A New Phytochemical Screening Programme used for Crops grown with Organic and Conventional Methods
A broad screening programme, covering the most general phytochemical groups of compounds, was developed on the basis of Thin Layer Chromatography (TLC). A total of 46 TLC systems, comprising 26 derivatization reagents, 3 stationary phases, and 4 mobile phases, were included. The TLC systems were classified according to the groups of phytochemical compounds detected: Alcohols and phenolic compounds; Carbohydrates; N-containing compounds; Organic acids and lipids; P-containing compounds; S-containing compounds, and Terpenoids. Furthermore, one group of TLC systems detected compounds from several of the mentioned groups.
The screening programme was applied in the screening of potatoes (S. tuberosum L.), peas (P. sativum L.), kale (B. oleracea L.), carrots (D. carota L.), and apples (M. domestica Borkh.), cultivated with combinations of organic and conventional methods for plant protection and nutrient supply, for phytochemical differences (biomarkers).
Distinctive phytochemical differences were found between the differently cultivated samples of these crops. In peas and carrots only one biomarker was found. In peas the biomarker was related to the soil conditions, while the biomarker in carrots was related to the use of pesticides. In potato, two biomarkers related to the use of pesticides were found. Three biomarkers were found in kale. Two of these could be related to the use of pesticide, while the last was related to either fertiliser or soil conditions. Several biomarkers were found apples, but a relation to the cultivation methods was not clear. Three of the biomarkers in apples could be related to either the use of pesticides or fertiliser, while no conclusions could be drawn from the other biomarkers found.
The results of the screening programme form the basis for a potential development of a kit to detect whether crops are organically- or conventionally cultivated. Furthermore, the results from this part and other parts of the project "Organic food and health â a multigenerational animal experiment" provide basis for the selection of which secondary compounds to quantify by specific chemical analysis, isolate, and/or structure elucidation
Kristallstrukturanalyse und Entwicklung von Computermodellen zur Beschreibung der SelektivitÀt von Enzymen am Beispiel der Carboanhydrase
Ein Ziel der modernen Wirkstoffforschung ist die Entwicklung von
Arzneistoffen, die selektiv mit einem Zielprotein wechselwirken.
Dadurch sollen Nebenwirkungen in einem frĂŒhen Stadium der
Entwicklung minimiert, wenn möglich sogar ausgeschlossen werden.
Bei der strukturbasierten Entwicklung von Wirkstoffen wurden im
Falle der Carboanhydrase (CA) die physikochemischen
Eigenschaften von CA-Inhibitoren mit den biologischen AffinitÀten
an drei Isoenzymen (CA I, CA II, CA IV) korreliert. Mit den daraus
erhaltenen 3D-QSAR-Modellen können diejenigen physikochemischen
und strukturellen Eigenschaften der Verbindungen bestimmt werden,
die die biologische AktivitÀt im Hinblick auf ein Isoenzym
beeinflussen.
Mit zwei verschiedenen AnsÀtzen wurden selektive
3D-QSAR-Modelle erhalten, mit deren Hilfe anhand
von Konturdiagrammen Bereiche ermittelt werden können, die fĂŒr
eine bestimmte physikochemische Eigenschaft (sterische, elektrostatische, hydrophobe, Akzeptor, Donor) die SelektivitÀt
hinsichtlich eines Isoenzyms erhöhen oder erniedrigen. Aus dieser
reinen Liganden-basierten Analyse konnten dennoch RĂŒckschlĂŒsse auf
die Gegebenheiten in der umgebenden Bindetasche gezogen werden.
Aber nicht nur die Auswertung von Ligandeninformationen kann
wichtige Hinweise fĂŒr die Entwicklung selektiver CA-Inhibitoren
liefern. Die Auswertungen von Kristallstrukturen der CA
bestÀtigten diese aus reiner Ligandeninformation erhaltenen
SelektivitĂ€tsmodelle und lieferten zusĂ€tzliche Informationen fĂŒr
die Entwicklung selektiver Verbindungen. Ferner konnte anhand eines
Homologiemodells der C AIX gezeigt werden, dass die
Modellierung von Enzymen, deren KristallstrukturaufklÀrung sich
als schwierig erweist, durchaus nĂŒtzliche Hinweise fĂŒr den
strukturbasierten Entwurf bzw. die Optimierung von Liganden
liefern kann.
