    A comparison of nicotine dose estimates in smokers between filter analysis, salivary cotinine, and urinary excretion of nicotine metabolites

    RATIONALE: Nicotine uptake during smoking was estimated by either analyzing the metabolites of nicotine in various body fluids or by analyzing filters from smoked cigarettes. However, no comparison of the filter analysis method with body fluid analysis methods has been published. OBJECTIVES: Correlate nicotine uptake estimates between filter analysis, salivary cotinine, and urinary excretion of selected nicotine metabolites to determine the suitability of these methods in estimating nicotine absorption in smokers of filtered cigarettes. MATERIALS AND METHODS: A 5-day clinical study was conducted with 74 smokers who smoked 1–19 mg Federal Trade Commission tar cigarettes, using their own brands ad libitum. Filters were analyzed to estimate the daily mouth exposure of nicotine. Twenty-four-hour urine samples were collected and analyzed for nicotine, cotinine, and 3′-hydroxycotinine plus their glucuronide conjugates. Saliva samples were collected daily for cotinine analysis. RESULTS: Each method correlated significantly (p < 0.01) with the other two. The best correlation was between the mouth exposure of nicotine, as estimated by filter analysis, and urinary nicotine plus metabolites. Multiple regression analysis implies that saliva cotinine and urinary output are dependent on nicotine mouth exposure for multiple days. Creatinine normalization of the urinary metabolites degrades the correlation with mouth exposure. CONCLUSIONS: The filter analysis method was shown to correlate with more traditional methods of estimating nicotine uptake. However, because filter analysis is less complicated and intrusive, subjects can collect samples easily and unsupervised. This should enable improvements in study compliance and future study designs

    Patient involvement in medical decision-making and pain among elders: physician or patient-driven?

    BACKGROUND: Pain is highly prevalent among older adults, but little is known about how patient involvement in medical decision-making may play a role in limiting its occurrence or severity. The purpose of this study was to evaluate whether physician-driven and patient-driven participation in decision-making were associated with the odds of frequent and severe pain. METHODS: A cross-sectional population-based survey of 3,135 persons age 65 and older was conducted in the 108-county region comprising West Texas. The survey included self-reports of frequent pain and, among those with frequent pain, the severity of pain. RESULTS: Findings from multivariate logistic regression analyses showed that higher patient-driven participation in decision-making was associated with lower odds (OR, 0.82; 95% CI, 0.75–0.89) of frequent pain, but was not significantly associated with severe pain. Physician-driven participation was not significantly associated with frequent or severe pain. CONCLUSIONS: The findings suggest that patients may need to initiate involvement in medical decision-making to reduce their chances of experiencing frequent pain. Changes to other modifiable health care characteristics, including access to a personal doctor and health insurance coverage, may be more conducive to limiting the risk of severe pain

    In this brief commentary, we provide some of our thoughts and opinions on the current and future use of zebrafish to model human eye disease, dissect pathological progression and advance in our understanding of the genetic bases of microphthalmia, andophthalmia and coloboma (MAC) in humans. We provide some background on eye formation in fish and conservation and divergence across vertebrates in this process, discuss different approaches for manipulating gene function and speculate on future research areas where we think research using fish may prove to be particularly effective

    Genetic analysis of soybean resistance to Fusarium solani f.sp. glycines

    In order to study the genetic control of soybean resistance to sudden death syndrome (SDS), a 5 x 5 diallel with the F2 generation, without the reciprocals, was carried out in a greenhouse. The following parents were used: Forrest, MG/BR-46 (Conquista), IAC-4, FT-Cristalina, and FT-Estrela. The first two cultivars are more resistant to SDS than IAC-4, which is considered to be moderately resistant to SDS, and the last two cultivars are highly susceptible. The fungus was inoculated with three colonized sorghum grains placed at the bottom of the holes with two soybean seeds. Single plants were evaluated between 14 and 37 days after emergency based on foliar severity symptoms (1-5) of SDS. The disease incidence and a disease index were also calculated for each plot (clay pots with five plants each). The analysis for severity and disease index was performed only with the data of the 37th day after emergence. Additive and dominant genetic effects were detected by Jinks-Hayman's analysis, but the dominant genetic effects were higher. The genetic parameters estimated indicated that the average degree of dominance showed the presence of overdominance; at least three loci or genic blocks that exhibited dominance were responsible for the genetic control of SDS resistance; the estimates of narrow-sense heritabilities were moderate (0.48 to 0.62), but in the broad-sense they were higher (0.90 to 0.95), thus reinforcing the presence of dominance effects; and the resistance to SDS was controlled mostly by dominant alleles. Five microsatellite markers (Satt163, Satt309, Satt354, Satt371 and Satt570), reported as linked to five QRLs of the SDS, were used to genotype the parents and showed the possibility of occurrence of multiallelism in those loci, but this evidence did not invalidate the fitting of the data to the Jinks-Hayman's model
