    Assessing Friction Characteristics of Liquid Lubricants

    The decline of fossil fuel reserves and the increasing awareness of greenhouse gas emissions have been the primary driving forces behind the need to conserve energy. To improve fuel efficiency friction modifiers are commonly blended into lubricants. Reduction of friction will clearly lead to less energy requirements. However, an accurate evaluation of lubricant performance is not possible using existing test equipment. The main reason is that current test rigs require operating conditions that induce wear so that the measurement of friction in these rigs is not a real evaluation of friction. The paper will detail the design and commissioning of a purpose built test rig to measure frictional characteristics of various oils as well as the results of the tests performed

    Torsion of parietal-peritoneal fat mimicking acute appendicitis: a case report

We present a rare case of torted fat on the parietal peritoneum over the anterior abdominal wall, mimicking clinical signs of acute appendicitis, which was diagnosed at laparoscopy. We are aware of only two other similar reported cases, both of which were diagnosed at the time of laparotomy. Case presentation A 41-year-old Caucasian woman presented with clinical signs of acute appendicitis. On diagnostic laparoscopy, a non-inflamed appendix was found. Further exploration revealed a necrotic torted appendage of fat overlying the parietal peritoneum of the right iliac fossa of the anterior abdominal wall. Conclusion Torted fatty appendages can be a diagnostic dilemma often mimicking more common causes of an acute abdomen. Laparoscopy is an excellent tool making the correct diagnosis in such cases

    Experimental and Computational Analysis of Polyglutamine-Mediated Cytotoxicity

    Expanded polyglutamine (polyQ) proteins are known to be the causative agents of a number of human neurodegenerative diseases but the molecular basis of their cytoxicity is still poorly understood. PolyQ tracts may impede the activity of the proteasome, and evidence from single cell imaging suggests that the sequestration of polyQ into inclusion bodies can reduce the proteasomal burden and promote cell survival, at least in the short term. The presence of misfolded protein also leads to activation of stress kinases such as p38MAPK, which can be cytotoxic. The relationships of these systems are not well understood. We have used fluorescent reporter systems imaged in living cells, and stochastic computer modeling to explore the relationships of polyQ, p38MAPK activation, generation of reactive oxygen species (ROS), proteasome inhibition, and inclusion body formation. In cells expressing a polyQ protein inclusion, body formation was preceded by proteasome inhibition but cytotoxicity was greatly reduced by administration of a p38MAPK inhibitor. Computer simulations suggested that without the generation of ROS, the proteasome inhibition and activation of p38MAPK would have significantly reduced toxicity. Our data suggest a vicious cycle of stress kinase activation and proteasome inhibition that is ultimately lethal to cells. There was close agreement between experimental data and the predictions of a stochastic computer model, supporting a central role for proteasome inhibition and p38MAPK activation in inclusion body formation and ROS-mediated cell death

    Data Mining for Gene Networks Relevant to Poor Prognosis in Lung Cancer Via Backward-Chaining Rule Induction

    We use Backward Chaining Rule Induction (BCRI), a novel data mining method for hypothesizing causative mechanisms, to mine lung cancer gene expression array data for mechanisms that could impact survival. Initially, a supervised learning system is used to generate a prediction model in the form of “IF <conditions> THEN <outcome>” style rules. Next, each antecedent (i.e. an IF condition) of a previously discovered rule becomes the outcome class for subsequent application of supervised rule induction. This step is repeated until a termination condition is satisfied. “Chains” of rules are created by working backward from an initial condition (e.g. survival status). Through this iterative process of “backward chaining,” BCRI searches for rules that describe plausible gene interactions for subsequent validation. Thus, BCRI is a semi-supervised approach that constrains the search through the vast space of plausible causal mechanisms by using a top-level outcome to kick-start the process. We demonstrate the general BCRI task sequence, how to implement it, the validation process, and how BCRI-rules discovered from lung cancer microarray data can be combined with prior knowledge to generate hypotheses about functional genomics

    Laboratory Evaluation of Dynamic Routing of Air Traffic in an En Route Arrival Metering Environment

    Arrival air traffic operations in the presence of convective weather are subject to uncertainty in aircraft routing and subsequently in flight trajectory predictability. Current management of arrival operations in weather-impacted airspace results in significant flight delay and suspension of arrival metering operations. The Dynamic Routing for Arrivals in Weather (DRAW) concept provides flight route amendment advisories to Traffic Management Coordinators to mitigate the impacts of convective weather on arrival operations. DRAW provides both weather conflict and schedule information for proposed route amendments, allowing air traffic managers to simultaneously evaluate weather avoidance routing and potential schedule and delay impacts. Subject matter experts consisting of retired Traffic Management Coordinators and retired Sector Controllers with arrival metering experience participated in a simulation study of Fort Worth Air Route Traffic Control Center arrival operations. Data were collected for Traffic Management Coordinator and Sector Controller participants over three weeks of simulation activities in October, 2017. Traffic Management Coordinators reported acceptable workload levels, a positive impact on their ability to manage arrival traffic while using DRAW, and initiated weather mitigation reroutes earlier while using DRAW. Sector Controllers also reported acceptable workload levels while using DRAW