Um potentielle neue (selektive) Leitstrukturen zu finden, können
neben dem experimentellen Hochdurchsatz-Screening virtuell am
Computer Datenbanken mit gespeicherten MolekĂŒlbibliotheken
durchmustert werden. Die korrekte Vorhersage des
Bindungsmodus und die korrekte AbschÀtzung der
BindungsaffinitĂ€t der platzierten MolekĂŒle ist entscheidend fĂŒr
die erfolgreiche Identifizierung von neuen Leitstrukturen. Es konnte erfolgreich gezeigt werden, dass
fĂŒr CA-Inhibitoren mit einem rigiden RinggerĂŒst und mit bekannter
Kristallstruktur der Bindungsmodus korrekt vorhergesagt werden
konnte. In den FĂ€llen, in denen die Bewertungsfunktionen der
Dockingprogramme in der AffiniÀtsvorhersage der Verbindungen
fehlschlagen, konnte durch die Kombination mit den zuvor
erhaltenen 3D-QSAR-Modellen die AffinitÀt dieser gedockten
Verbindungen richtig abgeschĂ€tzt werden. DarĂŒber hinaus wurde in
Datenbanken und in der Literatur nach potentiellen neuen
CA-Inhibitoren gesucht und ihre Bindung lieĂ sich biochemisch in
einem Assay bestÀtigen. Eine Platzierung in der Bindetasche von
CA II ermöglicht die korrekte AbschÀtzung der AffinitÀt.
Bei der Optimierung von Leitstrukturen/Wirkstoffen ist es sehr
schwierig Wechselwirkungen mit Proteinen vorherzusagen
beziehungsweise abzuschÀtzen, die sich in ihrer Sequenz und
Funktion stark von dem Ausgangsprotein unterscheiden. Es konnte gezeigt werden, dass
Cyclooxygenase (COX)-2-selektiven Verbindungen (Valdecoxib, Celecoxib) ebenfalls CA hemmen.
Obwohl beide Enzyme ? COX und CA - unterschiedliche biologische Funktionen besitzen, weisen
beide Enzyme lokale Bereiche im aktiven Zentrum mit Àhnlichen
physikochemischen Eigenschaften auf, so dass die Bindung von
Celecoxib und Valdecoxib an beiden Enzymen ermöglicht wird. Diese KreuzreaktivitÀt von
Celecoxib und Valdecoxib wurden durch
kinetische Daten und mit der
Kristallstruktur von Celecoxib im Komplex mit CA II belegt
Zehn Jahre differenzierte Grundbodenbearbeitung im ökologischen Ackerbau (Projekt Ăkologische Bodenbewirtschaftung) - Entwicklung der organischen Bodensubstanz, NĂ€hrstoffgehalte sowie bodenbiologischen Eigenschaften
This study investigated the lasting impact of reduced and conservation tillage, namely two-layer ploughing (LP) and layer-cultivating (LC) on soil ecological properties (physical, chemical, microbiological) in an organic farming system relative to ploughing (P). As a major trend, the amounts of soil organic C, microbial biomass âC and âN, Cmic to Corg ratio, and microbial activity increased in 0-15cm depth in the order P âLP â LC while they decreased in 15-25cm depth in the same order, both in green fallow and winter rye plots. For example, in 0-15 cm depth soil microbial biomass âC concentration was significantly 21 % (LP) and 30 % (LC) greater in green fallow plots or 15 % to 23 % greater in winter rye plots than in P. This was highly correlated with an increase in aggregate stability at these sites. Non-ploughing also resulted in a significant higher bulk density. In addition, there was much evidence that the microbial community changed towards fungi in 0-15 cm depth in reduced and conservation tillage plots. The abundance and fresh biomass of earthworms as well as species richness seemed to benefit from reduced soil tillage intensity. The highest amounts of abundance, fresh biomass and numbers of earthworm species in the current study were found in the plots with the layer cultivation treatment. No tillage effects were found for the amount of plant available P in soil. It can be concluded that reduced or conservation tillage in organic farming systems has beneficial effects on soil organic matter, microbial biomass and potential activity, as well as on earthworms
Die Bedeutung von Myo5 fĂŒr das polare Wachstum, die pathogene Entwicklung und den Transport polarer Chitinsynthasen in Ustilago maydis
Der phytopathogene Pilz Ustilago maydis ist der
Erreger des Maisbeulenbrandes. Die Bildung des infektiösen
dikaryotischen Filaments setzt die Fusion zweier kompatibler
Sporidien voraus, die stimuliert durch das Pheromon des
Partners Konjugationshyphen ausbilden, aufeinander zu wachsen
und miteinander fusionieren. Sowohl fĂŒr den Kreuzungsprozess
als auch fĂŒr die erfolgreiche Infektion der Wirtspflanze ist
die FĂ€higkeit des Pilzes, polar zu wachsen, von essentieller
Bedeutung. Polares Wachstum erfordert die gerichtete
Anlieferung von Wachstums- und Zellwandkomponenten entlang des
