    Lrp Acts as Both a Positive and Negative Regulator for Type 1 Fimbriae Production in Salmonella enterica Serovar Typhimurium

    Leucine-responsive regulatory protein (Lrp) is known to be an indirect activator of type 1 fimbriae synthesis in Salmonella enterica serovar Typhimurium via direct regulation of FimZ, a direct positive regulator for type 1 fimbriae production. Using RT-PCR, we have shown previously that fimA transcription is dramatically impaired in both lrp-deletion (Δlrp) and constitutive-lrp expression (lrpC) mutant strains. In this work, we used chromosomal PfimA-lacZ fusions and yeast agglutination assays to confirm and extend our previous results. Direct binding of Lrp to PfimA was shown by an electrophoretic mobility shift assay (EMSA) and DNA footprinting assay. Site-directed mutagenesis revealed that the Lrp-binding motifs in PfimA play a role in both activation and repression of type 1 fimbriae production. Overproduction of Lrp also abrogates fimZ expression. EMSA data showed that Lrp and FimZ proteins independently bind to PfimA without competitive exclusion. In addition, both Lrp and FimZ binding to PfimA caused a hyper retardation (supershift) of the DNA-protein complex compared to the shift when each protein was present alone. Nutrition-dependent cellular Lrp levels closely correlated with the amount of type 1 fimbriae production. These observations suggest that Lrp plays important roles in type 1 fimbriation by acting as both a positive and negative regulator and its effect depends, at least in part, on the cellular concentration of Lrp in response to the nutritional environment

    The objective of this study was to determine the profile of the screening test for syphilis in pregnant women from a metropolitan hospital in Fortaleza-Ce. The research was carried out analyzing the quantitative epidemiological profile of gestational syphilis in patients who underwent the screening test for syphilis, through the reports of the clinical analysis laboratory of the registered hospital of MaracanaĂș, referring to the period from January to June, 2018. Gestational syphilis 72% of the total number of tests performed in the study period were found in 72 patients, of which 72% belong to the age range of 18-29 years. The data show that gestational syphilis still persists in the municipality, which indicates that the threat of congenital syphilis may still be an unresolved problem in the municipality, if the case is not treated properlyO trabalho teve como objetivo determinar o perfil do exame de triagem para sĂ­filis em gestantes de um hospital metropolitano de Fortaleza-Ce. A pesquisa foi realizada analisando o perfil epidemiolĂłgico quantitativo da sĂ­filis gestacional em pacientes que realizaram o exame de triagem de sĂ­filis, atravĂ©s dos relatĂłrios do laboratĂłrio de anĂĄlises clĂ­nicas do hospital de MaracanaĂș registrados, referentes ao perĂ­odo de janeiro a junho de 2018. A sĂ­filis gestacional foi evidenciada em 72 pacientes cerca de 4% do total de exames realizados no perĂ­odo do estudo, sendo que destes, 72% pertence a faixa etĂĄria de 18-29 anos. Os dados revelam que, a sĂ­filis gestacional ainda persiste no municĂ­pio, o que aponta que a ameaça da sĂ­filis congĂȘnita ainda pode ser um problema nĂŁo solucionado no municĂ­pio, se o caso nĂŁo for tratado devidamente.Trabajo tuvo como objetivo determinar perfil del examen de tamizaje para sĂ­filis en gestantes de un hospital metropolitano de Fortaleza-Ce.     Functional genomic analysis of cell division in C. elegans using RNAi of genes on chromosome III.

    Genome sequencing projects generate a wealth of information; however, the ultimate goal of such projects is to accelerate the identification of the biological function of genes. This creates a need for comprehensive studies to fill the gap between sequence and function. Here we report the results of a functional genomic screen to identify genes required for cell division in Caenorhabditis elegans. We inhibited the expression of approximately 96% of the approximately 2,300 predicted open reading frames on chromosome III using RNA-mediated interference (RNAi). By using an in vivo time-lapse differential interference contrast microscopy assay, we identified 133 genes (approximately 6%) necessary for distinct cellular processes in early embryos. Our results indicate that these genes represent most of the genes on chromosome III that are required for proper cell division in C. elegans embryos. The complete data set, including sample time-lapse recordings, has been deposited in an open access database. We found that approximately 47% of the genes associated with a differential interference contrast phenotype have clear orthologues in other eukaryotes, indicating that this screen provides putative gene functions for other species as well
