancer C85 cells are deciphered in the present study from validated competitive expression microarray data, analyzed with the use of Ingenuity Pathways Analysis (IPA) software. The nitrosative/oxidative stress, inferred from upregulated expression of inducible nitric oxide synthase (iNOS) and mitochondrial dysfunction-associated genes, including monoamine oxidases MAOA and MAOB, β-amyloid precursor protein (APP) and presenilin 1 (PSEN1), is identified as the main determinant of signaling pathways operating during senescence establishment. Activation of p53-signaling pathway is found associated with both apoptotic and autophagic components contributing to this process. Activation of nuclear factor κB (NF-κB), resulting from interferon γ (IFNγ), integrin, interleukin 1β (IL-1β), IL-4, IL-13, IL-22, Toll-like receptors (TLRs) 1, 2 and 3, growth factors and tumor necrosis factor (TNF) superfamily members signaling, is found to underpin inflammatory properties of senescent C85 cells. Upregulation of p21-activated kinases (PAK2 and PAK6), several Rho molecules and myosin regulatory light chains MYL12A and MYL12B, indicates acquisition of motility by those cells. Mitogen-activated protein kinase p38 MAPK β, extracellular signal-regulated kinases ERK2 and ERK5, protein kinase B AKT1, as well as calcium, are identified as factors coordinating signaling pathways in senescent C85 cells

Acriflavine, a clinically approved drug, inhibits SARS-CoV-2 and other betacoronaviruses
The COVID-19 pandemic caused by SARS-CoV-2 has been socially and economically devastating. Despite an unprecedented research effort and available vaccines, effective therapeutics are still missing to limit severe disease and mortality. Using high-throughput screening, we identify acriflavine (ACF) as a potent papain-like protease (PLpro) inhibitor. NMR titrations and a co-crystal structure confirm that acriflavine blocks the PLpro catalytic pocket in an unexpected binding mode. We show that the drug inhibits viral replication at nanomolar concentration in cellular models, in vivo in mice and ex vivo in human airway epithelia, with broad range activity against SARS-CoV-2 and other betacoronaviruses. Considering that acriflavine is an inexpensive drug approved in some countries, it may be immediately tested in clinical trials and play an important role during the current pandemic and future outbreaks. © 2021 The Author