Cytoskeletts zur Wachstumsspitze hin. In der vorliegenden
Arbeit konnte gezeigt werden, dass das Klasse V Myosin, Myo5,
fĂŒr verschiedene Stadien des Pilzes wĂ€hrend seines sexuellen
Lebenszyklus von enormer Bedeutung ist. Myo5 ist essentiell fĂŒr
die erfolgreiche Perzeption des Pheromons und die Ausbildung
von Konjugationshyphen. Auch das Wachstum dikaryotischer Hyphen
ist in myo5ts-Mutanten gestört. Zudem zeigten
myo5ts-Infektionshyphen deutliche Störungen des polaren
Wachstums wĂ€hrend der frĂŒhen Infektionsphase, was sich in der
Ausbildung geschwollene Hyphenverzweigungen Ă€uĂerte, und sie
induzierten bereits bei permissiver Temperatur nur bei 2,7% der
Pflanzen Tumore. Aufgrund der aus Vorversuchen resultierenden
Erkenntnisse wurden Chitinsynthasen als mögliche
?Cargos? von Myo5 in Betracht gezogen und nÀher
untersucht. Das Genom von U. maydis kodiert fĂŒr sieben
Chitinsynthasen (Chs1-7) und eine Myosin-Chitinsynthase (Mcs1).
Sowohl die PhÀnotypen der Chitinsynthase-DeletionsstÀmme wurden
nÀher analysiert, als auch in vivo Lokalisationsstudien der
Chitinsynthasen durchgefĂŒhrt. Dabei zeigte sich, dass Dchs5-
und auch Dchs7-Zellen Àhnliche Zelltrennungsdefekte wie
Dmyo5-Zellen sowie Störungen in der Ausbildung von
Konjugationshyphen aufwiesen. Zudem fĂŒhrte die Deletion von
chs7 zu einer starken BeeintrÀchtigung der Filamentbildung auf
aktivkohlehaltigem Medium und der PathogenitÀt. Die Deletion
von chs6 und mcs1 störte die Filamentbildung kompatibler StÀmme
nicht, resultierte allerdings in einer völligen ApathogenitÀt.
Ăhnlich wie Myo5 lokalisierten die vier Chitinsynthasen Chs5,
Chs6, Chs7 und Mcs1 in der Spitze wachsender Sporidien.
Inhibitorversuche ergaben, dass die Lokalisation dieser polaren
Chitinsynthasen von Aktin abhÀngt. Untersuchungen zur
Lokalisation von Chs5, Chs6, Chs7 und Mcs1 in myo5ts-Mutanten
zeigten eine deutliche Fehlverteilung von Chs7 in
myo5ts-Mutanten nach einstĂŒndiger Inkubation bei restriktiver
Temperatur. Durch den neu etabliertem in vivo Aktinmarker
Fim1GFP konnte eine Störung des Aktin-Cytoskeletts als Ursache
ausgeschlossen werden. Sowohl die phÀnotypischen Analysen als
auch die Lokalisations- und Inhibitorstudien weisen deutlich
auf eine Beteiligung von Myo5 an der Lokalisation von Chs7
hin
Theorising creative industry management: rebooting the woolly mammoth
Purpose â The purpose of this paper is to highlight the paradox inherent in the term âCreative Industry Managementâ. The challenges of applying creative industry experiences within a managerial context are explored through a careful selection of papers that identify linkages between creative industry practice and management theories. Design/methodology/approach â The paper is a narrative commentary using the Jurassic Park franchise to highlight the potential application of management theory to explain the success or failure of a creative venture. The analogy of filmmaking and the creation of blockbuster movie sequels is useful in determining the tensions between creative production and management of a profitable franchise. Findings â This paper identifies opportunities for theory building at the meso level in the management domain, born of the experiences of those in the creative industries. The papers presented add to the continuing discovery of ways of applying management theory in the creative industries. However, there remain opportunities for a cross-pollination of theory from the creative industry to management domains. Research limitations/implications â It is not possible to claim more than observation and exploratory inference from the selection of papers presented. The special issue has only uncovered one half of the theoretical perspective, namely management theory that can be applied in the creative industries. Originality/value â The approach taken to liken the challenge of managing creativity to the production of creative works in moviemaking is novel and highlights the dearth of creative industry experiences that currently influence management theory
Ertrag und QualitÀt von Winterweizengenotypen unter integrierten und ökologischen Anbaubedingungen
An annually identical assortment of each of 32 winter wheat varieties and lines bred
under conventional conditions was tested between 2004 and 2007 both in a
conventionally and in an organically managed environment. Previously the assortment
had been evaluated with respect to yield and indirect parameters of baking quality.
