    Traction force microscopy on soft elastic substrates: a guide to recent computational advances

    The measurement of cellular traction forces on soft elastic substrates has become a standard tool for many labs working on mechanobiology. Here we review the basic principles and different variants of this approach. In general, the extraction of the substrate displacement field from image data and the reconstruction procedure for the forces are closely linked to each other and limited by the presence of experimental noise. We discuss different strategies to reconstruct cellular forces as they follow from the foundations of elasticity theory, including two- versus three-dimensional, inverse versus direct and linear versus non-linear approaches. We also discuss how biophysical models can improve force reconstruction and comment on practical issues like substrate preparation, image processing and the availability of software for traction force microscopy.Comment: Revtex, 29 pages, 3 PDF figures, 2 tables.     Formation of small-scale structure in SUSY CDM

    The lightest supersymmetric particle, most likely the lightest neutralino, is one of the most prominent particle candidates for cold dark matter (CDM). We show that the primordial spectrum of density fluctuations in neutralino CDM has a sharp cut-off, induced by two different damping mechanisms. During the kinetic decoupling of neutralinos, non-equilibrium processes constitute viscosity effects, which damp or even absorb density perturbations in CDM. After the last scattering of neutralinos, free streaming induces neutralino flows from overdense to underdense regions of space. Both damping mechanisms together define a minimal mass scale for perturbations in neutralino CDM, before the inhomogeneities enter the nonlinear epoch of structure formation. We find that the very first gravitationally bound neutralino clouds ought to have masses above 10^{-6} solar masses, which is six orders of magnitude above the mass of possible axion miniclusters.Comment: 7 pages, 3 figures, to appear in proceedings of "IDM 2002, 4th International Workshop on the Identification of Dark Matter

    Suitability of a CMV/EGFP cassette to monitor stable expression from human artificial chromosomes but not transient transfer in the cells forming viable clones

    Human artificial chromosomes (HACs) were generated by transfer of telomerized PAC constructs containing alpha satellite DNA of various human chromosomes. To monitor which cells took up constructs and subsequently formed stable clones under blasticidin S (BS) selection, a CMV/EGFP expression cassette was inserted into a HAC construct based on chromosome 5 alpha satellite DNA (142 kb). Lipofection into HT1080 cells resulted in a small proportion of cells exhibiting bright green fluorescence on day 1. Areas containing such early green cells were marked, and plates monitored over 2 weeks. In only one out of 41 marked areas, a viable clone developed. In the remaining 40 areas, the green cells ceased division at 1-8 cells. In contrast, outside the marked areas, 16 stable clones formed which did not exhibit green fluorescence during the first cell divisions, but all cells of each became green around day 4 -6. Fluorescence in situ hybridization (FISH) analysis of isolated clonal lines demonstrated low copy HAC formation without integration. We conclude that transient expression of an EGFP marker on HAC DNA is not a suitable means for the identification of the proportion of transfected cells which are capable of forming viable clones. One explanation could be that the high copy number required to consistently detect transient EGFP expression (Schindelhauer and Laner, 2002) impairs viability and clone formation.     We show that on three-dimensional Riemannian manifolds without boundaries and with trivial first real de Rham cohomology group (and in no other dimensions) scalar field theory and Maxwell theory are equivalent: the ratio of the partition functions is given by the Ray-Singer torsion of the manifold.     Scale dependence of cosmological backreaction

    Due to the non-commutation of spatial averaging and temporal evolution, inhomogeneities and anisotropies (cosmic structures) influence the evolution of the averaged Universe via the cosmological backreaction mechanism. We study the backreaction effect as a function of averaging scale in a perturbative approach up to higher orders. We calculate the hierarchy of the critical scales, at which 10% effects show up from averaging at different orders. The dominant contribution comes from the averaged spatial curvature, observable up to scales of 200 Mpc. The cosmic variance of the local Hubble rate is 10% (5%) for spherical regions of radius 40 (60) Mpc. We compare our result to the one from Newtonian cosmology and Hubble Space Telescope Key Project data.Comment: 6 pages, 2 figures; v3: substantial modifications, new figure