The objective was to obtain evidence for a targeted selection of lines for their later use
as bread varieties in organic farming. It was discovered that for a successful selection
of lines for the subsequent cultivation under organic conditions especially the
evaluation of yield must be conducted strictly under the same conditions

Chemical characterization of water-soluble organic carbon aerosols at a rural site in the Pearl River Delta, China, in the summer of 2006
Online measurements of water-soluble organic carbon (WSOC) aerosols were made using a particle-into-liquid sampler (PILS) combined with a total organic carbon (TOC) analyzer at a rural site in the Pearl River Delta region, China, in July 2006. A macroporous nonionic (DAX-8) resin was used to quantify hydrophilic and hydrophobic WSOC, which are defined as the fractions of WSOC that penetrated through and retained on the DAX-8 column, respectively. Laboratory calibrations showed that hydrophilic WSOC (WSOCHPI) included low-molecular aliphatic dicarboxylic acids and carbonyls, saccharides, and amines, while hydrophobic WSOC (WSOCHPO) included longer-chain aliphatic dicarboxylic acids and carbonyls, aromatic acids, phenols, organic nitrates, cyclic acids, and fulvic acids. On average, total WSOC (TWSOC) accounted for 60% of OC, and WSOCHPO accounted for 60% of TWSOC. Both WSOC HIP and WSOCHPO increased with photochemical aging determined from the NOx/NOy ratio. In particular, the average WSOCHPO mass was found to increase by a factor of five within a timescale of âŒ10 hours, which was substantially larger than that of WSOCHPI (by a factor of 2-3). The total increase in OC mass with photochemical aging was associated with the large increase in WSOCHPO mass. These results, combined with the laboratory calibrations, suggest that significant amounts of hydrophobic organic compounds (likely containing large carbon numbers) were produced by photochemical processing. By contrast, water-insoluble OC (WIOC) mass did not exhibit significant changes with photochemical aging, suggesting that chemical transformation of WIOC to WSOC was not a dominant process for the production of WSOC during the study period. Equity, Diversity, and Inclusion Climate Assessment Activities: Development and Strategic Use in Diversity Action Plans
This project report describes climate assessment activities in support of the development of a college-level diversity, equity, and inclusion (EDI) action plan. Elements of the climate assessment activities are described along with their purpose and rationale for inclusion. Recommendations are made for libraries to design and deploy their own EDI assessment activities with the goal of developing robust action plans supportive of inclusive excellence
The Ankyrin Repeat Domain Controls Presynaptic Localization of Drosophila Ankyrin2 and Is Essential for Synaptic Stability
The structural integrity of synaptic connections critically depends on the interaction between synaptic cell adhesion molecules (CAMs) and the underlying actin and microtubule cytoskeleton. This interaction is mediated by giant Ankyrins, that act as specialized adaptors to establish and maintain axonal and synaptic compartments. In Drosophila, two giant isoforms of Ankyrin2 (Ank2) control synapse stability and organization at the larval neuromuscular junction (NMJ). Both Ank2-L and Ank2-XL are highly abundant in motoneuron axons and within the presynaptic terminal, where they control synaptic CAMs distribution and organization of microtubules. Here, we address the role of the conserved N-terminal ankyrin repeat domain (ARD) for subcellular localization and function of these giant Ankyrins in vivo. We used a P[acman] based rescue approach to generate deletions of ARD subdomains, that contain putative binding sites of interacting transmembrane proteins. We show that specific subdomains control synaptic but not axonal localization of Ank2-L. These domains contain binding sites to L1-family member CAMs, and we demonstrate that these regions are necessary for the organization of synaptic CAMs and for the control of synaptic stability. In contrast, presynaptic Ank2-XL localization only partially depends on the ARD but strictly requires the presynaptic presence of Ank2-L demonstrating a critical co-dependence of the two isoforms at the NMJ. Ank2-XL dependent control of microtubule organization correlates with presynaptic abundance of the protein and is thus only partially affected by ARD deletions. Together, our data provides novel insights into the synaptic targeting of giant Ankyrins with relevance for the control of synaptic plasticity and maintenance
